"Latar Belakang
Penuaan merupakan suatu proses yang tidak dapat dihindari. Namun, pencegahan penyakit terkait usia yang seringkali menyertainya dapat dilakukan dengan memodifikasi faktor risiko utamanya, termasuk inflamasi. Salah satu pendekatan yang dapat digunakan adalah dengan memanfaatkan tanaman yang kaya antioksidan. Rosella (Hibiscus sabdariffa Linn.), terbukti memiliki aktivitas antioksidan yang tinggi. Namun, saat ini belum ada penelitian spesifik yang menilai dampaknya terhadap biomarker khusus penuaan seperti Sirtuin 1.
Metode
Penelitian eksperimental in vivo ini dilakukan pada 18 tikus Sprague Dawley jantan yang dibagi secara acak kedalam 3 kelompok: kontrol (K) yang merupakan tikus muda normal yang diberi pakan standar; Accelerated Aging (AA) yang diberi pakan tinggi lemak dan D-galaktosa; dan Hibiscus sabdariffa Linn. (HSL) yang diberi pakan tinggi lemak, D-galaktosa, dan ekstrak etanol bunga H. sabdariffa dengan dosis 400 mg/KgBB. Perlakuan dilakukan selama 6 minggu. Pada akhir penelitian, kadar SIRT1 diukur menggunakan kit ELISA standar.
Hasil
Perbedaan rerata kadar SIRT1 yang diperoleh antar kelompok tidak signifikan secara statistik.
Kesimpulan
Ekstrak HSL belum mampu meningkatkan kadar SIRT1.

Introduction
Aging is an inevitable process. However, prevention of age-related diseases that often follows can be done by modifying its main risk factors, including inflammation. One approach that can be used is to utilize plants that are rich in antioxidants. Rosella (Hibiscus sabdariffa Linn.), has been shown to have high antioxidant activity. However, there is currently no specific study that assesses its impact on a specific aging biomarker like Sirtuin 1.
Method:
This in vivo experimental study was conducted on 18 male Sprague Dawley rats that were randomly divided into 3 groups: control (K) which were normal young rats that are fed with standard feed; Accelerated Aging (AA) which were fed high fat feed and D-galactose; and Hibiscus sabdariffa Linn. (HSL) which were fed high fat feed, D-galactose, and the ethanol extract of H. sabdariffa flowers at a dose of 400 mg/KgBW. The treatment was carried out for 6 weeks. At the end of the study, SIRT1 levels were measured using a standard ELISA kit.
Results:
The difference in mean SIRT1 levels obtained between groups was not statistically significant.
Conclusion:
HSL extract has not yet been able to increase SIRT1 levels.

"Penuaan kulit merupakan perubahan biologis yang disebabkan oleh faktor intrinsik, seperti akumulasi waktu, dan dapat dipercepat oleh faktor ekstrinsik, seperti paparan sinar ultraviolet dan gaya hidup. Faktor-faktor tersebut menjadi penyebab terjadinya stres oksidatif yang merupakan mekanisme utama dalam penuaan kulit dan dapat mengarah pada disfungsi mitokondria. Stres oksidatif dapat disebabkan oleh metilglioksal (MGO) yang meningkat pada kondisi hiperglikemik. Stres oksidatif dan disfungsi mitokondria dapat dicegah menggunakan liquiritin yang memiliki sifat antioksidan dan antiinflamasi. Strategi sel dalam mengatasi stres oksidatif diregulasi oleh beberapa gen, di antaranya Pgc-1a, Sirt1, dan Foxo3. Penelitian ini menggunakan sel fibroblas L929 yang berasal dari mencit dan umum digunakan untuk uji sitotoksisitas dan biologi sel. Belum ada penelitian yang membahas potensi liquiritin sebagai antipenuaan. Oleh karena itu, penelitian ini bertujuan untuk mengetahui potensi antipenuaan liquiritin terhadap viabilitas, migrasi sel, dan ekspresi gen Pgc-1a, Sirt1, dan Foxo3. Metode pada penelitian ini mencakup uji viabilitas dengan MTS, uji migrasi (scratch assay dan transwell migration assay), dan uji ekspresi gen menggunakan qPCR. Pada hasil viabilitas, konsentrasi MGO 1.000 µM menurunkan viabilitas secara signifikan dan konsentrasi liquiritin 5; 7,5; dan 10 µM mampu meningkatkan viabilitas sel fibroblas L929 yang diinduksi oleh MGO. Hasil uji migrasi menunjukkan peningkatan penutupan luka dan jumlah sel yang bermigrasi meningkat setelah diberi liquiritin. Ekspresi gen Pgc-1a, Sirt1, dan Foxo3 memiliki pola peningkatan ekspresi yang sama, yaitu optimal pada konsentrasi 5 µM. Berdasarkan hasil penelitian, liquiritin memiliki potensi sebagai antipenuaan.

