Triclustering merupakan salah satu teknik data mining pada data tiga dimensi untuk mengelompokkan data secara bersamaan pada baris dan kolom di titik waktu yang berbeda menjadi tricluster. Metode ini umumnya diterapkan pada bidang bioinformatika, khususnya data ekspresi gen tiga dimensi. Salah satu triclustering dengan pendekatan biclustering-based adalah THD-Tricluster. Langkah utama dari algoritma ini ialah generate bicluster dan genereate tricluster. Algoritma THD-Tricluster menggunakan pola pergeseran dan penskalaan dengan nilai Shifting-and-Scaling-Similarity (SSSim) untuk mengelompokkan gen dan menghasilkan tricluster. Hasil dari THD-Tricluster dievaluasi dengan Multi Slope Measure (MSL) yaitu sebuah pengukuran kualitas melalui representasi grafik dari tricluster. Dalam penelitian ini, data yang digunakan adalah data respon tiga sel individu terhadap pemberian sitokin berupa interleukin-1-beta pada sel mesenkim amnion manusia atau sel pada membran janin. Sitokin memicu regulasi gen inflamasi yang berkontribusi pada kelahiran prematur. Metode THD-Tricluster diimplementasikan pada 15 skenario dengan nilai threshold berbeda. Skenario yang optimal dipilih menggunakan nilai validasi coverage. Pada skenario optimal, diperoleh delapan tricluster yang kemudian dievaluasi menggunakan Multi Slope Measure (MSL). Tricluster 2 yang memiliki nilai MSL paling kecil dan dipilih sebagai tricluster optimal terdiri atas kumpulan gen dari sel yang responsif terhadap pemberian sitokin berupa interleukin-1-beta. Gen-gen pada Tricluster 2 inilah yang dapat digunakan sebagai bahan pertimbangan bagi para peneliti di bidang biologis dan medis untuk untuk penelitian lebih lanjut terkait kelahiran prematur. ......Triclustering is one of the data mining techniques on three-dimensional data to cluster data simultaneously in rows and columns at different time points into triclusters. This method is generally applied to the field of bioinformatics, especially three-dimensional gene expression data. One of the triclustering methods with a biclustering-based approach is THD-Tricluster. The main steps of this algorithm are generate bicluster and generate tricluster. THD-Tricluster algorithm uses shifting and scaling patterns with Shifting-and-Scaling-Similarity (SSSim) values to cluster genes and generate tricluster. The result of THD-Tricluster is evaluated by Multi Slope Measure (MSL), a measurement of tricluster quality through graphical representation. In this study, the data used is the response data of three individual cells to cytokine in the form of interleukin-1-beta in human amniotic mesenchymal cells or cells in the fetal membrane. Cytokines stimulate the regulation of inflammatory genes that contribute to preterm birth. The THD-Tricluster method was implemented on 15 scenarios with different threshold values. The optimal scenario was selected using the coverage validation value. In the optimal scenario, eight triclusters were obtained which were then evaluated using Multi Slope Measure (MSL). Tricluster 2 which has the smallest MSL value and selected as the optimal consists of a collection of genes from cells that are responsive to cytokine administration in the form of interleukin-1-beta. The genes in Tricluster 2 can be used by biological and medical researchers to develop treatments to prevent premature birth.

Latar Belakang: Di era ini, terapi dengan sel punea untuk mengobati berbagai penyakit semakin dirninati. Sejauh ini, banyak pendapat mengenai keamanan sel punea, yang dikatakan aman untuk manusia. NaIllun" belum ada penelitian lebih jauh mengenai keamanan sel punea yang disuntikan lewat vena, untuk kesehatan pembuluh darah. Tujuan dari peneiitian ini adalah untuk menganalisis efek sel punea yang disuntikkan secara IV, pada pembuluh darah arteri yang nannal. Metode: Data didapatkan dari eksperimen klinik tikus Wistar yang dilaksanakan di Instilut Pertanian Bogor, merupakan studt awal pada tikus dengan tekanan darah normal. Tikus dengan tekanan darah nonnal (140/100 mmHg, diukur dengan CODA) tersebut dibagi menjadi 3 kelompok. Kelompok pertama ada kelompok kontrol, kelompok kedua disumikan secara IV sel punca 1 x 106 , dan kelompok keliga 3xl 06 sel, dalam sekali penyuntikan. Kelompok kedua dan ketiga diamati selama 1 bulan setelah penyuntikan sel punca. Setelah 1 bulan, tikus dinekropsi dalam keadaan anastesi. Selanjutnya, dengan bantuan J Image Software, dilakukan pcngamatan terhadap diameter dan ketebalan dinding dari meri karotis, karotis interna, karotis eksterna, aorta abdominal, iliaka kiri, dan iliaka kanan. Analisis statistik dilakukan dengan metode ANOV A terhadap 3 kelompok tikus untuk. mengukur perbedaan diameter pembuluh darah dan ketebalan dinding pembulu darah. Jika persebaran data tidak rata, maka digunakan metode T Independent Test. Hasil: Hasil analisis menunjukkan bahwa tidak ada perbedaan signifikan dalam diameter dan ketebalan dinding untuk kelompok I, kelompok 2, dan kelompok 3 untuk semua pembuluh darah kecuali arteri iliaka kiri. Arteri iliaka kiri menunjukkan perbedaan yang bennakna dalam diameter dan ketebalan pembuluh darah. Diskusi: Hasil riset ini, memmjukkan keamanan sel punca pada arteri bila disuntikan ke dalam vena. Ini ditunjukkan dengan hasil yang tidak. terdapat perbedaan bermakna dalam 5 pembuluh darah yang dipenksa. kecuali arteri iliaka kiri menunjukkan hasil yang bermaknadan perlu diteliti lebih lanjutdenganjumlah sam pel yang memadai (30 tikus). Setiap kelompok mempunyai sampel minimum 9 ekor, namun dan hasil penelitian masing-masing kelompok mempunya 5 ekor tikus yang dapat dianalisis. Sehingga hasil analisis yang didapatkan tidak bisa mencerminkan hubungan yang kuat secara statistik, namun merupakan suatu kecenderungan.

