Today, Indonesia has the largest palm oil plantation in the world. Palm area in Indonesia, in 2008, according Ditjenbun, Department of Agriculture, estimated to reach 7 million ha (Ditjenbun Deptan 2009). Palm fruit produced by palm oil trees collected in bunches, therefore, often called as Fresh Fruit Bunches (FFB). FFBs are then transported to the factory for further process to produce palm oil. The main product of oil mill is Crude Palm Oil (CPO) and Palm Kernel Oil (PKO). In 2008, about 90,5 ton FFB yielded 18,1 ton of CPO from the fruit (mesocarp) and 2,1 ton of PKO from the nut (palm kernel) (Naibaho 1998; Ditjenbun Deptan 2009; PPKS 2010). When the nut is processed, it yields PKO, Palm Kernel Meal (PKM), and shell. The PKM is the solid part remaining from palm kernel after the extraction of PKO by a mechanical screw pressing. From 90,5 ton of processed FFB can produce about 2,5 ton of PKM. The number of this by-product always be increased due to the growth of the oil palm industry in Indonesia. Institut de Recherche pour le Développement (IRD) has done researches about the application of palm oil industry waste (PKM) as fish feed through bioconversion process using maggot. Maggot is Hermetia illucens L. larvae that has the ability to convert organic material (PKM) into it's biomass. Maggot biomass has then been used to feed fish (Hem et al 2008b). This bioconversion process of PKM using maggot produced waste, which is called Bioconversion Fertilizer Palm Kernel Meal (BFPKM). The aim of this study is to know the effect of BFPKM, as organic fertilizer, to Vigna unguiculata L. Walp (yardlong bean) var. mutiara. The study was held in Balai Riset Ikan Hias and IRD laboratorium Depok, on July until October 2008. This research is an experimental study using a randomized complete design. The parameters observed including the growth of stems, leaves, flowers, pods and roots, between the plants cultivated with BFPKM and without BFPKM. The result shows that BFPKM as organic fertilizer tend to gave a positive effect on the growth of stem, leaf, flower and pod. BFPKM also seem to stimulate the formulation of new branches and make earlier formation of flower. Inside underground, BFPKM tend to show indication to stimulate the growth of the root faster and more branching. Well root development could give more nutritions supply to whole of plant. It was showed by this study. BFPKM supplies nitrogen, phospor, and potassium for plant's growth. That nutriens are needed by plant to grow. So, this study shows that BFPKM can be applied as organic fertilizer that seem to give effect in the growth of yardlong bean.

Chromium sulphate Cr(III) and pentachlorophenol (PCP) widely used as tanning and biocide respectively in leather preparation are highly toxic and recalcitrant. Biosorption of chromium by Aspergillus niger FIST1 was evaluated, and process parameters were optimized in presence of carbon, nitrogen, carbon:nitrogen, pH, temperature, different concentration of chromium. The potency of Acinetobacter sp.IST3 for degradation of pentachlorophenol was determined by HPLC after formation of tetrachlrohydroquinone and chlrorohydroquinone. Bioremediation of chromium and PCP were tested in bioreactors in sequential way where bacterium treated effluent subsequently treated by fungus showed reduction of chromium (82%) and PCP (85%) after 120 hrs. Biosorption of chromium was determined by transmission electron microscopy (TEM), scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDX). Recovery of chromium in tannery effluent was initially obtained by CaO:MgO (2:1) and pH adjusted to 7.0-7.6, and chromium absorbed by fungus and bacteria, was further used for tanning of the leather. Results of the study indicated that quality of the leather prepared by absorbed chromium of fungus and bacteria was better than chromium recovered by CaO:MgO determined by SEM. In view of above results ‘tanning cake’ was prepared which is better substitute of raw chromium used for tanning in leather mills.

