"Pengetahuan yang tepat diperlukan dalam penanganan sediaan sitostatika untuk mengurangi paparan akibat kecelakaan kerja dan meningkatkan keamanan pasien. Tujuan dari penelitian ini adalah untuk mengetahui gambaran tingkat pengetahuan mahasiswa keperawatan terhadap pemberian obat sitostatika (kemoterapi intravena) yang aman. Metode yang digunakan adalah penelitian deskriptif dengan teknik total sampling. Kuesioner yang dipergunakan merupakan adaptasi dari Exposure of Chemotherapy Drug Questionnaire. Hasil menunjukkan sebanyak 81,6% (71 dari 87) responden berada pada tingkat pengetahuan yang rendah dengan skor kurang dari tiga puluh. Hanya terdapat 16% responden yang berada pada tingkat pengetahuan tinggi, dengan skor antara tiga puluh sampai empat puluh. Responden tanpa kepemilikan informasi memiliki kecenderungan berada pada tingkat pengetahuan yang rendah. Hasil yang diperoleh menunjukkan bahwa mahasiswa keperawatan memiliki pengetahuan yang tidak memadai terhadap penatalaksanaan kemoterapi yang aman. Penyesuaian kurikulum diperlukan untuk memberikan pembekalan yang sesuai pada mahasiswa keperawatan sehingga dapat meningkatkan pelayanan keperawatan profesional yang berkualitas.

Sufficient knowledge needed when handling cytostatic drug to minimize exposure due to work accident and to increase patient safety. The aim of this study is to evaluate nursing students’ knowledge in safe handling of chemotherapy drug. Descriptive method used with total sampling technique. The questionnaire was adapted from Exposure of Chemotherapy Drug Questionnaire. The result shows 81,6% (71 of 87) respondents had low level of knowledge with a score less than thirty. Only 16% respondents had high level of knowledge, with a score between thirty to forty. Respondents with no possession of knowledge had tendency to have low rate of knowledge. The result shows majority of nursing students have insufficient knowledge in handling with chemotherapy. This report suggests conforming curriculum in nursing education is needed to give sufficient knowledge for nursing student in purpose to increase professional nursing care."

"ABSTRAKLatar belakang: Kemoterapi pilihan untuk Diffuse Large B Cell Lymphoma (DLBCL) adalah regimen yang mengandung doksorubisin. Doksorubisin merupakan obat kemoterapi golongan antrasiklin yang bekerja sebagai anti Topoisomerase II (Top2). Penelitian sebelumnya terhadap galur sel tumor menunjukkan bahwa ekspresi Topoisomerase IIα (Top2A) yang tinggi berhubungan dengan sensitifitas terhadap antrasiklin yang tinggi pula. Tujuan penelitian ini adalah untuk mengetahui ekspresi protein Top2A pada DLBCL dan hubungannya dengan respon terapi.Bahan dan cara kerja: Dilakukan studi analitik potong lintang terhadap 38 kasus DLBCL dengan pulasan CD20 positif dan telah mendapatkan kemoterapi minimal 4 siklus. Dilakukan pulasan imunohistokimia terhadap protein Top2A dan dinilai menggunakan H-score.Hasil: Secara keseluruhan ekspresi Top2A ditemukan pada 37 dari 38 kasus (97,4%) dengan nilai H-score sangat bervariasi yaitu antara 101,5 sampai dengan 215,0 dan median 124,1. H-score Top2A digolongkan tinggi jika H-score lebih dari 124,1. Analisis statistik menunjukkan bahwa ekspresi Top2A pada DLBCL tidak berhubungan bermakna dengan respon terapi (p=0,670).Kesimpulan: Tidak ditemukan hubungan bermakna antara ekspresi Top2A dengan respon terapi. Ekspresi Top2A tidak dapat dijadikan faktor prediktor respon terapi pada DLBCL.

ABSTRACTBackground: Standard of chemotherapy for Diffuse Large B Cell Lymphoma (DLBCL) is a regimen containing doxorubicin. Doxorubicin is a component of anthracycline based chemotherapy that work as anti Topoisomerase II (Top2). Previous study on tumor cell lines showed that high expression of Topoisomerase IIα (Top2A) was related to higher sensitivity to anthracycline. The aim of this study is to know the expression of Top2A and its relation to treatment response. Material and methods: This is an analytic cross-sectional study on 38 CD20 positive DLBCL cases that have been treated with at least 4 cycles of chemotherapy. The immunohistochemical staining for Top2A protein was performed assesed using H-score.Result: Expression of Top2A protein were found in 37 of 38 (97,4%) cases (H-score range: 101.5-215.0 and median 124.1). Top2A was defined as high if H-score was higher than 124.1. Statistical analysis showed that Top2A expression in DLBCL was not significantly related to treatment response (p=0.670).Conclusion : There was no significant relation between Top2A expression to treatment response. Top2A expression in DLBCL cannot be used as a predictor of treatment response."

