"Latar belakang: Endoglin adalah faktor angiogenesis spesifik yang diduga terkait dengan patogenesis endometriosis. Tujuan penelitian ini adalah mengevaluasi hubungan kadar endoglin pada cairan peritoneum pasien dengan endometriosis.
Metode: Penelitian ini merupakan penelitian observasional potong lintang yang dilaksanakan pada bulan Maret 2011 sampai Juli 2012 di RS Dr. Wahidin Sudirohusodo dan rumah sakit swasta di Makassar. Semua pasien yang akan menjalani laparoskopi untuk masalah infertilitas dan masalah lainnya serta memenuhi kriteria inklusi diminta untuk mengisi
kuesioner dan diambil cairan peritoneum sebanyak 5 cc untuk mengukur kadar endoglin dengan metode ELISA. Stadium endometriosis ditegakkan dengan memakai kriteria ASRM dan dipisahkan menjadi endometriosis ringan dan berat. Data dianalisis dengan Excel dan analisis korelasi Spearman.
Hasil: Pada kelompok endometriosis didapatkan konsentrasi endoglin antara 14,43-15,65 ng/mL (median 15,04 ng/mL) sedangkan pada kelompok kontrol antara 7,42-10,26 ng/mL (median 8,84 ng/mL). Kadar endoglin peritoneum sama atau lebih tinggi dari 11 ng/mL mungkin dipakai sebagai titik potong prediktor kasus endometriosis.
Kesimpulan: Kadar endoglin peritoneum meningkat secara proporsional sesuai dengan meningkatnya stadium endometriosis, sehingga kadar endoglin dapat dipakai sebagai prediktor endometriosis.

Abstract
Background: Endoglin is a specific angiogenic factor suspected to involve in the pathogenesis of endometriosis. The aim of this study was to evaluate the significance of endoglin concentration in the peritoneal fluid of patients with endometriosis.
Methods: This pilot study was performed between March 2011 and July 2012 at Dr. Wahidin Sudirohusodo Hospital and another private hospital in Makassar, Indonesia. This was an observational, cross-sectional study. All patients undergoing laparoscopy for infertility and other cases with suitable inclusion criteria were asked to answer a questionnaire and had a 5 cc peritoneal fluid sample taken for measurement of peritoneal endoglin concentration using ELISA. Endometriosis stage was classified using ASRM criteria and divided into two groups, mild and severe. All data were analyed using Excel and Spearman correlation analysis.
Results: In the endometriosis group peritoneal endoglin concentration ranged between 14.43- 15.65 ng/mL (median 15.04 ng/mL) which is higher than control group 7.42-10.26 ng/mL (median 8.84 ng/mL). A peritoneal endoglin concentration equal to or higher than 11 ng/mL could be used to predict endometriosis.
Conclusion: Endoglin concentrations increased proportionally with the severity of endometriosis, and many be used as predictive factor of endometriosis cases."

"Peritonitis tuberkulosis adalah peradangan peritoneum parietal atau visceral yang disebabkan oleh mycobacterium tuberculosis. Efusi pleura terjadi karena komplikasi dari penyakit yang menyertai maupun proses infeksi yang mengenai rongga pleura. Masalah keperawatan yang dapat ditegakkan yaitu gangguan pertukaran gas dan intoleransi aktivitas. Karya Ilmiah Akhir Ners ini bertujuan untuk menganalisis asuhan keperawatan dengan pemberian intervensi pursed lip breathing dan graded exercise therapy. Metode yang digunakan berupa laporan kasus yang dikelola selama 4 hari berdasarkan tinjauan literatur. Hasil analisis kombinasi intervensi mengenai efektifitas pemberian intervensi pursed lip breathing dan graded exercise therapy terbukti efektif dalam mengatasi masalah gangguan pertukaran gas dan intoleransi aktivitas. Dibuktikan dengan perubahan tanda-tanda vital menjadi lebih stabil, frekuensi pernapasan dan frekuensi nadi membaik, serta keluhan sesak pasien berkurang. Toleransi aktivitas pasien menjadi lebih meningkat dengan kemampuan self care yang meningkat. Pemberian intervensi pursed lip breathing dan graded exercise therapy direkomendasikan dalam mengatasi gangguan pertukaran gas dan intoleransi aktivitas.

