Abstrak :
Minuman kefir merupakan suatu produk fermentasi yang dapat dibuat secara mudah dan murah. Minuman kefir dikenal luas sebagai suatu minuman probiotik. Pembuatan kefir dapat dilakukan dengan menggunakan baik susu sapi maupun susu kambing. Penelitian ini bertujuan untuk menguji aktivitas antibakteri dari kefir susu sapi dan susu kambing, serta mengisolasi bakteri lactobacilli yang berperan. Aktivitas antibakteri dari kefir diuji berdasarkan perbedaan pada jenis susu yang digunakan dan lama waktu fermentasi. Isolasi dan karakterisasi isolat dilakukan berdasarkan Cowan and Steel’s Manual for the Identification of Medical Bacteria. Kefir dibuat dengan menginokulasikan 5% (w/v) granula kefir lokal ke dalam 50 mL susu sapi atau kambing yang telah dipasteurisasi. Fermentasi dilakukan selama 3, 4, dan 5 hari untuk kedua jenis susu. Uji antibakteri dari kefir dilakukan dengan metode difusi menggunakan silinder (cylinder diffusion method) terhadap 5 bakteri uji, yaitu Staphylococcus aureus NBRC 100910, Pseudomonas aeruginosa DRK 9.1, Eschericia coli NBRC 3301, Bacillus subtilis NBRC 13719 dan Kocuria rhizophila NBRC 12708. Pengukuran nilai pH kefir dilakukan dengan pH meter dan nilai total asam kefir dengan metode titrasi. Hasil uji aktivitas antibakteri dari kefir susu sapi maupun susu kambing menunjukkan adanya aktivitas antibakteri terhadap kelima bakteri uji. Secara umum kefir susu sapi menunjukkan aktivitas antibakteri yang lebih kecil dari kefir susu kambing, baik dari hasil fermentasi dengan lama waktu 3, 4, maupun 5 hari. Selanjutnya, aktivitas antibakteri yang paling optimal secara umum diperoleh pada kefir dengan lama fermentasi 4 hari baik untuk kefir susu sapi maupun susu kambing. Sebanyak 9 isolat bakteri berhasil diisolasi. Seluruhnya menunjukkan karakteristik bakteri yang berasal dari kelompok lactobacilli. ......Kefir is a fermented beverage that can be made easily and cheaply. Kefir is widely known as a probiotic beverage. The production of kefir can be done using either cow milk or goat milk. This study aims to examine the antibacterial activity of cow milk and goat milk kefir, as well as to isolate responsible lactobacilli bacteria. The antibacterial activity of kefir is examined based on differences in type of milk used and fermentation time. The isolation and characterization of isolates is done according to Cowan and Steel’s Manual for the Identification of Medical Bacteria. The kefirs are made by inoculating 5% (w/v) local kefir grains into 50 mL pasteurized cow milk or goat milk. Fermentation was carried out for 3, 4, and 5 days for both types of milk. The antibacterial test of kefirs was carried out using diffusion method utilizing cylinders (cylinder diffusion method) against 5 test bacteria, namely Staphylococcus aureus NBRC 100910, Pseudomonas aeruginosa DRK 9.1, Eschericia coli NBRC 3301, Bacillus subtilis NBRC 13719 and Kocuria rhizophila NBRC 12708. The measurement of kefir pH values was performed using pH meter and kefir total acidities by using titration method. Antibacterial activity test results from either cow milk or goat milk kefir showed the presence of antibacterial activity against five test bacteria. In general, cow milk kefir showed lower antibacterial activity than goat milk kefir in fermentation times of either 3, 4, or 5 days. Furthermore, the most optimal antibacterial activity was generally obtained in kefirs with a fermentation time of 4 days for both cow milk and goat milk kefir. A total of 9 bacterial isolates were successfully isolated. All of which shows the characteristics of bacteria from the lactobacilli group.