"6-Merkaptopurin 6-MP adalah obat kemoterapi golongan antimetabolit purin yang digunakan dalam terapi leukemia limfositik akut. 6-Merkaptopurin di dalam tubuh di metabolisme menjadi metabolit aktifnya. 6-Tioguanin nukeotida. Penelitian ini bertujuan untuk memperoleh metode optimum dan tervalidasi dalam menganalisis 6-Merkaptopurin dan 6-Tioguanin 6-TG secara simultan dalam sampel Dried Blood Spot menggunakan kromatografi cair kinerja ultra tinggi tandem spektrometri massa. Larutan kontrol kualitas dan kurva kalibrasi dibuat dengan menotolkan masing-masing sebanyak 40 L pada kertas CAMAG DBS dan dikeringkan selama 3 jam. Kertas DBS dipotong dengan diameter 8 mm dan diekstraksi menggunakan larutan metanol yang mengandung 5-fluorourasil 5-FU sebagai baku dalam. Analisa dilakukan dengan kolom Waters AcquityTM UPLC BEH Amide 1,7 m 2,1 x 100 mm dengan fase gerak berupa asam format 0,2 dalam air ndash; asam format 0,1 dalam asetonitril ndash; metanol dengan gradient elusi dan laju alir 0,2 mL/menit. Deteksi massa dilakukan dengan Waters Xevo TQD dengan Electrospray Ionization ESI positif untuk 6-MP dan 6-TG dan ESI negative untuk 5-FU pada mode Multiple Reaction Monitoring. Deteksi 6MP, 6-TG, 5-FU berturut-turut adalah 153,09 > 119,09; 168,09 > 107,06; 129,15 > 42,05. Metode ini linear dalam rentang 25 ndash; 1000 ng/mL untuk 6-MP dan 6-TG, nilai r berturut turut adalah ge; 0,996 dan ge; 0,995. Nilai diff dan koefisien variasi KV untuk akurasi dan presisi intra hari dan antar hari tidak lebih dari 15 dan tidak lebih dari 20 pada konsentrasi LLOQ. Metode ini memenuhi persyaratan validasi sesuai Guideline on bioanalytical Method Validation oleh European Medicines Agency tahun 2011.

6 Mercaptopurine 6 MP is a chemotherapeutic drug that belongs to a class of purine antagonist antimetabolite used for acute lymphocytic leukemia. 6 Mercaptopurine has to go through the metabolic pathway to become it rsquo s active metabolites 6 Thioguanin nucleotide. This study aimed to obtain an optimum and validated method for analyzing 6 Mercaptopurine and 6 Thioguanine 6 TG simultaneously in Dried Blood Spot samples using ultra high performance liquid chromatography tandem mass spectrometry. The quality control and calibration curves solutions were made by respectively spot 40 L at DBS CAMAG paper and dried for 3 hours. DBS papers were cut with a diameter of 8 mm and extracted with methanol containing internal standard 5 fluorouracil 5 FU. Separation was performed with Waters AcquityTM UPLC BEH Amide column 1.7 m 2.1 x 100 mm with a mobile phase consists of 0.2 formic acid in water ndash 0.1 formic acid in acetonitrile ndash Methanol with gradient elution and flow rate 0.2 mL minute. Mass detection was done using Waters Xevo TQD with positive electrospray ionization ESI for 6 MP and 6 TG and negative ESI for 5 FU in Multiple Reaction Monitoring mode. Detection of 6 MP, 6 MMP, 5 FU respectively was 153,09 119,09 168,09 107,06 129,15 42,05. This method is linear with the range 25 1000 ng mL for 6 MP and 6 TG with consecutive r value is ge 0,996 and ge 0,995. Diff value and coefficient of variation CV for accuracy and precision of intra day and inter day are not more than 15 and not more than 20 at a concentration LLOQ. This method fulfilled the requirements of validation which refers to the European Medicines Agency guideline."
