Abstrak :
Pengujian stabilitas terhadap produk obat merupakan salah satu pengujian yang harus dilakukan sebelum produk diproduksi dan dipasarkan. Pada pengujian, apabila hasil uji yang dilakukan tidak masuk kedalam syarat keberterimaan, hasil uji tersebut disebut Hasil Uji di Luar Spesifikasi (HULS). Pada proses analisis kadar asam folat dalam sediaan multivitamin kaplet salut selaput, diperoleh hasil uji yang berada di luar syarat keberterimaan asam folat. Data hasil pengujian diperoleh dari penelitian penetapan kadar sebelumnya dan pengamatan di lapangan secara langsung.Hasil pengujian yang dilakukan terhadap dua bets dengan perbedaan tahun produksi menunjukkan bahwa nilai kadar asam folat yang rendah tidak disebabkan oleh umur simpan produk. Nilai kadar yang tidak memenuhi syarat dapat disebabkan oleh pemisahan peak yang tidak sempurna. Modifikasi pada tahap preparasi sampel belum optimal. Optimasi kondisi kromatografi pada laju alir 1,0 mL/menit menunjukkan peningkatan kadar asam folat yang signifikan. Perlu dilakukan verifikasi metode untuk dapat menetapkan kadar asam folat dengan metode yang tepat. ......Stability testing of drug products is one of the tests that must be carried out before the product is produced and marketed. In testing, if the test results carried out do not enter into the acceptance requirements, the test results are called Out-of-Specification Test Results (HULS). In the process of analyzing folic acid levels in film-coated multivitamin caplet preparations, test results were obtained that were outside the acceptance requirements for folic acid. Data on test results were obtained from previous assay studies and direct field observations. Test results carried out on two batches with different years of production showed that the low folic acid content was not caused by product shelf life. Level values that do not meet the requirements can be caused by imperfect peak separation. Modifications at the sample preparation stage have not been optimal. Optimization of chromatographic conditions at a flow rate of 1.0 mL/min showed a significant increase in folic acid levels. It is necessary to verify the method to be able to determine folic acid levels with the right method.

Abstrak :
Investigasi terhadap terhadap Hasil Uji di Luar Spesifikasi (HULS) diperlukan untuk mengidentifikasi akar permasalahan dari HULS sehingga Tindakan Korektif dan Tindakan Perbaikan (TKTP) atau Corrective Action and Preventive Action (CAPA) dapat diinisiasi untuk mencegah terjadinya keterulangan (recurrence) dari HULS. Pemantauan parameter air berdasarkan parameter nilai organik total (Total Organic Carbon/TOC) dibutuhkan sebagai bentuk pemastian mutu sistem air yang digunakan dalam kegiatan produksi obat. Laporan ini disusun untuk mengetahui imlementasi dari investigasi HULS dan inisiasi terhadap TKTP atau CAPA. Laporan ini disusun di PT. Kalbio Global Medika, Cikarang, Jawa Barat pada periode Mei hingga Juni 2022 terhadap sampel air murni (Pure Water). Berdasarkan hasil investigasi HULS, HULS disebabkan oleh pengambil sampel (sampler) tidak menempatkan vial TOC langsung pada kotak sampel yang mengakibatkan sampel terkontaminasi oleh alkohol. Pada laporan ini, diagram fishbone digunakan sebagai langkah awal dalam mengidentifikasi akar permasalahan HULS. TKTP yang diinisasi adalah penambahan tanda (sign tag) pada kotak sampel TOC. ......Investigation on Out-of-Specification (OOS) results are needed to identify the root causes of OOS so that Corrective Action and Corrective Action (CAPA) can be initiated to prevent recurrence of OOS. Water quality parameters based on total organic carbon (TOC) is needed as a form of quality assurance of water systems used in drug production activities. This report aims to find out the implementation of OOS investigations and to initiate CAPA. This report was composed on May to June 2022 at at PT. Kalbio Global Medika, Cikarang, West Java, with Pure Water (PW) as the samples. Based on the results of the OOS investigation, OOS was caused by the sampler not placing the TOC vial directly in the sample box which resulted in the sample being contaminated with alcohol. In this report, a fishbone diagram is used as a first step in identifying the root causes of OOS. The CAPA initiated is the addition of a sign (sign tag) to the TOC sample box.
