"Pedagang besar farmasi (PBF) adalah perusahaan berbentuk badan hukum yang memiliki izin untuk pengadaan, penyimpanan, penyaluran obat dan/atau bahan obat dalam jumlah besar sesuai ketentuan peraturan perundang-undangan (CDOB, 2020) harus memiliki fasilitas (warehouse facility) yang memadai dan mendukung untuk menyimpan produk obat sebelum didistribusikan kembali seperti PT Enseval Putera Megatrading 4 fasilitas gudang yaitu ambient room, cool room, cold storage, dan frozen storage. Ambient room merupakan fasilitas yang digunakan untuk menyimpan produk di bawah 30°C, cool room di bawah 25°C, cold storage 2-8°C, frozen storage -40°C. Cool room, cold storage, dan frozen storage merupakan fasilitas yang digunakan pada cold chain management. Fasilitas tersebut didukung oleh ruangan dan alat yang harus memenuhi spesifikasi dari hasil kualifikasi. Kualifikasi alat atau mesin meliputi kualifikasi desain, kualifikasi instalasi, kualifikasi operasional, dan kualifikasi kinerja (CDOB, 2020). PT Enseval Putera Megatrading DC 1 memiliki freezer yang akan digunakan untuk mendukung cold chain management yang ditandai sebagai F.16. Freezer F.16 telah dilakukan kualifikasi desain dan kualifikasi instalasi yang selanjutnya akan dilakukan kualifikasi operasional pada kondisi kosong. Dari hasil kualifikasi operasional yang dilakukan, Freezer F.16 memenuhi spesifikasi kualifikasi operasional dengan pengaturan posisi pada beberapa titik yang akan dipastikan kembali pada kualifikasi kinerja.

Selain itu, proses rekualifikasi perlu dilakukan secara berkala untuk menjaga kualitas penyimpanan yang optimal.

Chest Freezer, as one of the essential equipment used in making and storing ice packs for CCP product shipments, needs to undergo a performance qualification process first. This performance qualification of the Chest Freezer is conducted to ensure that the storage temperature of the ice packs meets the standards. The results of the qualification on Chest Freezer AB-1200-T-X ID. P05 indicate that the temperature distribution falls within the expected range (-10-(-15)oC), with some deviations at the lowest temperatures. These deviations are caused by operational activities such as opening and closing the freezer or refilling ice packs. However, the distribution of cold temperatures tends to be uniform. Chest Freezer AB-1200-T-X ID. P05 meets the acceptance criteria in performance qualification with a uniform temperature distribution. Setting the temperature at -18oC aims to prevent the ice pack from melting during storage. Additionally, periodic requalification processes are necessary to maintain optimal storage quality."

Agar menjamin kinerja freezer berjalan sesuai spesifikasi yang diinginkan, maka perlu dilakukan kualifikasi kinerja secara berkala. kualifikasi kinerja merupakan prosess yang dilakukan untuk mendapatkan dan mendokumentasikan sistem dan atau alat bekerja sesuai dengan metode dan spesifikasi yang diinginkan dan telah disetujui. Kualifikasi kinerja dilakukan dengan cara meletakan thermometer data logger yang telah dibungkus plastik ke dalam freezer di beberapa lokasi selama 3 x 24 jam. Berdasarkan hasil kualifikasi kinerja diketahui bahwa pendistribusian dingin pada combi cooler Freezer merk Gea model PF-20 F.15 tidak merata dan terjadi Penyimpangan suhu di beberapa lokasi pada freezer. Tindak lanjut yang dapat dilakukan yaitu pembuatan ice pack dilakukan pada posisi yang memenuhi kriteria kualifikasi kinerja pada freezer.

To ensure the performance of the freezer runs according to the desired specifications, it is necessary to carry out performance qualifications on a regular basis. Performance qualification is a process carried out to obtain and document systems and or work tools according to the desired and approved methods and specifications. Performance qualification is carried out by placing a plastic-wrapped data logger thermometer into the freezer at several locations for 3 x 24 hours. Based on the outcomes of performance qualifications, it can be concluded that the Gea brand Freezer combi cooler Freezer model PF-20 F.15's cold distribution is uneven and that temperature deviations exist in several places inside the freezer. The follow-up that can be done is to make ice packs in a position that meets the performance qualification criteria in the freezer."