Penelitian ini dilakukan untuk mencari beberapa senyawa kimia dari ekstrak kulit pohon Garcinia tetrandra serta uji aktivitas biologi terhadap Artemia salina L. dan aktivitas antibakteri. Senyawa tersebut diisolasi dengan cara ektraksi dalam n-heksan selanjutnya ekstraksi dengan aseton . Masing-masing ekstrak dipisahkan dengan cara kromatografi kolom dengan fasa diam silika gel dan fasa geraknya adalah n-heksan , etil asetat, metanol secara gradien. Senyawa kimia yang telah murni ditentukan struktur molekulnya dengan cara spektrofotometri UV-Vis, spektrofotometri Infra Merah, Spektrometri Massa, Spektrometri Resonansi Magnet inti 3H dan 13 C. Dari hasil penelitian ini diperoleh senyawa GT-1 yang mempunyai rumus molekul C23HxoO6 , diperkirakan sama dengan Thwaitesixanton, senyawa GT-2 dengan rumus molekul C30H500 yang diperkirakan senyawa 3-a- (22) (29) Hopen-ol; Senyawa GT-3 dengan rumus molekul C38H50O6 yang diperkirakan sama dengan Camboginol; Senyawa GT-4 dengan rumus molekul C38H5006 yang diperkirakan sama dengan cambogin. Dari senyawa - senyawa tersebut yang memiliki aktivitas antimikroba adalah senyawa GT-3 dengan diameter harnbatan rata-rata = 8 mm/1000 ppm.terhadap bakteri B. subtilis ATCC 6633, E. coli ATCC 25922, S. aureus ATCC 6538, M. phlei DSM 43286. Dari hasil uji aktivitas terhadap larva-udang Artemia salina Leach., senyawa yang memiliki aktivitas yang cukup signifikan adalah senyawa GT-3 dan GT-4 dengan LC50 masing-masing 7µg/ ml dan 18µg/ ml. ......Isolation, Structure Elucidation, Biological Activity Test of Chemical Constituents of Stem Bark of Garcinia Tetrandra PierreThis study was caned out to obtain several chemical constituents of the extract of the stem bark of Garcinia tetrandra Pierre, their biologic are activity to Artemia salina Leach and their antibacterial activity. The compounds were isolated from the n-hexane and acetone - soluble extracts. Each extract was separated through a column chromatography on silica gel G , as the stationary phase which gradient mixtures, of n-hexane, ethyl-acetate and methanol as the mobile phase. The purified chemical constituents were identified their structures based on their UV -- Visible , Infra Red Spectrophotometry , Mass Spectrometry, 1H and '3C - Nuclear Magnetic Resonance Spectrometry data. The first isolate of GT-I with its molecular formula of C23HxoO6 was identified as Thawaitesixantone; GT-2 with and its molecular ion formula of C30H50O was identified as 3-a-(22)(29) Hopen-ol; GT-3 with its molecular formula of C38H50O6 was identified as camboginol; GT- 4 with its molecular formula of C38H5006 was identified as cambogin. From of all of the isolated compounds GT-3, showed antibacterial activity. In a concentration of 1000 ppm, its inhibition against the growth off B. subtilis ATCC 6633, E. code ATCC 25922, S. aureus ATCC 6538, M phlei DSM 43286 were 8 mm. From the results of toxicity evaluation to the Artemia salina Leach , GT-3 and GT-4 with its LC50 7µg/ ml and 18 µg/ ml, respectively.

Abstrak :

Latar Belakang: Peningkatan jumlah bakteri Salmonella Typhi dengan resistensi terhadap antibiotik lini pertama menjadi beban ganda bagi negara berkembang di Asia termasuk Indonesia sehingga diperlukan antibiotik alternatif untuk menghadapinya. Daun Carica papaya memiliki komponen-komponen antibiotik sehingga berpotensi menjadi antibiotik alternatif.