Palm Kernel Meal is solid waste from Palm Oil extraction (Ng, 2003). Akubuo & Eje (2002) reported that mechanical extraction produced Palm Kernel Oil (PKO) dan Palm Kernel Meal (PKM). Perez (1997) mentioned that Palm Kernel Meal contains rich arginin, leusin, and sistein matters. Hem et al., (2008), utilizing Palm Kernel Meal pass through bioconversion process for developing larvae Hermetia illucens L. as alternative natural feedstuff in aquaculture industry. Macromolecule composition of Palm Kernel Meal like cellulose, hemicellulose and lignin can be degrade to be simply compound and can be used by another organism like larvae Hermetia illucens L. in bioconversion process. Bioconversion Palm Kernel Meal for feedstuff nutrition consist with microorganism assistance. Suharyanto et al., (2006) define bioconversion as a certain biological process which involving microorganism or enzyme that can change organic matters. Slime molds have great play role in process reduction macromolecule composition of Palm Kernel Meal. Molds have enzyme which can reduce cellulose, hemicellulose and lignin become more simple compound. Study about fermentation fungi already been done through isolation, identification, and fungi screening. However, only a few study about fungi related consist in process bioconversion Palm Kernel Meal reported in Indonesia. This study consist of two part. First part describes the isolation, identification, and growth screening fungi from bioconversion Palm Kernel Meal. Second part of this study describes the fermentation Palm Kernel Meal by selected indigenous fungi. The selected indigenous fungi obtained from result of the first part. The fermentation result included ash matters, crude fiber, crude protein and dry matters experiment. The study was carried out at the Institut de Recherche pour le Developpement (IRD) Laboratory, Depok and the Laboratory of Microbiology, Departement of Biology, UI, Depok during April?Oktober 2009. The isolation of fungi was conducted with spread methods on Potato Dectrose Agar (PDA). Identification of the isolates was carried out on Potato Dectrose Agar (PDA), Czapeck Dox Agar (CDA), and Malt Extract Agar (MEA) based on macroscopic and microscopic morphological observation of the colonies. The Mimura agar (MA) was used for growth fungi screening. The isolation resulted in 15 representative isolates consisting of 4 group of fungi (Aspergillus, Mucor, Penicillium, and Geotrichum). Based on 7 days periods of fermentation processing, Mucor groups had the highest frequency distribution and Geotrichum had the highest quantity. After the growth fungi screening, 4 isolates (P3, P4, P10, P15) was selected for futher study in part II. Microscopic identification showed P3 (Penicillium chrysogenum), P4 (Mucor racemosus), P10 (Aspergillus flavus), and P15 (Geotrichum candidum). Mucor racemosus was the most wide diameter colony on Mimura agar?MA (9 cm) comparing to other isolates. These selected fungi was used for fermentation of Palm Kernel Meal as inoculant. After process bioconversion which fermented was done, proximate analysis were carried out to examine crude protein, crude fiber, ash matters, and dry matters. Ng (2003) methods was used for this Palm Kernel Meal fermentation and Hart & Fisher (1971) was used for proximate analysis. The results after 7 days fermentation showed that the increased nutrition of crude protein composition of Palm Kernel Meal fermented by fungus Aspergillus flavus (1,33%), Geotrichum candidum (5,90%), Mucor racemosus (0,29%), and Penicillium chrysogenum (12,09%). The increased crude fiber contains fermented by Aspergillus flavus (3,03%), Geotrichum candidum (1,93%), Mucor racemosus (4,32%), and Penicillium chrysogenum (14,11%). Chemical cellulose structure and fungi species influence the difference percentage of crude protein and crude fiber. Chemical cellulose structure which amorf shape was more easy to degrade better than crystal shape. Fungi species have difference complexity enzymes (cellulose, hemicellulose, ligninase) and optimum growth level. High oil that can blocked the optimum growth of fungi and raising temperature matter that have involved in aeration and water activity alteration were another influence factor that have made difference percentage of crude protein and crude fiber.