Metodologi penelitian yang digunakan yaitu menggunakan rancangan observasional dengan memeriksa kualifikasi operasional distribusi dingin freezer dalam keadaan kosong. Didapatkan kesimpulan diantaranya, Hasil kualifikasi operasi 1, 2, dan 4 memenuhi persyaratan sedangkan hasil kualifikasi operasi 3 memenuhi persyaratan dengan adanya justifikasi pada posisi 1, 2, 3 dan 4.

Therefore, it is necessary to carry out operational qualifications to ensure that the equipment used meets the acceptance criteria and can function properly. The purpose of preparing this work practice report, among other things, is to find out the results of operation qualification 1: turning on and off the power of the combi cooler PF-20 F.15 freezer; To find out the results of operation qualification 2: setting the temperature of the combi cooler PF-20 F.15 freezer; To find out the results of operation qualification 3: cold distribution in an empty freezer freezer combi cooler PF-20 F.15; To find out the results of operation qualification 4: defrost freezer combi cooler PF-20 F.15 operation. The research methodology used was an observational design by examining the operational qualifications of the cold freezer distribution in an empty state. The conclusions obtained include the results of operational qualifications 1, 2 and 4 meet the requirements while the results of operational qualification 3 meet the requirements with justification in positions 1, 2, 3 and 4."

Machine qualification consists of several qualification stages which are carried out before the machine is used for daily operations. In this research, operational qualification was carried out in accordance with the company's qualification procedures. Operational qualification (OQ) is performed to establish that the installation and all systems and subsystems operate effectively and consistently when the storage area is empty. Operational qualification results for Chest Freezer AB-1200-T-X ID. P05 empty condition has been implemented with the results meeting the acceptance criteria with justification. Therefore, qualification testing can proceed to the performance qualification stage."

Penelitian ini bertujuan untuk memastikan bahwa Up Right Medical Freezer LF-600 menunjukkan kinerja yang sesuai dengan pengoperasian alat yang konsisten. Kualifikasi kinerja Up Right Medical Freezer dilakukan dengan pemetaan suhu menggunakan sembilan titik pemantauan data logger yang dilakukan selama 3 x 24 jam. Hasil penelitian menunjukkan kualifikasi kinerja Up Right Medical Freezer memenuhi kriteria penerimaan dengan adanya justifikasi untuk penyimpanan produk sesuai dengan spesifikasi yang dipersyaratkan.

This study aims to ensure that the Up Right Medical Freezer LF-600 shows appropriate performance with consistent equipment operation. Up Right Medical Freezer performance qualification was carried out by temperature mapping using nine data logger monitoring points carried out for 3 x 24 hours. The research results show that the performance qualifications of the Up Right Medical Freezer meet the acceptance criteria with justification for product storage by the required specifications."

This storage condition is very influenced by temperature and velocity distribution in cold storage to decide the effectiveness of cold storage that is made, so that the need of uniform temperature and velocity distribution is very important to achieve the uniform condition and temperature storage.

This research uses three research methods, are: designing method bases on ASHRAF' HANDBOOK 1998 to predict total cooling loads, experiment method of temperature distribution data with thermocouple until measurement temperature of 8C is obtained, and computation method with CFD aided through PHOENICS soffware in making grid and object computation on Cartesian coordinate through windows VREditor, and simulating the cotd storagemethod."