Metode: Pada penelitian ini dilakukan uji eksperimental dengan ekstraksi daun C.papaya dengan pengenceran menjadi 4 konsentrasi; 100 mg/ml, 75 mg/ml, 50 mg/ml, dan 25 mg/ml. Tiap konsentrasi diuji aktivitas antibakteri terhadap Salmonella Typhi dengan metode difusi cakram dan uji konfirmasi dengan metode broth dilution. Diameter zona inhibisi pertumbuhan bakteri dengan metode difusi cakram diukur kemudian dianalisis

Hasil: Hasil dari penelitian difusi cakram tidak membuktikan adanya aktivitas antibiotik dari ekstrak daun C.papaya terhadap Salmonella Typhi sementara uji konfirmasi dengan metode broth dilution membuktikan adanya aktivitas antibakteri oleh keempat konsentrasi ekstrak terhadap Salmonella Typhi.

Kesimpulan: Kesimpulan dari penelitian ini adalah ekstrak daun C.papaya memiliki aktivitas antibakteri terhadap Salmonella Typhi


Background: Increasing incidence of resistant Salmonella Typhi strain towards first-line antibiotics has become a high burden in Asia’s developing countries including Indonesia impacting the need of an alternate antibiotic. Carica papaya leaves extract contain antibiotic components making it a potential alternative antibiotic.

Methods: This experimental research uses extraction of the C.papaya leaves with dilution into 4 concentrations; 100 mg/ml, 75 mg/ml, 50 mg/ml, and 25 mg/ml. Each concentration undergo antimicrobial susceptibility testing towards Salmonella Typhi with disc diffusion method and confirmatory test with broth dilution method. Diameter of inhibition zone of Salmonella Typhi bacterial growth in disc diffusion method will be measured and analyzed.

Results: Results show that the disc diffusion method was not able to prove the antibacterial activity of C.papaya leaves extract against Salmonella Typhi while the confirmatory test with broth dilution method has successfully proven antibacterial activity of the four extract concentrations towards Salmonella Typhi.

Conclusion: This research concluded that Carica papaya leaves extract has antibacterial activity towards Salmonella Typhi.

Penyakit infeksi masih menjadi masalah kesehatan di Indonesia, dimana salah satu bakteri penyebabnya adalah Streptococcus pyogenes. Bakteri ini dapat menyebabkan penyakit-penyakit penting mulai dari infeksi kulit hingga penyakit yang dapat membahayakan nyawa seperti glomerulonephritis. Hingga saat ini, penyembuhan untuk bakteri Streptococcus pyogenes masih bergantung dengan antibiotik jenis penicillin maupun ciprofloxacin. Penelitian ini bertujuan untuk mengetahui apakah ekstrak biji pepaya (Carica papaya L) memiliki aktivitas antibakteri terhadap bakteri Streptococcus pyogenes dengan melihat konsentrasi hambat minimum (KHM) dan konsentrasi bunuh minimum (KBM). Penelitian ini dilakukan menggunakan uji in-vitro dengan cara mikrodilusi tabung. Ekstrak biji pepaya digunakan dengan variasi konsentrasi 16.5%, 11%, 8.25%, dan 5.5%. KHM ekstrak biji pepaya ditemukan pada konsentrasi 16.5% ditandai dengan larutan yang bening pada tabung dengan konsentrasi ekstrak sebesar 16.5%. Dilain pihak, KBM ekstrak biji pepaya ditemukan pada konsentrasi 5.5%, yang ditandai dengan tidak adanya pertumbuhan koloni bakteri pada agar darah. Dengan demikian, dapat disimpulkan bahwa ekstrak biji pepaya berpotensi sebagai agen antibakteri untuk melawan bakteri Streptococcus pyogenes ......Nowadays, infection is still a major problem in Indonesian health management. Streptococcus pyogenes is an example of a bacteria that needs more attention since it can cause a mild infection on skin untill a deadly infection such as glomerulonephritis. In Indonesia, treatment for Streptococcus pyogenes infection is still heavily dependent on the use of penicillin or ciprofloxacin. This reasearch’s objective is to discover if papaya’s seed (Carica papaya L) has an antibacterial activity for Streptococcus pyogenes by measuring the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC). This reasearch was done by in vitro test using a microdilution tube. Papaya’s seed extracted in varied concentration which is 16.5%, 11%, 8.25%, and 5.5%. The results showed that Minimum Inhibition Concentration (MIC) of papaya’s seed extract concentration is 16.5% shown by a clean solution in tube. On the other hand, Minimum Bactericidal Concentration (MBC) of papaya’s seed extract is 5.5% with no colony growth found in the blood agar specimen. In conclusion, papaya’s seed extract has a good potential to be an antibacterial to treat Sptretococcus pyogenes.