Peritoneal tuberculosis is an inflammation of the parietal or visceral peritoneum caused by mycobacterium tuberculosis. Pleural effusion occurs due to complications from the accompanying disease as well as infectious processes that affect the pleural cavity. Enforceable nursing problems are gas exchange disorders and activity intolerance. This study aims to analyze nursing care by providing pursed lip breathing and graded exercise therapy   interventions. The method that used is a case report that is managed for four days based on a literature review. The results of the combination analysis of interventions regarding the effectiveness of giving pursed lip breathing and graded exercise therapy interventions have been shown to be effective in overcoming the problems of gas exchange disorders and activity intolerance. Evidenced by the change of vital signs to become more stable, the frequency of breathing and the frequency of the pulse improve, as well as dyspnea of the patient are reduced. The tolerance of patient activity improved with the increaseasing self-care ability. Intervention of pursed lip breathing and graded exercise therapy is recommended in overcoming gas exchange disorders and activity intolerance."

"ABSTRAK
Karakteristik peritoneum dan konsentrasi glukosa mempengaruhi perbedaan tekanan osmotik sepanjang dwell time pada CAPD. Penelitian ini bertujuan menganalisis peran karakteristik peritoneum dan konsentrasi glukosa terhadap pengeluaran cairan. Desain penelitian cross-sectional dengan pendekatan retrospektif. Berdasarkan consecutive sampling didapat 53 responden. Data berasal dari tahun 2005-2010. Hasil uji t menunjukkan nilai p<0,05 pada rerata pengeluaran cairan berdasarkan konsentrai glukosa pada siang dan malam. Hasil uji ANOVA menunjukkan nilai p<0,05 pada rerata pengeluaran cairan berdasarkan karakteristik peritoneum pada pagi dan sore. Peneliti menyimpulkan karakteristik peritoneum dan konsentrasi glukosa berperan terhadap pengeluaran cairan pada CSPD."

"[ABSTRAK
Latar Belakang : Continous ambulatory peritoneal dialysis (CAPD) telah menjadi
alternatif selain hemodialisis untuk pengobatan penyakit ginjal tahap akhir. Fibrosis
peritoneum merupakan penyebab utama terjadinya kerusakan membran peritoneum.
Mekanisme fibrosis peritoneum belum diketahui secara pasti, namun ditengarai
transforming growth factor ? β (TGF ?β) berhubungan erat terhadap terjadinya fibrosis
peritoneum.
Tujuan : Tujuan penelitian ini adalah untuk mengetahui pengaruh kombinasi ACE
inhibitor (ACEI) dan calcium channel Blocker (CCB) terhadap penurunan ekspresi TGF
? β dan fibrosis peritoneum tikus jantan yang telah dilakukan CAPD.
Metode Penelitian : Penelitian eksperimental, post test only control group design. Tiga
puluh tikus Dawley spraque dibagi menjadi lima kelompok yaitu kelompok kontrol
(kelompok 1) dan kelompok perlakuan dengan pemberian masing-masing cairan CAPD
4,25% (kelompok2) lisinopril 1,44 mg oral dan CAPD (kelompok 3) diltiazem CD 6,48
mg oral dan CAPD (kelompok 4) lisinopril 1,44 mg dan diltiazem CD 6,48 mg oral dan
CAPD (kelompok 5). Setelah 4 minggu tikus dikorbankan dengan cara dislokasi cervical
kemudian diperiksa ekspresi TGF ? β dan terjadinya fibrosis pada peritoneum tikus,
selanjutnya dibuat sediaan histopatologi dan diwarnai dengan hematoksilin eosin serta
imunohistokimia menggunakan antihuman TGF-ß.
Hasil : Dua puluh peritoneum tikus berhasil diperiksa. Rerata skor TGF-β kelompok
kontrol 1,8, kelompok CAPD 2, kelompok lisinopril dan CAPD 1,8, kelompok diltiazem
CD dan CAPD 1,8, kelompok lisinopril dan diltiazem CD dan CAPD 1,7 (p=0,959).
Rerata skor fibrosis peritoneum kelompok kontrol 1,1, kelompok CAPD 2,6, kelompok
lisinopril dan CAPD 1,2, kelompok diltiazem CD dan CAPD 1,3, kelompok lisinopril dan
diltiazem CD dan CAPD1,5 (p=0,268)
Simpulan : Kombinasi lisinopril dan diltiazem mempunyai kecenderungan menurunkan
ekspresi TGF ? β lebih baik dibandingkan lisinopril maupun diltiazem yang diberikan
secara terpisah tetapi tidak bermakna secara statistik. Kombinasi lisinopril dan diltiazem
mempunyai kecenderungan mengurangi fibrosis peritoneum tetapi tidak bermakna secara
statistik dan tidak lebih baik dibandingkan lisinopril maupun diltiazem bila diberikan
secara terpisah.

