"Untuk mencapai sirkulasi sistemik absorpsi suatu obat harus melewati satu atau beberapa membran sel. Sifat membran dan struktur molekul obat sangat berhubungan dengan permeabilitas obat. Tujuan penelitian ini untuk melihat pengaruh penambahan surfaktan tween 80 terhadap laju absorpsi ibuprofen secara in vitro menggunakan alat absorption simulator. Ibuprofen sebanyak 500 mg dalam 100 ml cairan lambung buatan tanpa enzim pH 1,0; 3,0 atau cairan usus buatan tanpa enzim pH 6,5 pada kompartemen I akan diabsorpsi secara difusi pasif ke kompartemen II (100 ml cairan plasma simulasi pH 7,4). Membran artifisial pada percobaan ini dibuat dari kertas penyangga bentuk bulat tipe GV 0,22 μm (Milipore), dengan luas permukaan 13,2 cm2 yang diimpregnasi dengan campuran lesitin-kolesterol (1:1) dalam pelarut parafin cair. Percobaan absorpsi berlangsung 5 jam pada suhu 37°C ± 1°C dan sampel diambil pada jam ke-1, 3, dan 5, kemudian dianalisis menggunakan spektrofotometer UV-Vis pada λ 264 dan 263,5 nm. Hasil percobaan menunjukan adanya peningkatan ibuprofen yang diabsorpsi dari kompartemen I ke kompartemen II dengan meningkatnya jumlah konsentrasi tween 80. Jumlah ibuprofen yang diabsorpsi dari kompartemen I ke kompartemen II paling besar terjadi pada cairan usus buatan pH 6,5. Tween 80 dapat meningkatkan kelarutan ibuprofen pada konsentrasi maksimal 1,5%.

Drugs have to pass one or more cell membranes in order to reach systemic absorption. Cell membrane and molecular structure characteristics are strongly related to drugs permeability. The objective of this research is to evaluate the effect of surfactant tween 80 addition based on in vitro absorption rate of ibuprofen by using absorption simulator. 500 mg Ibuprofen is added to simulation gastric liquid without enzim pH 1.0; 3.0 or simulation colon liquid without enzim pH 6.5 at compartment I that will be absorbed by passive diffusion to compartment II (100 ml plasma liquid simulation pH 7.4). Artificial membrane in this trial is made of spherical buffer paper GV 0.22 μm type (milipore) with 13.2 cm2 surface area that was impregnated to lecithin-cholesterol mixture in liquid paraffin solution. Absorption experiment has been conducted for 5 hours at temperature 37oC+1oC and the mixture is sampled at the first hour, 3rd hour and 5th hour. Then it is analyzed using spectrophotometer UV-Vis with λ 264 and 263,5 nm. Experiment result shows intensity of ibuprofen that is absorbed from compartment I to compartment II with increment of tween 80 intensity at compartment I. Ibuprofen intensity that was absorbed from compartment I to compartment II will be increased due to the increment of pH simulation liquid."