The volatile chemicals from species of wild Cinnamomum spp. (C. racemosum, C. cuspidatum, C. politum, C. javanicum), Etlingera spp. (E. pyramidosphaera, E. megalocheilos, E. coccinea, E. elatior) and Schizostachyum spp. (S. blumei, S. brachycladum, S. lima, S. pilosum) found in Sabah were investigated. The oils were obtained from the bark, rhizome and culm of respective specimens by hydrodistillation and the profile of volatile chemicals was obtained using Gas Chromatography- Mass Spectrometry (GCMS). Dominance of eucalyptol, terpinen-4-ol and eugenol were consistent among the species from genus Cinnamomum. aromadendrane oxide, lauryl aldehyde, elemicin, borneol and 1-dodecanol were predominant among the species from genus Etlingera. α-elemol, coumaran, guiacol-4-vinyl, palmitic acid and phytol acetate predominate the species from genus Schizostachyum. Strong inhibition against Staphylococcus aureus (MIC: 5.62 ± 0.5 μg mL-1) were exhibited by essential oils of C. cuspidatum and E. coccinea, oil of S. blumei inhibited Listeria monocytogenes (MIC: 4.60 ± 0.5 μg mL-1), oil of C. javanicum inhibited Salmonella typhimurium (MIC: 5.50 ± 0.5 μg mL-1). meanwhile the oil of C. politum suppressed Salmonella enteritidis (MIC: 5.20 ± 0.5 μg mL-1) was measured using microdilution method. these findings reveal the potential of selected plants used by indigenous communities of Borneo as antimicrobials in food, cosmetics and pharmaceutical industries.

Abstrak :
Copper(II) and nickel(II) complexes of 14-membered hexaazamacrocyclic ligand with C-methyl substituent have been synthesized by one-pot reaction and characterized by elemental analyses, FT-IR, electronic spectra, and LC-MS. Complex types are [Cu(C18H42N6)(ClO4)2] (1), [Ni(C18H42N6)](ClO4)2 (2), and [Cu(C22H50N6)(ClO4)2] (3). The results of spectroscopic techniques indicated that both complexes 1 and 3 containing copper(II) are octahedral geometry, whereas the complex 2 is a square-planar geometry in which the metal center coordinates to the four nitrogen atoms of macrocyclic ligand in a square planar fashion.All complexes were also screened for their antibacterial activity against on two bacteria strains. These results show that copper(II) and nickel(II) complexes were tested for antibacterial activity against gram positive, Staphylococcus aureus which was more active than the against gram negative, bacteria Escherichia coli.The results showed a good antibacterial activity of all complexes which suggests their potential applications as antibacterial agents.

