"Latar belakang: Indonesia terletak dalam sabuk talasemia dengan prevalensi karier yang tinggi. Saat ini, terapi kelasi besi dibutuhkan oleh pasien talasemia dependen transfusi. Namun, terapi kelasi besi yang tersedia saat ini memiliki harga yang mahal serta efek samping yang berbahaya. Oleh karena itu, mangiferin merupakan salah satu senyawa alternatif yang berpotensi sebagai terapi kelasi besi karena mampu mengikat besi serta bertindak sebagai antioksidan. Studi ini bertujuan untuk menilai pengaruh mangiferin dan penggunaan nanopartikel kitosan-alginat terhadap aktivitas SOD jantung.
Tujuan: Mengetahui kemampuan mangiferin sebagai alternatif terapi besi berlebih
Metode: Sebanyak 25 tikus Sprague-Dawley dibagi ke dalam 5 kelompok perlakuan yaitu: kelompok normal (N), kelompok besi berlebih (IO), dan kelompok terapi (IO+M50, IO+MN50, IO+MN25). Iron dekstran 15mg diberikan dua kali seminggu secara intraperitoneal selama 4 minggu, sedangkan mangiferin dan mangiferin dalam nanopartikel diberikan secara intraperitoneal sesuai dosis setiap hari selama 4 minggu. Aktivitas SOD diukur pada organ jantung menggunakan kit SOD dan spektrofotometer. Analisis statistik dilakukan dengan menggunakan uji ANOVA Welch post-hoc Games-Howell dengan nilai kemaknaan 0,05.
Hasil: Studi menunjukkan bahwa tidak ada perbedaan signifikan antara aktivitas SOD jaringan jantung tikus Sprague-Dawley dalam setiap kelompok percobaan (ANOVA Welch, p=0,071). Kelompok dengan besi berlebih menunjukkan kecenderungan penurunan aktivitas SOD dibandingkan kelompok normal (rerata 0,0092 vs 0,0175 U/mg), peningkatan jika diberikan mangiferin 50mg/kgBB (rerata 0,092 vs 0,0108 U/mg), dan penurunan ketika mangiferin diberikan dalam nanopartikel kitosan-alginat (rerata 0,0108 vs 0,0093 vs 0,0054 U/mg).
Simpulan: Tidak terdapat perbedaan signifikan pada aktivitas SOD jantung tikus Sprague-Dawley antara setiap kelompok, baik dari pemberian besi berlebih maupun dengan terapi mangiferin.

Introduction: Indonesia is located inside the thalassemia belt with high prevalence. Until now, transfusion dependent thalassemia needs iron chelator therapy. But currently, the iron chelator therapies available are so expensive and have side effects. Currently, mangiferin is extensively studied because of its ability to act as an iron chelator and also as antioxidant. This study aims to evaluate the effect of mangiferin and mangiferin in chitosan-alginate nanoparticle to SOD activity in heart with iron overload condition.
Purpose: To know the extent of mangiferin potential as iron chelator therapy alternative
Methods: Twenty-five Sprague-Dawley rats were divided into 5 groups of 5 each: normal (N), iron overload (IO), 50 mg/kg/day mangiferin therapy (IO+M50), 50 mg/kg/day mangiferin therapy in nanoparticle delivery (IO+M50), and 25 mg/kg/day mangiferin therapy in nanoparticle delivery (IO+MN25). Iron dextran and mangiferin were administered intraperitoneally according to each group dose for 4 weeks. SOD activity was measured with spectrophotometer and analysed with Welch’s ANOVA followed by Games-Howell post-hoc test with significance level of 0.05.
Results: This study demonstrated no significant difference of SOD activity compared in each group (Welch’s ANOVA, p=0.071). SOD activity tend to be higher in N group when compared to IO group (mean 0.01175 vs 0.0092 U/mg), tend to increase with mangiferin therapy (mean 0.0108 U/mg), but tend to be lower when carried by chitosan-alginate nanoparticle with IO+MN25 being the lowest (mean 0.0108 vs 0.0093 vs 0.0054 U/mg).
Conclusion: There is no significant difference of SOD activity from comparison of each group of Sprague-Dawley rat’s hearts.
