Telah dilakukan ekstraksi dan isolasi senyawa kimia serta uji antibakteri terhadap kulit batang tumbuhan Combretum punctatum yang merupakan salah satu tumbuhan terdapat di Indonesia. Metoda yang digunakan dalam pengerjaan isolasi adalah cara ekstraksi fraksinasi dengan pelarut campuran kloroform-metanol (1:1), kemudian dilanjutkan dengan pelarut n-heksana dan kloroform pada temperatur ruang. Untuk pemisahan dilakukan secara kromatografi kolom dan kromatografi lapis tipis (KLT). Penetapan struktur molekul dilakukan dengan cara spektroskopi. Pada uji antibakteri, untuk penentuan diameter daerah hambatan (DDH) digunakan metode kertas cakram dan untuk penentuan konsentrasi hambatan minimum (KHM) digunakan metoda penipisan lempeng agar dan pengenceran serial tabung. Larutan uji yang digunakan adalah larutan infus dan ekstrak kloroform-metanol (1:1). Hasil isolasi dari fraksi n-heksana diperoleh senyawa β-sitosterol dan dari fraksi kloroforrn diperoleh senyawa yang diperkirakan adalah asam 3β-hidroksi-19αH-2O(29)-lupen 28α-oat dan diduga merupakan suatu senyawa baru. Hasil percobaan uji antibakteri menunjukkan bahwabaik larutan infus maupun ekstrak kloroform-metanol (1:1) mempunyai daya hambat terhadap kuman Escherichia cols, Staphylococcus aureus dan Leptospira bataviae.

The method used in this isolation is the fractionated extraction with the mixed solvents chloroform-methanol (1:1) with is later proceed with the n-hexane and chloroform at the room temperature. The isolated compounds were characterized by spectroscopic methods. The antibacterial test was carried out, based on inhibition zona diameter by using diffusion method and minimum inhibitory concentration by using dilution method. The samples being used for antibacterial activity are the infuse and the extract from chloroform-methanol (1:1) fraction. The isolated compounds from the n-hexane extract is identified as R-sitosterol and from chloroform extract was isolated 3β-hidroxy-19αH-20(29)-lupen-28α-oic acid and predicted as a new compound. For antibacterial test, both the infuse and the extract chloroform-methanol (1:1), respectively, exhibit activities against Escherichia coil, Staphylococcus aureus and Leptospira bataviae.