6,48 mg oral and CAPD (group 5). After 4 weeks, rats sacrificed. Expression of
TGF ? β and peritoneal fibrosis are conducted by histopatology with hematoxillineosin
staining and immunology with anti human-TGF-β.
Result : Twenty peritoneal of rats can be examined. Mean score TGF-β control
group is 1,8, CAPD group is 2, lisinopril and CAPD group is 1,8,diltiazem CD

"Latar belakang: Endometriosis merupakan pertumbuhan jaringan mirip endometrium yang abnormal diluar uterus Studi menunjukkan peran infeksi yang memicu proses inflamasi berhubungan dengan awal mula terjadinya endometriosis. Berbagai macam mikroorganisme penyebab infeksi dari vagina dapat migrasi keatas kemudian menginfeksi dan mengkontaminasi dinding uterus. Akibatnya terjadi akumulasi endotoksin pada cairan mentruasi maupun cairan peritoneal menyebabkan inflamasi dan memicu pertumbuhan endometriosis.
Tujuan: Membuktikan adanya korelasi antara mikroorganisme yang ditemukan pada hasil kultur bilasan vagina dengan mikroorganisme yang ditemukan pada cairan peritoneum hasil laparoskopi perempuan usia reproduksi yang terdiagnosis endometriosis.
Metode: Penelitian ini menggunakan desain penelitian analitik poltong lintang yang bertujuan untuk melihat adanya hubungan korelasi serta mengetahui tingkat korelasi antara mikroorganisme kultur bilasan vagina dengan mikroorganisme pada cairan peritoneum pasien endometriosis.
Hasil: Hasil kultur bilasan vagina dari 31 subjek penelitian yang diteliti, mikroorganisme terbanyak adalah Enterococcus faecalis (32.3%), Eschericia coli (29.1%), dengan 16.1% dengan hasil kultur negatif. Sedangkan dari hasil kultur bilasan peritoneum terdapat 3 subjek (9.6%) dengan hasil positif yaitu dengan jenis bakteri Eschericia coli, Enterococcus faecalis, dan Pseudomonas. Terdapat korelasi lemah antara hasil kultur bilasan vagina dengan kultur bilasan peritoneum (r 0.13). Terdapat korelasi sedang antara kultur positif bilasan vagina dengan nyeri pelvik kronis, korelasi lemah antara kultur positif bilasan vagina dengan nilai Ca 125, dan korelasi lemah antara kultur positif cairan peritoneum dengan tuba kiri yang non paten.
Kesimpulan: Sebagian besar bakteri dari bilasan vagina dan bilasan peritoneum pada pasien endometriosis memiliki hasil bakteri dari organ pencernaan. Terdapat korelasi lemah antara hasil kultur bilasan vagina dengan kultur bilasan peritoneum pada pasien endometriosis.

