Hiperurisemia adalah kondisi tubuh ketika kadar asam urat di dalam darah yang melebihi batas normal. Peningkatan kadar asam urat di dalam darah, dapat menyebabkan inflamasi pada sendi dan kematian karena kerusakan kardiovaskuler. Inhibitor xantin oksidase bekerja dengan cara menghambat kerja pusat molybdenum pterin yang merupakan tempat aktif xantin oksidase. Xantin oksidase dibutuhkan untuk mengoksidasi hipoxantin dan xantin menjadi asam urat. Allopurinol digunakan secara luas untuk mengontrol dan menurunkan kadar asam urat di dalam darah yang penggunaannya dapat mengalami kegagalan dalam menurunkan kadar asam urat di dalam darah. Isorhamnetin merupakan senyawa yang berasal dari Propolis Sulawesi Selatan (Tetragonula biroi), yang mana menunjukan hasil aktivitas inhibisi xantin oksidase yang mendekati nilai dari senyawa allopurinol. Ekstrak etanol propolis (EEP) T. biroi yang dipartisi menggunakan sistem pelarut P Arizona memiliki nilai Rf mendekati 1 pada fasa atas dan bawahnya, tidak dapat menghasilkan senyawa Isorhamnetin dengan metode Centrifugal Partition Chromatography (CPC) berdasarkan identifikasi menggunakan metode LC-MS.
ABSTRACT
Hyperuricemia is a condition of the body that composes uric acid levels in the blood which exceed the normal limit. Increased levels of uric acid in the blood, can cause inflammation in the joints and death due to cardiovascular damage. The xanthine oxidase inhibitor works by inhibiting the work of the center of molybdenum pterin which is the active site of xanthine oxidase. Xanthine oxidase is needed to oxidize hypoxanthine and xanthine to uric acid. Allopurinol is widely used to control and reduce the level of uric acid in the blood which can cause failure to reduce uric acid levels in the blood. Isorhamnetin is a compound derived from Propolis South Sulawesi (Tetragonula biroi), which shows that the inhibitory activity of xanthine oxidase is close to the value of allopurinol compounds. Ethanol extract of propolis (EEP) T. biroi partitioned using the P Arizona solvent system has a value of Rf close to 1 in the upper and lower phases, unable to produce Isorhamnetin compounds by Centrifugal Partition Chromatography (CPC) method based on identification using the LC-MS method.