Insect larvae, such as the Black Soldier Fly (Hermetia illucens) larvae (BSFL), have been considered as a protein source which can be produced at a large scale using low cost organic wastes as feeding material. This study was designed to determine the response of BSFL to various waste combinations of vegetable and animal remains, Pak Choi (Brassica chinensis) residues (S) and carp (Cyprinus caprio) fish offal (I)). A total of 540 BSFL were fed with 100 mg/larvae/day combination of vegetable wastes: animal waste 70%: 30% (S > I), 50%: 50% (S = I), and 30%: 70% (S < I). Among the feed combinations, the S < I group showed the best results as it produced the significantly highest weight of BSFL at 122.8 mg/larvae and approximate digestibility of 62.01%, with the least pupae mortality rate at 4.29%.Keywords: bioconversion, biomass, Brassica chinensis, Cyprinus carpio, Hermetia illucen

Sampah organik merupakan salah satu jenis sampah yang memiliki jumlah terbesar di Indonesia. Rumah tangga dan Restoran merupakan sumber terbanyak yang menghasilkan sampah organik khususnya sampah makanan. Salah satu teknologi untuk mengatasi hal tersebut adalah melalui proses biokonversi dengan larva Black Soldier Fly (BSF). Penelitian ini bertujuan untuk [1] menganalisis karakteristik sampah organik rumah tangga dan restoran; [2] menganalisis proporsi sampah organik dari rumah tangga dan restoran terhadap efektivitas biokonversi larva BSF; [3] menganalisis pengaruh penambahan substrat dengan campuran sampah organik rumah tangga dan restoran terhadap efektivitas biokonversi larva BSF. Instrumen yang digunakan dalam penelitian ini adalah eksperimen. Berdasarkan hasil analisis proksimat, sampah organik rumah tangga, sampah organik restoran, dan lumpur susu memiliki kadar air masing-masing sebesar 73%, 81%, dan 82%. Berdasarkan hasil biokonversi, memberikan gambaran bahwa kelompok kontrol dengan proporsi sampah rumah tangga dan restoran 30:70 memiliki nilai WRI tertinggi jika dibandingkan dengan proporsi lainnya. Hasil tersebut dapat disebabkan karena sampah organik restoran memiliki keadaan yang lebih murni dibandingkan sampah organik rumah tangga. Sementara, penambahan substrat limbah susu tidak dapat meningkatkan nilai WRI, namun penambahan substrat dapat meningkatkan terhadap nilai ECD, RGR, dan RSR. Hasil tersebut dapat disebabkan karena lumpur susu memiliki kadar air yang lebih tinggi jika dibandingkan dengan kadar air yang dimiliki oleh sampah organik rumah tangga dan sampah organik restoran. ......Organic waste is one type of waste that has the largest amount in Indonesia. Households and restaurants are the largest sources of organic waste, especially food waste. One technology to overcome this is through a bioconversion process with Black Soldier Fly (BSF) larvae. This research aims to [1] analyze the characteristics of household and restaurant organic waste; [2] analyzed the proportion of organic waste from households and restaurants on the effectiveness of bioconversion of BSF larvae; [3] analyzed the effect of adding a substrate mixed with household and restaurant organic waste on the effectiveness of bioconversion of BSF larvae. The instrument used in this research was an experiment. Based on the results of proximate analysis, household organic waste, restaurant organic waste, and milk sludge have water contents of 73%, 81%, and 82% respectively. Based on the bioconversion results, it shows that the control group with a proportion of household and restaurant waste of 30:70 has the highest WRI value when compared to other proportions. These results can be caused because restaurant organic waste is purer than household organic waste. Meanwhile, the addition of milk waste substrate cannot increase the WRI value, but the addition of substrate can increase the ECD, RGR and RSR values. This result could be caused by milk sludge having a higher water content compared to the water content of household organic waste and restaurant organic waste.