Background: Endometriosis is an abnormal endometrial like-tissue growth outside the uterus. Studies show the role of infection that triggers the inflammatory process associated with the onset of endometriosis. Various kinds of microorganisms even normal flora causing infection of the vagina can migrate upwards then infect and contaminate the uterine wall. Due to retrogade mestruation, mestrual fluid can entered the peritoneal cavity. As a result, accumulation of endotoxin in menstrual fluid and with retrograde menstruation endotoxins in peritoneal fluid causes inflammation and triggers the growth of endometriosis.
Objective: To prove correlation between microorganisms in vaginal rinse cultures with microorganisms in peritoneal fluid culture in reproductive age women with endometriosis.
Methods: This research use consecutive sampling with 31 subjects reproductive age women with endometriosis who performed surgery procedure. Vaginal bilasan and peritoneal fluid culture were performed. Research was approved by our institutional ethics commitee for health research in 2016.
Results: Results of vaginal rinse culture of 31 subjects studied. Most of vaginal bilasan culture result in gastrointestinal bacteria. Most microorganisms were Enterococcus faecalis (32.3%), Eschericia coli (29.1%), with 16.1% with negative culture results. While the result of peritoneum rinse culture there are 3 subject (9.6%) with positive result that was with Eschericia coli bacteria type, Enterococcus faecalis, and Pseudomonas. There was weak correlation between vaginal rinse culture results and peritoneal rinse culture (r 0.13). There is a correlation between the positive culture of the vaginal rinse with chronic pelvic pain, between the positive culture of the vaginal rinse and Ca 125, and between the positive culture of the peritoneal rinse with the non-patent left tube.
Conclusion: Most of vaginal and peritoneal rinse culture in endometriosis patients result in gastrointestinal bacteria. There was weak correlation between vaginal swab and peritoneal rinse culture.

"Pendahuluan: Endometriosis berkaitan dengan adanya inflamasi  kronik,  gangguan  maturasi  oosit,  peningkatan stress  oksidatif  radikal,  dan  apoptosis  yang  kemudian  mendasari  terjadinya  infertilitas pada wanita usia subur.  Fertilisasi In Vitro (FIV)  merupakan  salah  satu  teknik  yang dapat menangani  infertilitas  dengan  tingkat  keberhasilannya  bergantung  dengan  kualitas  oosit  yang diambil  untuk  menjadi  embrio.  Kualitas oosit dipengaruhi  oleh  nutrisi. Profil  mikronutrien  seperti  vitamin  D  dan  zink  dianggap mempengaruhi  fungsi  reproduksi  melalui aktivitas  anti-inflamasi,  anti-apoptosis dan  anti-oksidan  yang dimiliki. Sayangnya, urgensi untuk menjaga adekuasi nutrisi  ini  sering diabaikan. Selain itu juga hingga saat ini  belum  ada  acuan  untuk  memprediksi  kadar  profil  vitamin  D  dan  zink  dalam serum  dan  cairan  folikular  yang  berhubungan  pada  pasien  endometriosis  itu  sendiri.
Tujuan: Penelitian ini membandingkan kadar vitamin D dan zink di serum dan cairan folikular pada pasien endometriosis dan non-endometriosis.
Metode: Studi ini merupakan studi potong lintang yang dilaksanakan pada pasien Fertilisasi In Vitro (FIV) di Klinik Yasmin, RSCM Kencana selama Juli –  Desember 2020. Data klinis diperoleh melalui rekam medis dan wawancara pasien. Data laboratorium diperoleh melalui sampel darah dan cairan folikuler yang diperoleh bersamaan dengan prosedur Ovum Pick Up (OPU). Sampel kemudian dikelompokkan menjadi kelompok endometriosis dan non-endometriosis. Setelah itu data disajikan dalam tabel dan dianalisis dengan uji parametrik, yaitu uji-t berpasangan bila sebaran data normal atau uji non parametrik, yaitu uji Mann-Whitney bila sebaran data tidak normal. Data dianalisis dengan SPSS 24.
Hasil: Dari jumlah sampel 26 pasien pada studi ini, didapatkan tidak adanya perbedaan bermakna dari vitamin D serum pada pasien endometriosis (22,83 (5,00 – 40,00)) dan non endometriosis (30,11 (10,40-76,10)), namun secara rerata kadar vitamin D serum pada pasien endometriosis lebih rendah. Kadar vitamin D cairan folikular pada pasien endometriosis (15,33 (4,50-36,32)) dan non-endometriosis (23,64 (4,98-60,22)) tidak berbeda bermakna (P>0,05). Kadar zink serum pada pasien endometriosis (75,23 ± 11,58) dan non-endometriosis (79,46 ± 12,09) tidak berbeda bermakna (P>0,05). Konsentrasi zink paada cairan folikular tidak menunjukkan perbedaan bermakna antara pasien endometriosis (39,00 (22,00 – 49,00)) dan tanpa endometriosis (51,00 (19,00-95,00)) (P>0,05). 
Kesimpulan: Tidak ditemukan perbedaan bermakna antara kadar vitamin D serta zink serum dan cairan folikular pada pasien endometriosis dan non-endometriosis. 

