ABSTRACTTingginya konsumsi fruktosa dan kolesterol di masyarakat meningkatkan kejadianobesitas, dislipidemia, resistensi insulin, serta sindroma metabolik yang merupakanfaktor risiko utama terjadinya perlemakan pankreas atau non-alcoholic fattypancreas disease NAFPD . NAFPD merupakan kondisi yang masih minimdiketahui patogenesis serta tatalaksanya, namun kondisi NAFPD memiliki sifatreversibel apabila dilakukan modifikasi terhadap faktor risiko utamanya. Penelitian ini meneliti efek penggunaan kombinasi simvastatin dan ekstrak etanol akar tanaman Acalypha indica Linn. yang diketahui memiliki efek antihiperlipidemia dan antidiabetik terhadap perbaikan perlemakan pankreas tikus Sprague-Dawley pasca diet tinggi fruktosa dan kolesterol. Tikus diinduksi diet tinggi fruktosa dan kolesterol selama 4 minggu dan selanjutnya tikus dibagi menjadi 4 kelompok lalu diberikan perlakuan tatalaksana yang berbeda selama 4 minggu, yaitu kontrol negatif K , kontrol positif menggunakan simvastatin S , ekstrak A. indica AI , dan kombinasi S-AI. Hasil analisis uji One Way Anova dengan uji Post Hoc Bonferroni menunjukkan adanya perbedaan perlemakan pankreas yang bermakna antara kelompok tikus K dengan kelompok S p = 0,024, 95 CI: 0,038-0,696 serta antara kelompok tikus K dengan kelompok terapi kombinasi S-AI p = 0,001, 95 CI: 0,241 ndash; 0873 . Kombinasi simvastatin-ekstrak A. indica memberikan efek sinergis dalam perbaikan perlemakan pankreas, namun diperlukan penelitian lebih lanjut mengenai kemungkinan efek samping interaksi tersebut.
ABSTRACTHigh fructose and cholesterol diet nowadays has increased the incidence of obesity, dyslipidemia, insulin resistance, and metabolic syndrome which are the main risk factors of non fatty pancreas disease NAFPD . The pathogenesis and treatment of NAFPD is currently not well understood yet, but studies show that NAFPD is a reversible state which can be treated by modifying its underlying risk factors. This research aims to understand the effect of simvastatin ndash Acalypha indica Linn. extract combination which is known to have antihyperlidemic and antidiabetic properties on fatty pancreas improvement in Sprague Dawley mice induced with high fructose and cholesterol diet. Sprague Dawley mice were induced with high cholesterol and fructose diet for 4 weeks, followed by 4 weeks of different 4 treatments for different mice groups, which are negative control, positive control using simvastatin, A. indica extract, and simvastatin A. indica extraxt combination. One Way Anova and Post Hoc Bonferroni test results show significant improvement of fatty pancreas in mice without therapy and mice treated with simvastatin therapy p 0,024, 95 CI 0,038 0,696 , also in mice without therapy and mice treated with simvastatin A. indica extract therapi p 0,000, 95 CI 0,241 ndash 0873 . Simvastatin A. indica extract combination shows synergic effect on fatty pancreas improvement, but further research on adverse effects of the two substrates interaction is needed to confirm safety use of the therapy.
