ABSTRAKPenelitian studi pembentukan DNA adduct 8-Hidroksi-2'-Deoksiguanosin (8-OHdG) sebagai biomarker kerusakan DNA akibat oksidatif stress. Penelitian ini dilakukan dengan mereaksikan basa DNA 2'-deoksiguanosin 5'-monofosfat dengan TBHQ dan BHT. Profil pembentukan 8-OHdG dilakukan pada suhu 37°C dan 60°C, pH 7,4 dan pH 8,4, dengan waktu inkubasi 5 jam serta dengan penambahan FeSO4.Hasil adduct dianalisis menggunakan HPLC reversed phase dengan detektor UV pada panjang gelombang 254 nm. Hasil analisis diperoleh bahwa 8-OHdG terbentuk akibat reaksi dari 2'-Deoksiguanosin 5'Monofosfat dengan Hidroksi radikal dari TBHQ, BHT dan Fe (II) . Adduct yang terbentuk terdapat pada variasi suhu dan pH yang lebih tinggi dan cenderung lebih stabil dalam setiap variasi kondisi. Sedangkan pada penambahan Hidrogen Peroksida hanya terjadi pembentukan di zat uji TBHQ. Hasil penelitian menunjukan bahwa kondisi suhu dan pH yang lebih tinggi mempengaruhi pembentukan DNA Adduct.

ABSTRACT This research of study DNA adduct formation of 8 - hydroxy -2' - Deoksiguanosin ( 8 - OHdG ) as a biomarker of DNA damage due to oxidative stress . This research was carried out by reacting the DNA bases deoksiguanosin 2' -5'-monophosphate with TBHQ and BHT. Profile formation of 8-OHdG carried out at a temperature of 37° C and 60° C, pH 7.4 and pH 8.4 , with 5 hours of incubation time and with the addition FeSO4. The results of adducts were analyzed using reversed phase HPLC with UV detector at a wavelength of 254 nm. Results of the analysis showed that 8-OHdG is formed from the reaction of 2'-hydroxy Deoksiguanosin 5'Monofosfat with radicals of TBHQ, BHT and Fe ( II). Adducts formed are on the variation of temperature and pH are higher and tend to be more stable in every variation of conditions. While the addition of hydrogen peroxide formation only occurs in the test substance TBHQ . The results showed that the conditions of temperature and higher pH affects the formation of DNA adducts.
