"Protein sel merupakan makromolekul yang terdiri dari satu atau beberapa polipeptida yang tersusun dari rangkaian asam amino yang saling berikatan. Terdapat perbedaan profil protein antara sel normal dengan sel kanker.
Tujuan : Melihat ekspresi protein pada KSSRM dan mukosa mulut normal.
Metode : Sel galur HSC-3 dan HSC-4 dikultur hingga confluent. Sel skuamosa mukosa normal diambil dari jaringan gingiva pasien odontektomi. Semua sampel dilakukan prosedur ekstraksi protein, Bradford protein assay, dan SDS PAGE.
Hasil : KSSRM mengekspresikan protein dengan level cukup tinggi pada berat molekul 31-78 kDa. Namun, pada mukosa normal, kebanyakan mengekspresikan protein pada berat molekul antara 39 - 172 kDa.
Kesimpulan : Terdapat perbedaan ekspresi protein pada sel galur KSSRM dibandingkan dengan mukosa mulut normal.

Cells Proteins are macro molecules consist of one or several polypeptides which formed from amine acid chain that bound one another. There is a different of protein profile between normal and cancer cells.
Objective : To observe the protein expression in OSCC and normal oral mucous.
Method : Cell lines HSC-3 and HSC-4 were cultured until confluent. Normal squamous mucosa was taken from gingival tissues patient who had odontectomy procedure. Protein extraction, Bradford protein assay, and SDS PAGE procedure were performed for all samples.
Results : Oral squamous cells carcinoma expressed rather high level of protein which have molecular weight of 31-78 kDa compared to normal gingival which express protein molecular weight ranging between 39 - 172 kDa.
Conclusion : There are different protein expression between oral squamous cells carcinoma and normal oral mucous."

"Latar Belakang: p73, homolog p53, diketahui memiliki kemampuan serupa dalam menekan pertumbuhan tumor. Protein p73 diekspresikan dalam berbagai level pada sel kanker dan jaringan normal yang berbeda. Belum diketahui bagaimana pola ekspresi protein p73 pada KSSRM dan pada jaringan mukosa mulut normal.
Tujuan: Mengetahui profil protein p73 pada KSSRM tipe mutant p53 dan jaringan mukosa mulut normal berdasarkan berat molekul protein.
Metode: Ekstrak protein dari HSC-3 dan HSC-4 serta jaringan mukosa normal dianalisa dengan teknik SDS PAGE untuk mendeteksi protein p73 berdasarkan
berat molekulnya.
Hasil:. pita protein p73 pada HSC-3 lebih tebal daripada HSC- 4. Terdapat variasi profil protein p73 pada mukosa mulut normal dengan pita protein tebal (8/17) dan sedang (5/17).
Simpulan: Terdapat perbedaan profil protein p73 antara HSC-3 dan HSC-4 berkaitan dengan tingkat protein p53 dan SNP pada kodon 72. Kebanyakan sampel jaringan mukosa memperlihatkan ketebalan pita protein p73 yang cukup tinggi.

Backround: p73, the homolog of p53, has a similar ability in tumor suppression. p73 protein expressed at a different level in various cancer cells and normal tissues. Profile of p73 protein in mutant p53 OSCC cell line and normal human oral mucosa have not been known.
Objectives: To observe p73 protein profile in mutant p53 OSCC cell lines and normal human oral mucosa.
Methods: The extracted protein of HSC-3 and HSC-4 cell lines and normal mucosal tissues were analyzed with SDS PAGE to detect p73 protein based on the molecular weight.
Results:. The band of p73 protein in HSC-3 shows a higher density compared to its density of HSC-4. A thick p73 protein band was shown on 8/17 of normal mucosal tissues while medium level of p73 protein band was shown on 5/17.
Conclusion: The protein profile between HSC-3 and HSC-4 were different related with p53 protein and SNP on codon 72 of each samples. Most of mucosal tissues shows a quite high density of p73 protein bands.