Pendahuluan: Periodontitis merupakan suatu penyakit inflamasi terkait bakteri yang dikarakteristikan dengan interaksi antara sistem pertahanan tubuh host dan patogen. Porphyromonas gingivalis (P.gingivalis) merupakan salah satu bakteri red complex yang berperan dalam menginisiasi terjadinya periodontitis. Gingiva merupakan lini pertahanan mekanis, yang disebut sebagai gingival barrier function. Tight junction gingiva merupakan salah satu yang berperan dalam fungsi pertahanan tersebut dan kemampuan tubuh dalam merespon patogen dipengaruhi oleh usia. Tujuan: Mendapatkan perbedaan ekspresi gen tight junction gingiva terhadap stimulasi P.gingivalis. Metode: Studi eksperimental menggunakan jaringan gingiva tikus usia 18 minggu dan 35 minggu. Sampel kemudian dikultur dan diberikan stimulasi live P.gingivalis dan hasil sekresi P.gingivalis. Pengukuran dilakukan untuk menilai laju proliferasi, laju metabolisme, dan ekspresi tight junction sel gingiva tikus. Hasil: Sel gingiva tikus tua menunjukkan laju proliferasi yang lebih tinggi dibandingkan sel gingiva tikus tua, namun pada uji statistik tidak terdapat perbedaan signifikan pada kedua kelompok. Terdapat kecenderungan peningkatan laju metabolisme dan ekspresi gen tight junction gingiva yang lebih tinggi pada sel gingiva tikus muda dibandingkan sel gingiva tikus tua, namun pada uji statistik tidak terdapat perbedaan signifikan pada kedua kelompok. Kesimpulan: Tidak terdapat perbedaan ekspresi gen tight junction gingiva terkait usia. ......Introduction: Periodontitis is an inflamatorry disease associated with the interaction of host immune systemn and pathogen. Porphyromonas gingivalis is one of the red complex bacteria that plays important role in initiating periodontitis. Gingiva act as mechanical defense towards the pathogen, which is known as the gingival barrier function,including gingival tight junctions. Body’s ability to respond stimuli and environmental condition is influenced by age, this also affect the respond of host immune system to pathogens. Objective: To analyse the tight junction gene expression to Porphyromonas Gingivalis intervention. Material and Methods: Aging experimental model was conducted by using two age categories male rodents, 18 and 58 weeks. Rodents gingival cell was intervened with Porphyromonas gingivalis and its product from the broth medium. Meassurements were made to analyse the proliferation rate, metabolic rate, and gingival tight junction gene expression. Result: The old rodent group shows higher proliferation rate, but there was no statistically differences between two groups. There was a tendency of increase value for the metabolic rate and gingival tight junction gene expression in young rodent group compare to old rodent group. Conclusion: There was no differences of the gingival tight junction expression in related to aging.

Caveolae are 50-100 nm flask-shaped invaginations of the plasma membrane that are primarily composed of cholesterol and sphingolipids. Using modern electron microscopy techniques, caveolae can be observed as omega-shaped invaginations of the plasma membrane, fully-invaginated caveolae, grape-like clusters of interconnected caveolae (caveosome), or as transcellular channels as a consequence of the fusion of individual caveolae. The caveolin gene family consists of three distinct members, namely Cav-1, Cav-2 and Cav-3. Cav-1 and Cav-2 proteins are usually co-expressed and particularly abundant in epithelial, endothelial, and smooth muscle cells as well as adipocytes and fibroblasts. On the other hand, the Cav-3 protein appears to be muscle-specific and is therefore only expressed in smooth, skeletal and cardiac muscles. Caveolin proteins form high molecular weight homo- and/or hetero-oligomers and assume an unusual topology with both their N- and C-terminal domains facing the cytoplasm.
