"Inflamasi kronik saluran cerna merupakan salah satu faktor risiko untuk berkembang menjadi kanker kolorektal (KKR). Probiotik diharapkan dapat menjadi faktor proteksi imunologis pada host melalui regulasi, stimulasi, dan modulasi respons imun untuk menurunkan inflamasi. Dadih, merupakan probiotik lokal asal Sumatera Barat yang mengandung Lactococcus lactis ssp. lactis diharapkan dapat menjaga homeostasis saluran cerna. Tujuan dari penelitan ini adalah untuk menganalisis efek dadih terhadap inflamasi usus serta untuk mengetahui korelasi dengan ekspresi COX-2, TGF-β, dan IL-10 pada mencit Balb/c yang diinduksi DSS. Penelitian ini menggunakan Bahan Biologi Tersimpan (BBT) Formalin Fixed Paraffin Embedded (FFPE) jaringan usus mencit Balb/c dan dilakukan scoring histopatologi dan pulasan imunohistokimia. score luas inflamasi menunjukkan adanya perbedaan signifikan antar kelompok perlakuan (p 0,003). COX-2, TGF-β, dan IL-10 terekspresi kuat pada sel epitel mukosa dan sel limfosit. Ekspresi COX- 2 dan TGF-β pada mencit Balb/c yang diinduksi DSS lebih tinggi dibandingkan dengan mencit yang diinduksi dadih dan DSS. Sedangkan ekspresi IL-10 pada mencit Balb/c yang diinduksi DSS lebih rendah dibandingkan dengan mencit yang diinduksi dadih dan DSS. Induksi dadih dapat menekan luas inflamasi, menurunkan ekspresi COX-2 dan TGF-β, dan meningkatkan ekspresi sitokin anti inflamasi IL- 10. Terdapat korelasi yang signifikan antara score luas inflamasi dengan ekspresi COX-2, TGF-β, dan IL-10.

Chronic intestinal inflammation is a major risk factor that lead to the development of Colorectal Cancer. Probiotics confer immunological protection to the host through the regulation, stimulation, and modulation of immune responses. Dadih, a local probiotic from West Sumatera, Indonesia, containing Lactococcus lactis ssp. lactis expected can be used for preventive treatment to maintain the intestinal homeostasis. The aim of this study is to analyse the effect of dadih to the intestinal inflammation and correlation with COX-2, TGF-β, and IL-10 expression in DSS-induced Balb/c mice. The study was conducted using Formalin Fixed Paraffin Embedded of Balb/c mice colon with histopathological scoring and immunohistochemistry staining. Score of inflammation severity showed a significant difference between the four groups (p 0.003). Strong intensity of COX-2, TGF-β, and IL-10 expression was found in the mucosal epithelium and lymphocyte cells. COX-2 and TGF-β expression in DSS-induced Balb/c mice is higher than Dadih+DSS-induced. On the other hand, IL-10 expression is lower than Dadih+DSS-induced. The induction of dadih can suppress the inflammation severity score, reduce the expression of COX-2 and TGF-β, and increase the expression of anti-inflammatory cytokine IL-10. In addition, there is significant correlation between inflammation severity score to the expression of COX-2, TGF-β, and IL-10. "
