"Hiperurisemia adalah keadaan dimana tingginya kadar asam urat dalam tubuh. Salah satu penyebab tingginya kadar asam urat adalah aktifnya xantin oksidase. Pengobatan untuk kondisi tersebut yaitu dengan menghambat kerja xantin oksidase sehingga produksi asam urat berkurang. Daun salam (Syzygium polyanthum Wight.) merupakan salah satu contoh pengobatan tradisional dalam mengurangi produksi asam urat. Tujuan penelitian ini untuk memberikan bukti ilmiah mengenai aktivitas fraksi n-heksan, etil asetat, n-butanol, dan metanol dalam menghambat xantin oksidase pada Syzygium polyanthum Wight. Serbuk simplisia diekstraksi dengan cara maserasi menggunakan metanol 80%, kemudian dilakukan fraksinasi dengan pelarut n-heksana, etil asetat, n-butanol dan metanol secara berurut-urut. Diketahui dalam penelitian ini, fraksi yang paling aktif memiliki aktivitas penghambatan xantin oksidase adalah fraksi etil asetat dengan nilai IC50 sebesar 18,43 μg/mL. Uji kinetika enzim menunjukkan bahwa fraksi etil asetat memiliki tipe penghambatan kompetitif. Pada identifikasi golongan senyawa, fraksi etil asetat memiliki senyawa alkaloid, tanin, saponin, flavonoid, polifenol, glikosida, dan terpen.

Hyperuricemia is condition where the uric level in the body is high. One of the reason for the condition that uric acid level in blood is high is active of xanthine oxidase. The treatment for this condition is by inhibiting the xanthine oxidase so that the production of uric acid can be reduced. Syzygium polyanthum Wight. leaves is an example of traditional medicine for reducing uric acid production.

The purpose of this study to provide scientific evidence related with fraction nhexane, ethyl acetate, n-butanol, and methanol to inhibit xanthine oxidase activity in Syzygium polyanthum Wight. leaves. The simplicia powder was macerated by methanol 80%, and then fractionated successively by n-hexane, ethyl acetat, nbutanol, and methanol.

Research’s result shows that the most active fraction proved to give the best inhibitory activity of xanthine oxidase is the fraction of ethyl acetate with IC50 value of 18,43 μg/mL. Kinetic enzyme assay showed that ethyl acetate fraction had competitive inhibitory type. Phytochemical identification shows that the fraction of ethyl acetate contains alkaloid, tannins, saponin, flavonoids, polyphenol, glycosides, and terpene."