Background: Endometriosis associates with chronic inflammation, dysfunction of oocyte maturation, increase of oxidative stress, and apoptosis which underlie infertility problem in reproductive female. In-Vitro Fertilization (IVF) is a technique used to treat infertility with the success rate depending on the quality of the oocytes extracted to become embryos. Oocyte quality is influenced by nutrition. Micronutrient profiles such as vitamin D and zink are thought to influence reproductive function through their anti-inflammatory, anti-apoptotic and anti-oxidant activities. However, the urgency of maintaining nutritional adequacy is often overlooked. In addition, until now there is no reference for predicting levels of vitamin D and zink in serum and follicular fluid associated with endometriosis.
Method: This study is a cross-sectional study conducted on In Vitro Fertilization (FIV) patients at the Yasmin Clinic, RSCM Kencana during July – December 2020. Clinical data were obtained through medical records and patient interviews. Laboratory data were obtained through blood and follicular fluid samples obtained in conjunction with the Ovum Pick Up (OPU) procedure. The samples were then grouped into endometriosis and non-endometriosis groups. After that the data are presented in tables and analyzed by parametric test, namely paired t-test if the data distribution is normal or non-parametric test, namely the Mann-Whitney test if the data distribution is not normal. Data were analyzed by SPSS 24.
Result: From a total sample of 26 patients in this study, there was no significant difference in serum vitamin D in patients with endometriosis (22.83 (5.00 – 40.00)) and non-endometriosis (30.11 (10.40-76.10). )), but the mean serum vitamin D level in endometriosis patients was lower. Follicular fluid vitamin D levels in patients with endometriosis (15.33 (4.50-36.32)) and non-endometriosis (23.64 (4.98-60.22)) were not significantly different (P>0.05) . Serum zink levels in patients with endometriosis (75.23 ± 11.58) and non-endometriosis (79.46 ± 12.09) were not significantly different (P>0.05). Zink concentration in follicular fluid did not show a significant difference between endometriosis patients (39.00 (22.00 – 49.00)) and without endometriosis (51.00 (19.00-95.00)) (P>0.05) .
Conclusion: There was no significant difference between the levels of vitamin D and serum zink and follicular fluid in patients with endometriosis and non-endometriosis.

