ABSTRAKResep jamu kuno Au Fere II telah banyak digunakan oleh masyarakat Maluku sebagai pengobatan tradisional dalam terapi hipertensi. Namun potensi dari ekstrak Au Fere II belum dibuktikan secara lebih mendetail akan khasiatnya dalam terapi hipertensi. Penelitian ini bertujuan untuk mengetahui potensi yang dapat dihasilkan dari resep jamu kuno Au Fere II terhadap tekanan darah, berat badan, dan morfologi hewan uji. Tujuan lain adalah untuk mengevaluasi efek pemberian ekstrak resep jamu kuno Au Fere II pada hewan uji terhadap aktivitas level renin plasma. Pada penelitian ini digunakan enam jenis kelompok tikus (n=4) berbeda yang terdiri dari satu kelompok Normal dan lima kelompok tikus model 2K1C. Pada tikus model 2K1C, hipertensi diinduksi dengan pemasangan mikroklip stainless steel pada arteri ginjal kiri selama lima minggu. Kelompok tikus model terdiri dari kontrol negatif (2K1C, tanpa perlakuan), kontrol positif (kaptopril 4,5 mg/200 g BB), dan 3 kelompok yang diberikan ekstrak Au Fere II dengan dosis berbeda (0,495; 0,99; dan 1,98 ml/200 g BB). Pemberian ekstrak dan kaptopril dilakukan secara peroral selama satu minggu. Pemberian ekstrak Au Fere II memberikan penurunan terhadap tekanan darah, tidak mempengaruhi pertambahan berat badan, dan menyebabkan atrofi pada ginjal yang di klip. Efek pemberian ekstrak resep jamu kuno Au Fere II pada hewan uji secara signifikan menurunkan konsentrasi level renin tikus percobaan.

ABSTRACT

The ancient Au Fere II herbal recipe has been widely used by the Maluku people as a traditional treatment in hypertension therapy. However, the potential of Au Fere II extract has not been proven in for its efficacy in hypertension therapy. The study aims to determine the potential that can be constructed from the ancient Au Fere II herbal recipe on blood pressure, body weight, and renal morphology of test animals. And at the same time, it also evaluates the effect of extracts of ancient herbal medicine Au Fere II in test animals to determine plasma renin level activity. In this study six different groups of rats (n = 4) were used consisting of one Normal group and five groups of 2K1C rats. In 2K1C rats, hypertension was induced by placing stainless steel microclips in the left kidney artery for five weeks. The rats group model consisted of negative controls (2K1C, without treatment), positive controls (captopril 4.5 mg / 200 g BW), and 3 groups who were given Au Fere II extract with different doses (0.495; 0.99; and 1.98 ml / 200 g BW). Administration of extract and captopril were given orally for one week. The provision of Au Fere II extract gave a decrease in blood pressure, did not affect weight gain, and caused atrophy in the clipped kidney. The effect of prescribing extracts of ancient herbal medicine Au Fere II to test animals were significantly reduced the concentration of renin levels in experimental rats.