Potensi Indonesia sebagai salah satu penghasil minyak cengkeh terbesar di dunia didukung dengan pengembangan perkebunan cengkeh di Indonesia. Sulawesi Utara merupakan provinsi penghasil minyak cengkeh di Indonesia. Desa Liandok yang berada pada kabupaten Minahasa Selatan, provinsi Sulawesi Utara memiliki area perkebunan cengkeh yang luas. Penelitian ini bertujuan untuk mengkarakterisasi gen ech dan gen fcs pada bakteri tanah dari perkebunan cengkeh Desa Liandok, Minahasa Selatan. Bakteri tanah dari perkebunan cengkeh di Desa Liandok, Minahasa Selatan diisolasi dengan beberapa medium selektif. Ektraksi DNA dilakukan dengan menggunakan Geneaid PrestoTM Mini gDNA Bacteria Kit. Isolasi genom dari ekstraksi DNA dilakukan dengan elektroforesis gel agarosa. Primer forward dan primer reverse didesain dengan multiple alignment sekuens yang menyandi gen ech dan gen fcs dari bakteri Pseudomonas sp. pada data NCBI GenBank. Analisis PCR dilakukan melalui primer forward dan primer reverse untuk mendeteksi gen ech dan gen fcs pada isolat. Selanjutnya, amplikon dianalisis dengan elektroforesis gel agarosa untuk menunjukan pita pada daerah gen ech dan gen fcs. Analisis secara molekuler dilakukan dengan mengamplifikasi gen 16S rRNA dengan metode PCR menggunakan primer universal 27F dan 534R dan dilanjutkan dengan sekuensing terhadap gen 16S rRNA. Langkah terakhir, yaitu dilakukan analisis hasil sekuensing menggunakan metode BLAST di NCBI. Keberadaan gen ech dan gen fcs pada isolat bervariasi. Sembilan isolat dari total 22 isolat memiliki gen ech dan gen fcs. Hasil BLAST terhadap urutan nukleotida gen 16S rRNA dari tiga isolat yang disekuensing mempunyai kesamaan 99% dengan bakteri Pseudomonas nitroreducens dan satu isolat mempunyai kesamaan 96% dengan Pseudomonas denitrificans. Sebagai kesimpulan, bakteri tanah pada perkebunan cengkeh di Desa Liandok, Minahasa Selatan memiliki gen ech dan gen fcs yang berpotensi untuk melakukan konversi eugenol menjadi vanillin.
Indonesias potential as worlds largest clove oil producer is supported by the development of clove plantations in Indonesia. North Sulawesi is a province that playing the biggest role in producing clove oil in Indonesia. Desa Liandok is located in Minahasa Selatan, North Sulawesi which has a large areal of clove oil plantation. This study was aimed to characterize ech and fcs genes in soil bacteria from clove plantation in Desa Liandok, South Minahasa which has the potential to bioconvert eugenol to vanillin. The soil bacteria from clove plantations in Desa Liandok, South Minahasa was isolated using selective mediums. DNA extraction was carried out using Geneaid PrestoTM Mini gDNA Bacteria Kit. The isolated genomes from DNA extraction were analyzed and carried out by agarose gel electrophoresis. Both primers for PCR were designed by aligning multiple ech and fcs genes sequences of Pseudomonas sp. in NCBI GenBank data. PCR analysis was performed within forward and reverse primers to detect ech and fcs genes in the isolate. Furthermore, the amplicons was analyzed using agarose gel electrophoresis to show ech and fcs genes bands. Molecular analysis was carried out by amplifying the 16S rRNA gene with the PCR method using universal primers 27F and 534R and continued with sequencing of the 16S rRNA gene. The last step was to analyze the result of DNA sequencing using BLAST method in NCBI. The existence of ech and fcs genes in each isolate were varied. BLAST analysis against nucleotide sequence of the 16S rRNA gene from three isolates that were sequenced possess 99% similarities with Pseudomonas nitroreducens and one isolate possesses 96% similarities with Pseudomonas denitrificans. Nine out of 22 isolates contained both fcs and ech genes. To conclude, soil bacterias in clove plantation in Desa Liandok, South Minahasa have ech and fcs genes which have the potential to bioconvert eugenol to vanillin.