"Endometriosis merupakan penyakit inflamasi kronis pada organ reproduksi wanita dengan gejala utama nyeri pelvis kronis, dismenore, dan dispareunia yang dapat disebabkan oleh stres oksidatif akibat rendahnya kadar antioksidan, seperti vitamin C, sehingga terjadi kerusakan sel. Levonorgestrel adalah terapi hormonal yang sering digunakan untuk meredakan rasa nyeri pada endometriosis, tetapi dapat memperberat proses inflamasi, sehingga dibutuhkan suatu terapi adjuvan, seperti propolis yang mengandung antioksidan yang tinggi. Penelitian ini menggunakan desain uji klinis dengan alokasi acak dan tersamar ganda. Subjek penelitian adalah 24 wanita yang sedang mendapatkan terapi implan levonorgestrel dan diminta untuk menerima propolis atau plasebo dua kali sehari dengan dosis 1 tetes/10 kg berat badan (kgBB) per kali. Sampel darah kemudian diambil pada 4 minggu setelah intervensi dan dilakukan pemisahan serum. Pengukuran kadar vitamin C serum dilakukan dengan metode spektrofotometri dan analisis statistik dilakukan dengan uji t tidak berpasangan apabila data berdistribusi normal. Hasil penelitian menunjukkan bahwa terdapat perbedaan yang signifikan antara kedua kelompok (p<0,001), yaitu kadar vitamin C serum lebih tinggi pada kelompok propolis (0,202+0,057) dibandingkan kelompok plasebo (0,069+0,028). Dengan demikian, pemberian propolis meningkatkan kadar vitamin C serum pada pasien endometriosis setelah intervensi 4 minggu. 

Endometriosis is a chronic inflammatory reproductive disease in women which main symptoms are chronic pelvic pain, dysmenorrhea, and dyspareunia that can be triggered by oxidative stress due to decreased antioxidants, such as vitamin C that may cause cell damage. Levonorgestrel is a hormonal therapy that is commonly used for endometriosis to relieve pain but it can worsen the inflammatory process, so an adjuvant therapy is needed, such as propolis that contains high antioxidant level. This study used clinical trial design with random allocation and double-blinded. The study subject is 24 women that receive levonorgestrel therapy and were asked to consume propolis or placebo randomly two times a day with a dose of 1 drop/10 kg body weight (kgBW) per time. Blood samples were then taken after 4 weeks and serum separation was performed. Serum vitamin C levels were measured using spectrophotometric method and statistical analysis used independent t-test if the data were normally distributed. The result showed that there is a significant difference between the two groups (p<0,001), in which the concentration of serum vitamin C is higher in the propolis group (0,202+0,057) compared to the placebo group (0,069+0,028). In conclusion, the administration of propolis results in significantly higher serum vitamin C concentration after 4-week intervention."

"Latar belakang: Resistensi progesteron akibat gangguan ekspresi reseptor progesteron pada jaringan endometriosis telah diketahui menjadi faktor yang memperberat kondisi klinis pasien endometriosis. Tujuan penelitian ini adalah untuk menganalisis tingkat metilasi DNA pada promoter gen PR-B pada berbagai jaringan endometriosis seperti eutopik endometrium, lesi ektopik peritoneum, endometrioma dan darah menstruasi serta pengaruhnya terhadap ekspresi mRNAnya dibandingkan dengan kontrol endometrium normal; untuk mengetahui patomekanisme endometriosis pada berbagai lokasi terkait dengan resistensi progesteron.
Metode: Penelitian ini menggunakan desain potong lintang yang melibatkan 20 sampel untuk masing-masing kelompok kasus dan kontrol. Tingkat metilasi DNA dari gen PR-B diukur menggunakan metode Methylated Specific PCR MSP lalu intensitas pita di dalam gel agarose dihitung dengan software ImageJ. Presentase intensitas pita pada sampel dibandingkan dengan kontrol positif disebut dengan tingkat metilasi DNA. Pengukuran ekspresi relatif mRNA PR-B menggunakan qRT-PCR dua tahap dan analisis dilakukan dengan metode Livak.
Hasil: Dari penelitian ini didapatkan perbedaan bermakna antara tingkat metilasi DNA gen PR-B pada jaringan endometriosis ektopik peritoneum 72,4 termetilasi , endometrioma 85 termetilasi dan eutopik endometrium 72,21 termetilasi dibandingan dengan kontrol p