Skin aging is a biological process caused by intrinsic factors, such as the accumulation of time, and can be accelerated by extrinsic factors like ultraviolet exposure and lifestyle. These factors trigger oxidative stress, which is a key mechanism in skin aging and can lead to mitochondrial dysfunction. One contributor to oxidative stress is methylglyoxal (MGO), whose levels increase under hyperglycemic conditions. Oxidative stress and mitochondrial dysfunction may be prevented by liquiritin, a compound with known antioxidant and anti-inflammatory properties. The cellular strategies to counter oxidative stress are regulated by several genes, including Pgc-1a, Sirt1, and Foxo3. This study used L929 fibroblast cells, which are mouse-derived and commonly applied in cytotoxicity and cell biology assays. There has been no research discussing the potential of liquiritin as an anti-aging agent. Therefore, this study aimed to evaluate the anti-aging effects of liquiritin on cell viability, cell migration, and gene expression of Pgc-1a, Sirt1, and Foxo3. The methods included MTS assay for viability, scratch assay and transwell migration assay for cell migration, and qPCR for gene expression analysis. Results showed that 1.000 µM MGO significantly reduced cell viability, while liquiritin at 5; 7,5; and 10 µM restored the viability of MGO-induced L929 cells. Migration assays revealed enhanced wound closure and increased migrating cells after liquiritin treatment. Gene expression analysis showed a similar increase in Pgc-1a, Sirt1, and Foxo3, with optimal expression at 5 µM. These findings suggest that liquiritin shows potential as an anti-aging agent."

"Pendahuluan: Data Susenas tahun 2022 yang dikeluarkan oleh Badan Pusat Statistik (BPS) mencatat bahwa 10,48% penduduk Indonesia adalah lansia dengan 2 dari 5 lansia memiliki keluhan kesehatan. Hal ini bisa disebabkan karena banyaknya faktor ekstrinstik maupun intrinsik yang menyebabkan kerusakan seluler sehingga menginduksi protein p53 untuk melakukan cellular senescence. Senyawa thymol, carvacrol, dan flavonoid (quercetin, luteolin, apigenin, rutin, eriodictyol) dalam ekstrak tanaman Plectranthus amboinicus yang kaya akan antioksidan dipercaya dapat menghambat proses cellular senescence sehingga dapat menekan proses penuaan. Dengan itu, tujuan dari penelitian ini adalah untuk mengetahui interaksi antara komponen ligan pada senyawa aktif thymol, carvacrol, dan flavonoid yang terkandung dalam Plectranthus amboinicus dengan protein target p53 yang berkaitan dengan aktivitas cellular senescence pada proses penuaan. Metode: Studi in silico melalui molecular docking (penambatan molekuler) untuk menguji interaksi molekuler dari ligan senyawa thymol, carvacrol, dan flavonoid (quercetin, luteolin, apigenin, rutin, eriodictyol) dari tanaman Plectranthus amboinicus terhadap reseptor protein target p53 yang bertanggung jawab dalam proses cellular senescence. Hasil interaksi yang didapat merupakan sebuah histogram yang akan dianalisis dan diinterpretasikan untuk mengetahui afinitas ikatan (binding affinity) dari interaksi yang terjadi. Hasil: Terjadi interaksi antara ligan senyawa thymol, carvacrol, dan flavonoid (quercetin, luteolin, apigenin, rutin, eriodictyol) dari tanaman Plectranthus amboinicus terhadap reseptor protein target p53. Kesimpulan: Plectranthus amboinicus berpotensi menjadi agen anti-aging yang dapat menghambat dan memperlambat proses dari penuaan melalui studi in silico. Akan tetapi, perlu dilakukan penelitian lebih lanjut mengenai mengenai sifat-sifat dari Plectranthus amboinicus untuk mengetahui potensi tanaman ini terhadap tubuh manusia yang belum diketahui.

