ABSTRAKObat golongan α-glukosidase inhibitor merupakan salah satu obat yang digunakan untuk pengobatan diabetes melitus. Penelitian sebelumnya menunjukkan bahwa 80% ekstrak etanol kulit batang Cephalomappa malloticarpa J.J.Sm. memiliki penghambatan aktivitas α-glukosidase yang lebih baik dibandingkan dengan acarbose standar. Pada penelitian ini dilakukan fraksinasi dengan metode cair-cair pada ekstrak etanol 80% kulit batang Cephalomappa malloticarpa J.J.Sm. untuk melihat apakah hasil fraksinasi memiliki daya hambat aktivitas -glukosidase yang lebih baik dibandingkan ekstrak etanol 80%. Selain itu juga dilakukan penentuan kadar fenol total dan fitokimia ekstrak dan fraksi kulit batang Cephalomappa malloticarpa J.J.Sm. Uji daya hambat α-glukosidase dilakukan secara in vitro pada ekstrak dan fraksi kental yang diukur absorbansinya menggunakan microplate reader. Hasil pengujian menunjukkan bahwa ekstrak etanol 80% memiliki daya hambat aktivitas α-glukosidase yang lebih baik dengan IC50 sebesar 38,42 ± 1,20 μg / mL dibandingkan fraksi n-heksan, fraksi etil asetat dan fraksi metanol dengan IC50 sebesar 374,69 ± 6,75 μg / mL; 157,52 ± 4,01 μg / mL; dan 331,21 ± 4,46 μg / mL, masing-masing. Fraksi etil asetat memiliki kandungan total fenol tertinggi yaitu 450,34 mg GAE / g ekstrak diikuti oleh ekstrak etanol 80%, fraksi metanol dan fraksi n-heksan 196,74; 102.03; dan ekstrak GAE / g 22,95 mg. Skrining fitokimia menunjukkan bahwa ekstrak etanol 80% mengandung flavonoid, terpenoid, saponin, glikosida; fraksi n-heksan dan fraksi metanol mengandung terpenoid, saponin glikosida; Fraksi etil asetat mengandung flavonoid, terpenoid dan glikosida. Penelitian ini menunjukkan bahwa fraksinasi ekstrak etanol 80% dapat menurunkan aktivitas kulit batang Cephalomappa malloticarpa J.J.Sm. dalam menghambat α-glukosidase.
ABSTRACTThe α-glucosidase inhibitor class of drugs is one of the drugs used for the treatment of diabetes mellitus. Previous studies have shown that 80% ethanol extract of the stem bark of Cephalomappa malloticarpa J.J.Sm. has better α-glucosidase activity inhibition compared to standard acarbose. In this study, fractionation using the liquid-liquid method was carried out on 80% ethanol extract of the stem bark of Cephalomappa malloticarpa J.J.Sm. to see whether the fractionation has better inhibition of -glucosidase activity than 80% ethanol extract. In addition, the determination of total phenol content and phytochemical extracts and fraction of Cephalomappa malloticarpa J.J.Sm bark was also carried out. The α-glucosidase inhibition test was carried out in vitro on viscous extracts and fractions which were measured for absorbance using a microplate reader. The test results showed that 80% ethanol extract had better α-glucosidase activity inhibition with an IC50 of 38.42 ± 1.20 μg / mL compared to the n-hexane fraction, ethyl acetate fraction and methanol fraction with IC50 of 374.69 ± 6.75 μg / mL; 157.52 ± 4.01 μg / mL; and 331.21 ± 4.46 μg / mL, respectively. Ethyl acetate fraction had the highest total phenol content, namely 450.34 mg GAE / g extract followed by 80% ethanol extract, methanol fraction and n-hexane fraction 196.74; 102.03; and extract GAE/g 22.95 mg. Phytochemical screening showed that 80% ethanol extract contained flavonoids, terpenoids, saponins, glycosides; n-hexane fraction and methanol fraction containing terpenoids, saponin glycosides; The ethyl acetate fraction contains flavonoids, terpenoids and glycosides. This study shows that fractionation of 80% ethanol extract can reduce the activity of the stem bark of Cephalomappa malloticarpa J.J.Sm. in inhibiting α-glucosidase.