Background: Progesterone resistance, due to alteration of progesterone receptor PR expression in endometriosis, was known as a disrupt factor in response to progesterone. The aim of this study is to analyze DNA methylation level on PR B promoter in various tissues include eutopic endometrium, ectopic peritoneal, endometrioma and menstrual blood from endometriosis patient as well as the implication on it's mRNA relatif expression compare with normal endometrium control to know the patomechanisms of endometriosis in various lession in term of progesterone resistance.
Methods: It was a cross sectional study, involved 20 sample for both patient and control. DNA isolate from each sample were converted by bisulfite conversion. DNA methylation level of PR B gene was analysis by Methylated Specific PCR MSP method, then band intensity in gel agarose was measured by ImageJ software. Percentage of band intensity in sample compared with positive control was determined as DNA methylaton level. Quantitative real time PCR was conducted to assess expression of mRNA PR B for each sample and Livak method was used to analysis it's relatif expression compare with control.

"Endometriosis adalah pertumbuhan jaringan mirip endometrium di luar uterus. Jaringan ini memiliki kemampuan tertanam di berbagai tempat ektopik karena dipengaruhi sistem aktivator plasminogen yang berperan dalam proses fibrinolisis. Pada endometriosis terdapat ekspresi plasminogen activator inhibitor-1 (PAI-1) berlebih yang menyebabkan kurangnya fibrinolisis sehingga menyebabkan terbentuknya produk fibrin terdegradasi yang dapat mempengaruhi penempelan dan perkembangannya. Faktor epigenetik perubahan tingkat metilasi DNA berperan pada patogenesis endometriosis.
Penelitian ini bertujuan untuk menilai tingkat metilasi gen PAI-1 dan hubungannya dengan perkembangan jaringan endometriosis ovarium dan peritoneum. Studi potong lintang ini menggunakan 13 sampel wanita endometriosis ovarium, 5 wanita endometriosis peritoneum, dan 8 wanita tanpa endometriosis. DNA dari sampel diisolasi, dilakukan konversi bisulfit, kemudian diamati tingkat metilasi DNAnya dengan metode methylation specific polymerase chain reaction (MSP). Hasilnya dianalisis dengan uji Kruskal-Wallis dan uji Mann-Whitney. Terdapat perbedaan yang signifikan tingkat metilasi DNA gen PAI-1 pada ketiga kelompok sampel (p<0,05).
Penelitian ini menemukan perbedaan signifikan antara endometriosis ovarium dan peritoneum dibandingkan dengan kontrol (p=0,006 dan p = 0,003); namun tidak ada perbedaan yang signifikan pada endometriosis peritoneum dibandingkan dengan ovarium (p>0,05). Penelitian kami menunjukkan rendahnya tingkat metilasi gen PAI-1 yang dapat meningkatkan ekspresi gen PAI-1 dan hal ini disugestikan dapat berkontribusi sebagai faktor risiko endometriosis pada ovarium dan peritoneum."