Introduction: The 2022 Susenas data stated by the Central Statistics Agency (BPS) notes that 10.48% of Indonesia’s population are elderly with 2 out of 5 having health complaints. This could be due to the many extrinsic and intrinsic factors that causes cellular damage thereby inducing the p53 protein to perform cellular senescence. Compounds of thymol, carvacrol, and flavonoids (quercetin, luteolin, apigenin, rutin, eriodictyol) from the extract of the Plectranthus amboinicus plant which are rich in antioxidants are believed to have the potential to inhibit the cellular senescence process so as to suppress the aging process. The purpose of this research is to investigate the interaction between the ligand of thymol, carvacrol, and flavonoid compounds from Plectranthus amboinicus with the p53 target protein which is associated with cellular senescence activity in the aging procress. Methods: In silico study through molecular docking to examine the molecular interactions of the ligands of thymol, carvacrol, and flavonoids (quercetin, luteolin, apigenin, rutin, eriodictyol) from the Plectranthus amboinicus plant against the target protein receptor p53 which is responsible for the process of cellular senescence. The interaction results obtained is a histogram which will be analyzed and interpreted to determine the binding affinity of the interactions that occured. Results: Interactions existed between the ligands of thymol, carvacrol, and flavonoids (quercetin, luteolin, apigenin, rutin, eriodictyol) from the Plectranthus amboinicus plant against the target protein receptor p53. Conclusion: Plectranthus amboinicus has the potential to be an anti-aging agent that can inhibit and slow down the process of aging through in silico studies. However, it is necessary to carry out further research regarding the properties of Plectranthus amboinicus to determine the unknown potential of this plant to the human body.

"Penuaan merupakan proses biologis alami yang dapat dipercepat oleh faktor eksternal, salah satunya paparan sinar ultraviolet A (UV-A). Paparan UV-A meningkatkan produksi spesies oksigen reaktif dalam sel, yang memicu stres oksidatif dan ketika berlangsung secara terus-menerus, dapat menyebabkan apoptosis. Untuk mencegah kerusakan tersebut, diperlukan agen antipenuaan yang bersifat antioksidan dan antiinflamasi seperti ekstrak Morinda citrifolia (mengkudu). Penelitian ini bertujuan untuk mengevaluasi potensi ekstrak mengkudu sebagai agen antipenuaan pada sel fibroblas L929 yang diinduksi oleh sinar UV-A. Pengujian dilakukan melalui uji fototoksiksitas terhadap sel akibat paparan UV-A, uji sitotoksiksitas ekstrak mengkudu, uji migrasi sel (uji wound scratch dan uji migrasi transwell), serta analisis ekspresi gen yang berperan dalam biogenesis mitokondria, yaitu Sirt1, Pgc1a, dan FoxO3. Hasil penelitian menunjukkan bahwa induksi UV-A pada dosis 1,5 J/cm2 berhasil menurunkan viabilitas sel secara signifikan. Sementara itu, penambahan ekstrak M. citrifolia cenderung meningkatkan viabilitas sel secara signifikan pada pemberian ekstrak dengan konsentrasi 200 μg/mL. Uji migrasi menunjukkan adanya peningkatan migrasi sel pada perlakuan yang diberikan ekstrak M. citrifolia dengan konsentrasi 100, 200, 400, dan 600 μg/mL. Hasil analisis ekspresi gen menunjukkan adanya peningkatan ekspresi gen Pgc1a dan FoxO3 pada kelompok perlakuan yang diberi ekstrak M. citrifolia. Sementara itu, ekspresi gen Sirt1 cenderung tidak mengalami perubahan yang signifikan antara kelompok yang diinduksi UV-A maupun yang diberi ekstrak. Temuan ini mengindikasikan bahwa ekstrak M. citrifolia berpotensi sebagai agen antipenuaan alami melalui modulasi ekspresi gen terkait stres oksidatif dan fungsi mitokondria.

Aging is a natural biological process that can be accelerated by exposure to ultraviolet A (UV-A) radiation. UV-A exposure increases the production of reactive oxygen species (ROS) within cells, triggering oxidative stress which, when prolonged, can lead to apoptosis. Such damage can be prevented by treatment using anti-aging agents with antioxidant and anti-inflammatory properties, such as the extract of Morinda citrifolia (noni). This study aims to evaluate the potential of noni extract as an anti-aging agent in UV-A-induced L929 fibroblast cells. The methods used include phototoxicity assay on UV-A-exposed cells, cytotoxicity assay of the noni extract, cell migration assays (wound scratch and transwell migration), and gene expression analysis of key regulators involved in mitochondrial biogenesis and protection, including Sirt1, Pgc1a, and FoxO3. The results of this study showed that UV-A induction at a dose of 1.5 J/cm² significantly reduced cell viability. In contrast, the addition of Morinda citrifolia extract tended to increase cell viability, with a statistically significant increase observed at a concentration of 200 μg/mL. Migration assays showed upregulation of cell migration in groups treated with M. citrifolia extract at concentrations of 100, 200, 400, and 600 μg/mL. Gene expression analysis revealed increased expression of Pgc1a and FoxO3 in the groups treated with M. citrifolia extract. Meanwhile, Sirt1 expression remained relatively unchanged between the UV-A–induced group and the extract-treated group. These findings suggest that M. citrifolia extract holds potential as a natural anti-aging agent through the modulation of genes involved in oxidative stress response and mitochondrial function."
