Pada penelitian ini dilakukan pengujian deteksi 8-hidroksi-2' deoksiguanosin (8-OHdG), akibat adanya paparan bahan tambahan pangan Tertier Butyl Hidroquinon(TBHQ) dan Bisphenol A(BPA) secara in vitro kemudian dipelajari pula efek sinergis atau antagonis yang di timbulkan dengan adanya variasi waktu inkubasi dan paparan sinar UV A. Selain itu variasi eksperimen pada penelitian ini adalah pada pH 7,4 dan pH 8,4 waktu inkubasi dan paparan sinar UV-B 7 jam dan 5 jam. Analisis dilakukan dengan menggunakan alat UHPLC dengan detector UV-VIS pada panjang gelombang 254 nm. Digunakan metanol dan buffer Na-fosfat sebagai eluen dengan perbandingan (15:85). Hasil menunjukan bahwa konsentrasi 8-OHdG pada waktu inkubasi dan paparan sinar UV yang lebih lama akan menghasilkan konsentrasi yang lebih tinggi. Pada variasi pH, pH yang lebih tinggi didapatkan konsentrasi 8-OHdG yang lebih besar. Efek pencampuran pada 2 xenobiotika, yaitu BPA dan TBHQ menimbulkan efek sinergis yang berdampak pada meningkatnya konsentrasi 8-OHdG yang terbentuk.

In this study an 8-hydroxy-2-deoxiguanosin (8-OHdG) detection experiment was conducted, consequently it has to do with the research material of Tertier Butyl Hydroquinone (TBHQ) and Bisphenol A (BPA) in vitro and can also be accessed using synergistic or antagonistic properties. caused by the variation of incubation time and exposure to UV rays A. In addition to the variation of experiments in this study at pH 7.4 and pH 8.4 incubation time and presentation of UV-B rays 7 hours and 5 hours. The analysis was performed using a UHPLC with a UV-VIS detector at a wavelength of 254 nm. Used methanol and Na-phosphate buffer as eluent with allocated (15:85). The results show that the concentration of 8-OHdG at incubation time and longer exposure to UV light will result in higher concentrations. With pH variations, a higher pH is obtained at a greater concentration of 8-OHdG. The mixing effect on 2 xenobiotics, namely BPA and TBHQ, has a synergistic effect that affects the concentration of 8-OHdG that is formed.