"Pendahuluan : Infertilitas merupakan salah satu gejala pada endometriosis dengan prevalensi mencapai 40-50%. Endometriosis memiliki dampak merugikan terhadap kualitas oosit, sementara sampai saat ini belum ada biomarker baik dari serum ataupun cairan folikel yang dapat dijadikan acuan penilaian kualitas oosit untuk dapat digunakan pada pasien endometriosis yang menjalani fertilisasi in vitro (FIV). Telah ditemukan bahwa pada serum pasien endometriosis terjadi perubahan ekspresi microRNA dimana miRNA-125b memiliki peningkatan yang paling signifikan dengan sensitifitas dan spesifisitas yang paling tinggi. Pada cairan folikel, miRNA-125b berperan saat transisi fase folikular-luteal dengan mempengaruhi ekspresi leukemia inhibitory factor (LIF). LIF diketahui dapat menginduksi sel kumulus yang kemudian mempengaruhi maturasi oosit.
Tujuan: Tujuan penelitian ini adalah untuk mencari apakah terdapat hubungan antara miRNA-125b serta LIF dengan kualitas oosit pada pasien infertil dengan endometriosis.
Desain: Studi Analitik korelatif dengan desain potong lintang.
Material dan Metode: Sampel penelitian didapatkan dari 31 pasien infertil dengan endometriosis yang menjalankan program FIV di Klinik Yasmin RSCM Kencana, dan Klinik Melati RSAB Harapan Kita. Sesaat sebelum petik ovum, sebanyak 5cc sampel darah dari setiap pasien akan diambil untuk penilaian ekspresi miRNA-125b. Pada saat petik ovum, sebanyak 10cc dari total cairan dari folikel yang didapat akan diambil untuk penilaian ekspresi miRNA-125b dan kadar LIF. Oosit yang didapat dinilai oleh embriolog. Pemeriksaan ekspresi miRNA dilakukan dengan RT-PCR, dan kadar LIF menggunakan metode sandwich ELISA.
Hasil:Terdapat korelasi negatif antara miRNA-125b serum dengan LIF cairan folikel (p=0,042; r=-0,34). Tidak terdapat korelasi antara miRNA-125b serum dengan miRNA-125b cairan folikel. Tidak terdapat korelasi antara miRNA-125b cairan folikel dengan LIF cairan folikel. Tidak terdapat korelasi antara miRNA-125b serum, miRNA-125b cairan folikel, dan LIF cairan folikel dengan kualitas oosit. Terdapat hubungan yang bermakna antara kadar ekspresi miRNA-125b cairan folikel dengan angka kehamilan biokimia.
Kesimpulan: Terdapat ekspresi miRNA-125b pada serum dan cairan folikel pada pasien endometriosis, namun miRNA-125b belum dapat dijadikan sebagai parameter yang kuat untuk pemeriksaan kualitas oosit pada pasien endometriosis yang menjalani FIV.

Introduction : Infertility is one of the symptoms in endometriosis with prevalence reaching 40-50%. Endometriosis is known to have detrimental effect on oocyte quality, yet until now there is no biomarker derived from either serum, or even follicular fluid, which can be used as reference for oocyte quality assessment in endometriosis patients going through in vitro fertilization (IVF) procedures. Changes of some microRNAs expression has been found in serum of endometriosis patients, with miRNA-125b showing the most significant increase with the highest sensitivy and specificity. In follicular fluid, miRNA-125b play role during follicular-lutheal phase transition by targeting the expression of Leukemia Inhibitory Factor (LIF). LIF has been studied to have the ability to induce cumulus cell expansion which in turn will affect the oocytes maturation.
Purpose: The purpose of this study is to observe the correlation between miRNA-125b, LIF, and oocyte quality in infertile patient with endometriosis.
Design: this is a cross-sectional study with correlation analysis method.
Materials and Methods: in this study, samples were collected from 31 infertile women with endometriosis undergoing in vitro fertilization procedure at Yasmin Clinic of Dr. Cipto Mangunkusumo Hospital, and Melati Clinic of Harapan Kita Mother and Child Hospital. Shortly prior to ovum pick up (OPU) procedure, 5cc of blood sample from each patients was collected, and 10 cc of total follicular fluid was obtained during OPU. Harvested oocytes during the procedure were assessed and scored by embryologist. MiRNA-125b expressions from serum and follicular fluid samples were analyzed using RT-PCR, and LIF levels were analized using ELISA sandwich method.
Result: negative correlation was found between the expression of miRNA-125b serum and LIF follicular fluid (p=0,042; r=-0,34). No correlation was found between the expression of miRNA-125b in serum and in follicular fluid, as well as the expression of miRNA-125b in follicular fluid and LIF in follicular fluid. No correlation was found between the expression of miRNA-125b in serum, follicular fluid, also LIF in follicular fluid, with oocyte quality. Significant result was found between the expression of miRNA-125b in follicular fluid and biochemical pregnancy rate.
Conclusion: This study found miRNA-125 expression represented in serum and follicular fluid in endometriosis patient, but it still cannot be used as a strong parameter for assessing the oocyte quality in infertile women with endometriosis