"Salah satu cara untuk menjamin keaslian mutu obat diantaranya adalah memastikan peralatan dan instrumentasi penunjang yang digunakan selama proses supply chain obat bekerja dan berfungsi dengan akurat seperti yang dipersyaratkan pada CDOB. Pemastian tersebut dilakukan melalui kegiatan kualifikasi. Pelaksanaan kualifikasi tersebut dilakukan secara berurut-turut dimulai dari kualifikasi desain hingga kinerja. Dalam pelaksanaannya, PT. Enseval Putera Megatrading Cikarang melakukan kualifikasi terhadap semua peralatan yang berhubungan dengan produk, salah satunya yaitu Freezer untuk membuat dan menyimpan ice gel. Kualifikasi dilakukan dengan menguji dan memastikan kinerja freezer dalam membekukan ice gel dengan baik pada distribusi suhu antara -15 °C sampai dengan -25 °C. Hasil pengujian distribusi dingin pada freezer dengan muatan ice gel, didapatkan bahwa pada 9 titik pemantauan menghasilkan suhu rata-rata minimal -19.43, maksimal -5,53 dan rata-rata - 14.46°C. Pada 9 titik temperature mapping, hasil rekaman data logger pada posisi 1,2,3,4 berada di ruang range penerimaan (-15 C - (-25 C). Sementara pada data logger posisi 6,7,8,9 suhu baru stabil di range penerimaan setelah kurang lebih 12 jam. Berdasarkan penyimpangan yang terjadi, dapat dilakukan kualifikasi ulang dengan melakukan conditioning data logger terlebih dahulu sehingga data logger dapat merekam di suhu awal yang stabil.

In its implementation, PT. Enseval Putera Megatrading Cikarang has qualified all equipment related to the product, one of which is a freezer for making and storing ice gel. Qualification is carried out by testing and ensuring the freezer's performance in freezing ice gel properly at a temperature distribution between -15 °C to -25 °C. The results of testing cold distribution in freezers with ice gel loads, it was found that at 9 monitoring points the minimum average temperature was -19.43, maximum -5.53 and average -14.46°C. At 9 temperature mapping points, the data logger recording results at positions 1,2,3,4 were in the reception range (-15 C - (-25 C). Meanwhile at data logger positions 6,7,8,9 the temperature was just stable in the acceptance range after approximately 12 hours. Salah satu bab dalam CDOB membahas tentang aspek ketentuan khusus produk rantai dingin atau Cold Chain Product (CCP) yang merupakan produk farmasi yang sensitif terhadap temperatur sehingga penyimpanan dan pengirimannya memerlukan kontrol temperatur terus menerus untuk menjamin agar karakteristik, stabilitas, khasiat dan keamanan produk tidak berubah. Untuk menjaga kualitas produk rantai dingin yang didistribusikan sesuai dengan CDOB, maka diperlukan salah satu fasilitas berupa Chest Freezer yang digunakan untuk menyimpan ice pack yang digunakan untuk menjaga suhu produk rantai dingin saat pengiriman. Penelitian ini dibuat untuk melakukan pengujian kualifikasi operasional pada salah satu Chest Freezer yang ada di PT. Enseval Putera Megatrading Tbk untuk memastikan freezer tersebut memenuhi spesifikasi yang telah ditentukan berdasarkan protokol kualifikasi operasional dan dapat digunakan untuk menyimpan ice pack. Penelitian dilakukan dengan pengujian dan pengamatan kualifikasi operasional Chest Freezer AB-1200-T-X ID.P04 sesuai dengan protokol yang berlaku. Pengujian terdiri atas beberapa prosedur yaitu menyalakan dan mematikan daya, pengaturan suhu, distribusi dingin pada keadaan kosong dan operasi defrost. Setelah pengujian, hasil didokumentasikan dan diolah ke tabel dan grafik Microsoft Excel untuk melihat distribusi temperatur freezer dan keberterimaannya. Penelitian menunjukkan hasil memenuhi kriteria penerimaan.

To maintain the quality of distributed Cold Chain Products in accordance with GDP, facilities such as a Chest Freezer are required to store ice packs used to maintain the temperature of the products during transportation. This research was conducted to perform operational qualification testing on one of the Chest Freezers at PT. Enseval Putera Megatrading Tbk to ensure that the freezer meets specified requirements based on operational qualification protocols and can be used to store ice packs. The study involved testing and observing the operational qualification of the Chest Freezer AB-1200-T-X ID.P04 according to applicable protocols. Testing procedures included powering on and off, temperature setting, cold distribution in empty condition, and defrost operation. After testing, results were documented and processed into tables and graphs using Microsoft Excel to assess freezer temperature distribution and acceptance criteria. The research findings indicate that the results meet acceptance criteria."
