Infeksi Dengue merupakan penyakit endemik di daerah tropis dan subtropis yang disebabkan oleh virus dengue DENV . Hingga saat ini belum ada antivirus spesifik untuk infeksi DENV. Diketahui bahwa derajat viral load berkaitan dengan keparahan penyakit. Curcuma longa L. kunyit dengan senyawa aktif utama kurkumin diketahui memiliki aktivitas antivirus terhadap DENV secara in vitro. Penelitian ini merupakan studi awal untuk mengetahui efek antivirus ekstrak C. longa terhadap DENV-2 dan toksisitas akut di organ hati dan ginjal pada mencit ddY. Tahap pertama dilakukan uji toksisitas akut oral ekstrak C. longa pada mencit ddY untuk mengetahui LD50. Dosis aman yang diperoleh digunakan untuk uji toksisitas organ hati dan ginjal mencit dengan pengamatan histopatologi serta biokimia SGPT, SGOT, ureum, kreatinin . Kedua dilakukan uji potensi ekstrak C. longa terhadap sel Huh7it-1 terinfeksi DENV-2 pada mencit ddY. Ekstrak C. longa diberikan peroral dosis 0.147 mg untuk tiap mencit dua jam setelah inokulasi sel terinfeksi secara intraperitonial. Serum dikoleksi dari intraorbital pada jam ke-6 dan jam ke-24 setelah inokulasi. Titer virus dinilai dengan metode focus assay. Berdasarkan hasil uji toksisitas akut oral ekstrak C. longa hingga dosis 7500 mg/kgBB tidak ada kematian. Pemeriksaan histopatologi menunjukkan tidak ada kelainan spesifik pada organ hati dan ginjal. Tidak ada peningkatan nyata kadar SGPT, SGOT, ureum, dan kreatinin. Ekstrak C. longa menurunkan titer virus dibandingkan kontrol. Hasil penelitian ini membuktikan ekstrak C. longa tidak toksik terhadap ginjal dan hati mencit serta memiliki efek antivirus terhadap DENV-2.

Dengue infection, caused by Dengue Virus DENV , is one of endemic diseases in tropical and subtropical region. Until now, there is no specific antiviral for dengue infection. It is known that the degree of viral load is related to disease severity. Curcuma longa L. tumeric with curcumin as major active compound has been identified for its antiviral effect for dengue in vitro. This study was a preliminary study to determine antiviral effect of C. longa extract on DENV 2 and its acute toxicity in ddY mice liver and kidney. The acute oral toxicity of C. longa extract was observated to determine LD50. The safe doses obtained were used for toxicity tests of liver and kidney with histopathological and biochemical observations SGPT, SGOT, urea, creatinine . The antiviral effect of C. longa exstract was tested using ddY mice inoculated intraperitoneally with Huh7it 1 cells infected by DENV 2. The C. longa extract was given orally dose 147 mg for each mice two hours after infection. Serum was collected from intraorbital at 6 hours and 24 hours after infection.Viral load was assessed by focus assay method. Based on acute oral toxicity test results C. longa extract up to dose 7500 mg kgbw there was no demise. Histopathological examination showed no specific abnormalities in liver and kidney organ. There was no significant increase in levels of SGPT, SGOT, urea, and creatinine. Extract C. longa lowered the viral titer compared to controls. The results of this study prove that C. longa extract was not toxic mice liver and kidney as well as had antiviral effect against DENV 2.