Infeksi MRSA belum dapat ditangani secara efektif. Pilihan terapi yang saat ini digunakan adalah vankomisin, clindamycin, atau trimethoprim-sulfomethoxazole TMP-SMX . Penelitian ini dilakukan untuk mengetahui aktivitas antimikroba ekstrak kulit batang Aleurites moluccana L. willd terhadap MRSA dengan harapan dapat dijadikan pengobatan alternatif untuk infeksi MRSA. Ekstrak kulit batang A. moluccana diketahui memiliki zat aktif 3-acetyl aleuritolic acid yang memiliki aktivitas antibakteri terhadap Staphylococcus aureus. Ekstrak kulit batang A. moluccana dilarutkan dengan methanol, kemudian diencerkan dengan konsentrasi 50 g/mL, 100 g/mL, 200 g/mL, 400 g/mL, dan 800 g/mL. Pengujian dilakukan dengan metode difusi cakram, kemudian dibandingkan dengan clindamycin 20 g/mL sebagai kontrol positif dan akuades sebagai kontrol negatif. Hasil penelitian menunjukkan tidak terbentuk zona hambat pada kelima konsentrasi ekstrak yang diuji. Hal ini mungkin dipengaruhi oleh konsentrasi ekstrak yang diuji serta karakteristik tanaman A. moluccana yang tempat tumbuhnya berbeda antara daerah yang satu dengan yang lainnya sehingga mempengaruhi kandungan zat aktif dalam tanaman tersebut. ......MRSA infection cannot be treated effectively. Treatments being used now are vancomycin, clindamycin, or trimethoprim sulfomethoxazole TMP SMX . This research was conducted to know the antibacterial activity of Aleurites moluccana L. willd stem bark extract against MRSA so it can be used as an alternative treatment for MRSA infection. A. moluccana stem bark extract is known to have 3 acetyl that showed antibacterial activity against Staphylococcus aureus. The extraction of A. moluccana stem bark used methanol as solvent, and then diluted to five different concentration, 50 g mL, 100 g mL, 200 g mL, 400 g mL, dan 800 g mL. The research was conducted with disc diffusion method, and then compared to clindamycin 20 g mL as positive control and aquadest as negative control. The result showed no inhibition zone for all concentration that were tested. This result could be affected by several factors, such as the extract concentration and the different characteristic of the plant according to the plant rsquo s habitat.

Bakteri asam laktat merupakan kelompok bakteri yang sering ditemukan pada makanan fermentasi. Bakteri asam laktat seperti Lactobacillus diketahui memiliki aktivitas antibakteri terhadap bakteri pathogen dan dapat berperan sebagai agen probiotik. Penelitian yang telah dilakukan sebelumnya oleh Wang (2022) dan Andika (2022) menunjukan adanya aktivitas antibakteri dari filtrat tape ketan hitam dan kefir serta berhasil mengisolasi 13 isolat bakteri asam laktat. Tujuan penelitian ini adalah untuk melakukan penapisan aktivitas koagulasi susu serta aktivitas antibakteri dari masing-masing isolat bakteri asam laktat yang telah berhasil diisolasi dari tape ketan hitam dan kefir. Dari penapisan aktivitias antibakteri tersebut kemudian dipilih isolat terpilih yang kemudian dilakukan uji antibiosis. Sebanyak 13 isolat bakteri asam laktat telah berhasil dilakukan penapisan aktivitas kogulasi susu dan aktivitas antibakteri. Semua isolat bakteri asam laktat menunjukkan dapat mengkoagulasi susu. Kemudian berdasarkan penapisan aktivitas antibakteri didapatkan 3 isolat terpilih dengan kode isolat TM2, KNB2, dan KNB4 dengan nilai Indeks Aktivitas (IA) zona bening tertinggi disetiap perlakuan bakteri uji. Ketiga isolat terpilih tersebut kemudian dilakukan uji antibiosis. Hasil uji antibiosis dari filtrat fermentasi isolat terpilih (TM2, KNB2, dan KNB4) dengan menggunakan medium standar de Man Rogosa Sharpe Broth (MRSB) menunjukkan terdapat aktivitas antibiosis terhadap semua bakteri uji dan ketiga isolat tersebut berpotensi sebagai agen probiotik. ......Lactic acid bacteria are a group of bacteria that are often found in fermented foods. Lactic acid bacteria such as Lactobacillus are known to have antibacterial activity against pathogenic bacteria and can act as probiotic agents. Previous research by Wang (2022) and Andika (2022) showed that there was antibacterial activity from fermented black glutinous rice and kefir filtrate and succeeded in isolating 13 isolates of lactic acid bacteria. The purpose of this study was to screen the coagulation activity of milk and the antibacterial activity of each lactic acid bacteria isolated from fermented black glutinous rice and kefir. From the screening of antibacterial activity, selected isolates then subjected to an antibiosis test. A total of 13 isolates of lactic acid bacteria has been successfully screened for milk coagulation activity and antibacterial activity. All isolates of lactic acid bacteria showed the ability to coagulate milk. Then based on the antibacterial activity screening, 3 selected isolates were selected with the isolate codes TM2, KNB2, and KNB4 with the highest clear zone Activity Index (IA) value in each treatment of the test bacteria. The three selected isolates were then subjected to an antibiosis test. Antibiosis test results from the fermented filtrate of selected isolates (TM2, KNB2, and KNB4) using standard de Man Rogosa Sharpe Broth (MRSB) medium showed that there was antibiosis activity against all tested bacteria and the three isolates had the potential as probiotic agents.

