Ectonucleotide pyrophosphatase/phosphodiesterase1 (ENPP1) merupakan protein yang memiliki peran sebagai modulator mineralisasi pyrophosphate (PPi) yang merupakan struktur anorganik yang dapat menghambat proses minerlisasi tulang karena pembetukannya berlawanan dengan pembentukan hiroksiapatit. Ketidakseimbangan dalam mineralisasi tulang akan menghambat proses remodeling tulang dan berakibat pada penurunan Bone Mineral Density (BMD). Osteoporosis merupakan penyakit kerangka sistemik yang ditandai dengan kepadatan mineral tulang yang kurang dari 2,5 dan diukur oleh dual-emission x-ray absorptiometry (DXA). Selain berpengaruh pada osteoporosis, ENPP1 juga dicurigai memiliki pengaruh terhadap regulasi zat-zat yang memodulasi terjadinya Penyakit Neurodegeneratif. Tujuan dari penelitian ini adalah untuk menganalisis distribusi polimorfisme gen ENPP1 K121Q rs1044498 pada penyakit Osteoporosis dan keterkaitannya dengan penyakit Neurodegenertif. ENPP1 K121q rs1044498, ddH2O, dan MyTaq dicampur pada DNA template (sampel osteoporosis dan non-osteoporosis), kemudian dianalisis menggunakan teknik PCRRFLP menggunaan AVAII sebagai enzim restriksi dan dielektroforesis untuk melihat hasilnya. Selanjutnya dianalisis menggunakan uji Pearson Chi - Square dan Continuity Correction. Hasil penelitian menunjukkan terdapat perbedaan yang signifikan dalam distribusi frekuensi polimorfisme genotipe ENPP1 K121Q antara osteoporosis dan nonosteoporosis. Kesimpulannya, polimorfisme gen K121Q berkaitan dengan penurunan BMD dan merupakan faktor predisposisi osteoporosis.

Imbalances in bone mineralization will inhibit the bone remodeling process and resulting decrease in Bone Mineral Density (BMD). Osteoporosis is a systemic skeletal disease with a bone mineral density less than 2.5 and measured by dual-emission x-ray absorptiometry (DXA). Besides the effect on osteoporosis, ENPP1 is also suspected of having an influence on the regulation of substances that modulate the incidence of neurodegenerative diseases. The purpose of this study is to analyze the polymorphism distribution of the ENPP1 K121Q (rs1044498) gene in osteoporosis and its association with neurodegenerative diseases. ENPP1 K121Q (rs1044498), ddH2O, and MyTaq are mixed on the DNA template (osteoporosis and non-osteoporosis samples), then analyzed using the PCR-RFLP technique using AVAII as a restriction enzyme and electrophoresis to see the results. Then analyzed using the Pearson Chi - Square test and Continuity Correction. The results showed that there was a significant difference in the frequency distribution of the ENPP1 K121Q genotype polymorphism between osteoporosis and non-osteoporosis. In conclusion, ENPP! K121Q gene polymorphism is associated with decreased BMD and is a predisposing factor for osteoporosis.