Sampah sisa makanan menjadi salah satu masalah di Universitas Indonesia karena belum terdapat metode yang efektif dan efisien dalam pengelolaan limbah kantin dari fakultas-fakultas yang ada. Salah satu teknik pengelolaan limbah kantin adalah dengan pengomposan menggunakan larva Black Soldier Fly (BSF). Adanya proses fermentasi dan penguraian bahan kompleks oleh mikroorganisme pada substrat organik mampu mengoptimalkan daya biokonversi larva BSF. Namun, hingga saat ini, penelitian terkait pengaruh fermentasi limbah kantin terhadap efektivitas biokonversinya dengan larva BSF belum ditemukan sehingga penelitian ini perlu dilakukan. Penelitian ini bertujuan untuk mengidentifikasi pengaruh perlakuan durasi fermentasi limbah kantin dengan mikroorganisme EM4 terhadap efektivitas biokonversi limbah kantin menggunakan larva BSF dan juga kandungan kompos yang dihasilkan berdasarkan standar SNI 19-7030-2004. Metode penelitian yang digunakan, yakni berupa kombinasi fermentasi EM4 dengan perbedaan perlakuan durasi (0, 5, dan 10 hari) serta larva BSF sebagai agen pengomposan pada limbah kantin. Analisis data dilakukan terhadap berat biomassa larva BSF dan kompos, substrate consumption, growth rate, bioconversion rate, waste reduction index, serta analisis kandungan unsur hara kompos. Hasil penelitian menunjukkan bahwa efektivitas biokonversi larva BSF tertinggi didapatkan oleh perlakuan durasi fermentasi limbah kantin dengan EM4 selama 10 hari (F10) dengan berat biomassa total larva BSF sebesar 50,09 gram; nilai bioconversion rate sebesar 24,55%; nilai waste reduction index (WRI) sebesar 10,79%; dan berat kompos sebesar 27,38 gram. Sementara itu, sebagian besar kandungan unsur hara kompos pada semua perlakuan sudah sesuai dengan standar SNI 19-7030-2004, kecuali nilai kadar C-organik pada perlakuan F10 serta nilai rasio C/N pada semua perlakuan lainnya (F0, F5, F10, KL dan KF). Pada penelitian ini, didiskusikan juga potensi biokonversi dan peluang bioekonomi pengelolaan limbah kantin FMIPA UI dengan larva BSF. ......Food waste management has become one of the issues at the University of Indonesia due to the lack of an effective and efficient method in managing canteen waste from existing faculties. One common technique in food waste management is composting using larvae of Black Soldier Fly (BSF). The fermentation process and decomposition of complex substances by microorganisms in the organic substrate can optimize the bioconversion efficiency of BSF larvae. However, studies related to the influence of canteen waste fermentation on the effectiveness of bioconversion with BSF larvae have not been discovered to date. Hence, further study regarding this subject is necessary. This study aimed to identify the effects of canteen waste fermentation treatment with EM4 microorganisms on the effectiveness of canteen waste bioconversion using BSF larvae and the content quality of the resulting compost based on the SNI 19-7030-2004 standard. The method used in this study is combining EM4 fermentation with different durations treatment (0, 5, and 10 days) and BSF larvae as composting agent on canteen waste. Data analysis included the weight of BSF larvae biomass and compost, substrate consumption, growth rate, bioconversion rate, waste reduction index, and content quality analysis of the compost. The results showed that the highest effectiveness of bioconversion by BSF larvae was obtained with a 10-day EM4 fermentation treatment (F10). The total biomass of F10 BSF larvae reached 50,09 grams; with bioconversion rate of 24,55%; waste reduction index of 10,79%; and a compost weight of 27,38 grams. Meanwhile, most of the content qualities in the compost for all treatments were following the SNI 19-7030-2004 standard, except for the C-organic content value in the F10 treatment and the C/N ratio values in all other treatments (F0, F5, F10, KL, and KF). In this study, the bioconversion potency and the bioeconomic opportunities of managing the waste from the FMIPA UI canteen using BSF larvae were also discussed.