Aktivitas bakteri merupakan salah satu penyebab jerawat. Rimpang kencur (Kaempferia galanga L.) memiliki aktivitas antibakteri yang berasal dari senyawa etil p-metoksi sinamat (EPMS). Tujuan penelitian ini adalah untuk mengetahui aktivitas antibakteri dari EPMS ekstrak rimpang kencur terhadap P.acne, S.aureus dan S.epidermidis serta stabilitas fisik krim dan keamanan krim anti jerawat. Kristal EPMS diperoleh melalui proses maserasi dengan pelarut heksan, lalu pemurnian dan identifikasi karakteristik kristal EPMS. Uji aktivitas antibakteri dan konsentrasi hambat minimum (KHM) pada konsentrasi EPMS 0,3; 0,6; 1,2 dan 2,4% dengan metode difusi cakram dan dilusi cair. Hasil menunjukkan semua konsentrasi EPMS memiliki aktivitas antibakteri secara signifikan (p<0,01) dengan zona jernih secara berturut-turut terhadap P.acne (9,00; 11,50; 14,50; 16,00 mm), S.aureus (9,00; 11,50; 16,50; 22,00 mm) dan S.epidermidis (10,50; 12,50; 20,50; 27,00 mm). Senyawa EPMS dengan konsentrasi 0,6; 1,2 dan 2,4 % terbukti memiliki konsentrasi hambat minimum (KHM) terhadap bakteri P.acne, sedangkan pada S.aureus dan S.epidermidis pada konsentrasi 1,2 dan 2,4%.Berdasarkan hasil evaluasi stabilitas fisik, krim EPMS 1,2% memiliki stabilitas fisik baik hingga akhir penelitian. Dari hasil uji keamanan (pacth test) pada 12 subjek tidak terjadi iritasi alergi, sehingga krim EPMS 1,2% aman digunakan dalam sediaan topikal. ...... An activity of certain bacterias is one of the causes of acne. Rimpang kencur (Kaempferiagalanga L.) has an antibacterial agent from compound ethyl pmethoxy cinnamate (EPMC). The purpose of this research is to find out the activities of antibacterias using EPMC rimpang kencur extracts against P.acne, S.aureus and S.epidermidis with the physical stability of the cream and its safety use as an anti acne cream. The EPMC crystals are obtained through the process of maceration using hexane solvent, purification and identification of the characteristic of EPMC crystals. The activity test of the antibacterial and the minimum inhibitory concentration (MIC) of EPMC are 0,3; 0,6; 1,2 and 2,4% was done using disk diffusion method and broth dilution test. The result shows that all EPMC concentration has significant antibanterial activity (p<0,01) respectively gaining clear zone against P.acne (9,00; 11,50; 14,50; 16,00 mm), S.aureus (9,00; 11,50; 16,50; 22,00 mm) and S.epidermidis (10,50; 12,50; 20,50; 27,00 mm). EPMC compound with the 0,6; 1,2 dan 2,4 % concentration is proven to have minimum inhibitory concentration (MIC) against P.acne bacterias, while on the S.aureus dan S.epidermidis reaches up to 1,2 dan 2,4% concentration. Based on the results of the evaluation on the physical stability, EPMC 1,2% cream has a good physical stability until the final research. From the results of safety use (pacth test) on 12 subjects there were no evidence of allergic irritation, therefore cream EPMC 1,2% is safe to be used in topical preparation.
