"Latar Belakang: Penelitian menunjukkan bahwa paparan logam Ni pada alloy NiCr pada tubuh manusia melalui stres oksidatif.Tujuan: Tesis ini membahas pengaruh logam nikel pada protesa alloy Ni-Cr terhadap pembentukan DNA Adduct 8-Hydroxy-2-Deoxyguanosine yang dievaluasi berdasarkan konsentrasi 8-OHdG dalam urin dengan alat LC-MS/MS.Metode: Regresi linier dilakukan untuk menghitung jumlah konsentrasi 8-OHdG antara kelompok kontrol dan kelompok pengguna alloy NiCr yang kemudian dilanjutkan dengan uji SPSS Independent T-test.Hasil: Terdapat pengaruh logam nikel pada protesa alloy Ni-Cr terhadap pembentukan DNA Adduct 8-OHdG yang diuji dengan hasil rerata lebih tinggi dibandingkan kelompok kontrol namun tidak berbeda signifikan.Kesimpulan: Hasil penelitian menyarankan bahwa perlunya edukasi terhadap pasien pengguna alloy Ni-Cr untuk tidak melakukan kebiasaan buruk seperti merokok, serta menjaga kesehatan rongga mulut dengan baik agar tidak meningkatkan produksi ROS yang dihasilkan oleh alloy Ni-Cr.

Background: Research shows that exposure to Ni metal in NiCr alloys in the human body through oxidative stress.Objectives: This thesis discusses about the effect of nickel metal on Ni-Cr alloys on the formation of DNA Adduct of 8-Hydroxy-2-Deoxyguanosine which is evaluated based on 8-OHdG concentrations in urine with LC-MS/MS.Methods: Linear regression was carried out to calculate the 8-OHdG concentration between the control group and the NiCr alloy user group which was then followed by the SPSS Independent T-test.Results: The effect of nickel metal on Ni-Cr alloy prostheses on DNA formation of 8-OHdG adducts were tested with higher mean results than control group but not significantly different.Conclusions: The results suggest that needed for educating patients of Ni-Cr alloy users for not doing bad habits such as smoking, as well as maintaining oral hygiene very well in order not to increase production of ROS as generated by Ni-Cr alloys."
