"Xilitol merupakan senyawa gula polialkohol dengan lima atom karbon. Senyawa tersebut memiliki banyak manfaat dan telah digunakan secara luas dalam industri makanan, farmasi, dan kesehatan. Sumber karbon yang berlimpah seperti lignoselulosa dapat dimanfaatkan untuk produksi xilitol. Salah satu sumber karbon yang potensial dan prevalensinya tinggi di Indonesia adalah eceng gondok. Tanaman ini dikenal sebagai gulma dan kurang dimanfaatkan oleh masyarakat.
Tujuan dari penelitian ini adalah pemanfaatan tanaman eceng gondok yang mengandung hemiselulosa sebagai substrat dalam biokonversi xilosa menjadi xilitol oleh sel khamir Debaryomyces hansenii. Tahapan optimasi meliputi optimasi hidrolisis dari eceng gondok menggunakan response surface method dan optimasi kondisi fermentasi.
Kondisi hidrolisis yang optimum diperoleh dengan perbandingan bobot eceng gondok terhadap larutan asam oksalat 8% 1:7,5 (b/v) selama 75 menit pada suhu 121oC. Kondisi tersebut menghasilkan yield sebesar 20,66%. Kondisi optimum fermentasi produksi xilitol oleh Debaryomyces hansenii dicapai dengan waktu kultivasi selama empat hari, kondisi aerasi terbatas, dan penambahan ion logam CaCl2.2H2O 0,01%. Kondisi tersebut menghasilkan yield value sebesar 77,43%.

Xylitol is a five-carbon polyol sugar. It has many healthy benefits and is widely used in food, pharmaceutical, and healthcare. Sources with abundant carbon such as lignocellulose can be used for xylitol production. One of the potencial sources with high prevalency in Indonesia is water hyacinth. It is known as weeds and has not been fully utilized by people.
The aim of this research is the utilization of water hyacinth which contains hemicellulose as a substrate in the bioconversion of xylose into xylitol by Debaryomyces hansenii yeast. Stages of processing include the optimization of water hyacinth hydrolysis using response surface method and optimization of fermentation conditions.
Optimum hydrolysis condition obtained by water hyacinth mass to 8% oxalic acid volume ratio of 1:7.5 (w/v) for 75 minutes at temperature of 121oC. The condition obtained yield of 20.66%. The optimum fermentation condition for xylitol production by Debaryomyces hansenii was achieved by four day cultivation, limited aeration condition, and addition of metal ions CaCl2.2H2O 0.01%. The condition obtained yield value of 77.43 %.

"ABSTRACT
Xylitol is a five-carbon polyol sugar. It has many healthy benefits and is widely used in food, pharmaceutical, and healthcare. Natural sources with abundant carbon such as lignocellulose can be used for xylitol production. One of the potencial sources with high prevalency in Indonesia is water hyacinth. It is known as weeds and has not been fully utilized by people. The aim of this research was the utilization of water hyacinth which contains hemicellulose as a substrate in the bioconversion of xylose into xylitol by yeast cells Debaryomyces hansenii. Stages of processing include the optimization of water hyacinth hydrolysis using autohydrolysis method and optimization of fermentation conditions. Xylose and xylitol were determined by HPLC with RI detector and LiChrosorb® NH2 (4 mm x 125,00 mm, 5μm) column. Acetonitrile-water (90:10, v/v) was used as a solvent. 20 μL sample volume was injected at flow rate of 1.0 mL/min and room temperature. The results showed that optimum conditions for the acquisition of xylose were obtained through autohydrolysis methods for 75 minutes with 1:15 water hyacinth and water ratio and posthydrolysis for 45 min using 4% sulfuric acid. Xylose concentration in hydrolyzate obtained was 25.55 g/L. The optimum fermentation condition for xylitol production was achieved by four day cultivation, limited aeration condition, and addition of metal ions CaCl2.2H2O 0.01%. The yield of xylitol obtained using those conditions was 77.43 %. "