"ABSTRAK Karsinoma sel skuamosa kepala leher (KSSKL) menempati peringkat ke-6  kanker tersering di seluruh dunia.  Perkembangan  dalam  hal  terapi yang menggunakan  beragam modalitas untuk mengontrol  lokal dan  regional masih belum dapat meningkatkan angka survival lima tahun yang hanya sekitar 50% selama tiga dekade terakhir.  Metastasis jauh  menjadi penyebab utama  morbiditas dan  mortalitas pada KSSKL.  Sebagian besar pasien dengan metastasis jauh tidak terdeteksi secara klinis dan  radiologi.  Hingga saat ini sistem staging dan radiologi konvensional tidak mampu mendeteksi metastasis sejak dini . Mikrometastasis  merupakan proses awal dari suatu makrometastasis, sehingga sel tumor yang bersirkulasi dianggap sebagai silent predictor. Deteksi  CTC dianggap sebagai metode yang inovatif, sensitif, dan spesifik untuk dapat memprediksi kemungkinan terjadinya metastasis pada pasien-pasien  KSSKL. Penelitian ini menggunakan 90 sampel darah pasien penderita KSSKL stadium I-IV yang berobat di poli Bedah Onkologi dan THT RSUPNCM  pada periode Januari hingga Desember 2018.  Sampel darah  sebanyak 8 ml dikirim ke Laboratorium Patologi Klinik FKUI/RSUPNCM untuk dianalisa jumlah CTC -nya dengan  menggunakan flow cytometry, penanda epitel EpCAM dan CK19 serta penanda lekosit CD45. Analisa statistik dilakukan untuk mengetahui ada tidaknya asosiasi antara status klinikohistopatologi pasien KSSKL dengan deteksi CTC. Flow cytometry mampu  mendeteksi CTC positif sebanyak 28 dari 90 sampel darah (31.1%). Dari analisa bivariat dan multivariat, tidak didapatkan adanya asosiasi yang bermakna antara status klinikohistopatologi pasien KSSKL dengan deteksi CTC (p>0.05).    Pasien dengan stadium lanjut  memiliki kecenderungan 4.192 kali untuk mendapatkan CTC positif dibandingkan dengan stadium awal. Begitu pula pasien  yang memiliki sublokasi  tumor di daerah  faring  dan  laring   memiliki  kecenderungan 2.634 kali  untuk mendapatkan CTC positif bila dibandingkan dengan sublokasi anatomi bibir, rongga mulut, hidung dan sinus paranasal. Flow cytometry mampu mendeteksi CTC pada KSSKL dengan presentase 31.1%  bila menggunakan dua penanda epitel. Tidak terdapat asosiasi yang bermakna antara status klinikohistopatologi pasien KSSKL dengan deteksi CTC. Diperlukan studi lanjutan untuk meningkatkan sensitivitas dan spesifisitas serta mengkonfirmasi dapat tidaknya CTC dipakai sebagai prediktor dan prognostikator di bidang KSSKL.
ABSTRACT
HNSCC is the sixth most common cancer worldwide. The therapy of choice for newly diagnosed patients consists of combined modality treatment for locoregional control. Despite of improvement in treatment, for all stages combined, the 5-year survival rate is approximately 50% and this rate has not changed significantly in the last 3 decades. As distant metastases develop, the chance for cure becomes low and survival decreases. Tumor metastasis is an important contributor to mortality in cancer patients. Mostly distant metastases are not seen by clinical examination and/ or conventional imaging. To date, TNM staging  system and  the existing image technology are not sufficient to detect tumor metastasis in early stages. The concept of the development of metastasis involves a tumor cell that dissociates from the primary tumor (micrometastasis) and circulates within lymphatic or vascular channels. The true utility of a circulating tumor cell (CTC) is that it acts as a silent predictor of metastatic disease. Detection of CTCs serves as an innovative, sensitive and specific marker and providing early and definitive evidence of metastatic disease. In this study, peripheral blood samples from 90 patients with SCCHN stage I-IV from Surgical Oncology and ENT OPD were taken. CTCs were quantified using flow cytometry of anti-EpCAM, CK19 and CD45. Their detection was correlated with clinicohistopathologic characteristics. CTCs were identified in 28 (31.1%) patients at any time point with a mean + standard deviation of  0.9 + 3.2 CTCs. In bivariate and  multivariate analysis, we observed no significant correlation between the presence of CTCs and clinicohistopathologic characteristics (p>0.05). However the odds for the patients with advanced stages to have positive CTCs  is 4.192 times higher than early stage. Moreover, patients with pharynx and larynx cancer have the odds 2.634 times higher than those with lips, oral cavity and paranasal sinuses cancer. The rate of CTCs detection in HNSCC by flow cytometry was 31.1%. Detection of CTCs does not correlate with any clinicohistopathological characteristics. Further studies is needed to increase the sensitivity and specificity  and  also to confirm the potential of CTC to serve as a predictor and prognosticator in patients with HNSCC.

"Tongue squamous cell carcinoma, a relatively requent malignancy in the oral cavity has a great tendency to metastasize to regional lymph nodes that worsens the prognosis of the disease. The anti metastasis gene NM23, encodes a 17 KD cytoplasmic and nuclear proteins has shown controversy behaviour as metastatic suppressor in human cancer. An observational study using immunohistochemical staming method with LSAB (-) technique was conducted on 41 paraffin block specimens to show the relationship between significance of the expression of NM23 protein as a marker for cancer metastasis and the staging, cancer grading and the positive regional head and neck lymph nodes. Statistical significance of difference was evaluated using Chi square test (p<0.01). The result showed that there was an inverse relationship between expression of NM23 protein and the cancer grading as well as frequency of the positive regional head and neck lymph nodes supporting the role of NM23 as metastasis suppresor factor that may be useful for predicting tongue squamous cell carcinoma metastasis."
