Abstrak :
Objektif. Preeklampsia adalah penyakit pada kehamilan ditandai dengan hipertensi dan proteinuria. Di Indonesia, preelampsia/eklampsia merupakan penyebab utama mortalitas dan morbiditas ibu dan anak. Stress oksidatif pada plasenta dan sistem sirkulasi menyebabkan disfungsi dan kerusakan sel endotel. Stres oksidatif di plasenta menyebabkan gangguan pertumbuhan janin. HSP70 adalah molekul protein yang sangat penting untuk penyembuhan sel dan menjaga homeostasis. Tujuan penelitian untuk membandingkan kadar MDA dan HSP70 yang diproduksi di plasenta pada kehamilan dengan preeklampsia berat, ringan dan kehamilan normal. Plasenta didonorkan secara sukarela dari ibu2 yang melahirkan dengan preeklampsia ringan (N=10), preeklampsia berat (N=10) dan kehamilan normal (N=10). Plasenta dikultur dengan RPMI dan FBS 20%, pada hari ke 3, supernatant diambil. Diperiksa kadar Malondealdehida (MDA), petanda untuk stres oksidatif dan kadar HSP70. Kadar MDA diukur dengan spektrofotometer pada panjang gelombang 530nm. Kadar HSP 70 diukur dengan metoda enzyme-linked immunosorbent assay. Kadar rata2 MDA pada preeklampsia berat (7,13+5,36 nmol/ml), preeklampsia ringan (4,82+2,47 nmol/ml) dan hamil normal (4,87+2,4 nmol/ml). Kadar MDA pada preeklampsia berat paling tinggi, tetapi perbedaan tersebut tidak berbeda bermakna. Kadar rata2 HSP70 pada preeklampsia ringan tertinggi (10,15+12,39 nmo/ml) dibandingkan dengan kadarnya pada preeklampsia berat (3,78 +3,07 nmol/ml) dan kehamilan normal (3,76+4,65nmol/ml), namun perbedaan ini tidak berbeda bermakna. Walaupun demikian, kadar HSP sangat tinggi pada preeklampsia ringan menunjukkan respons homeostasis relatif tinggi. Hal ini tidak ditunjukkan pada preeklampsia berat. Kadar rata2 MDA dan HSP70 pada preeklampsia berat, ringan maupun hamil normal tidak berbeda bermakna. Kadar HSP yang sangat tinggi pada preeklampsia ringan menunjukkan respons homeostasis masih tinggi. ......Objective: Preeclampsia is a disease in pregnancy and characterized by hypertension and proteinuria. Preeclampsia and eclampsia are the most causes of maternal and fetal mortality and morbidity in Indonesia. Placental and systemic oxidative stress caused endothelial cell dysfunction and injury. Placental oxidative stress also linked to fetal growth restriction. HSP70 is essential for cellular recovery, survival and maintenance of homeostasis. The purpose of this study was to compare the MDA, a marker for oxidative stress and HSP70 production in placental of severe preeclampsia, mild preeclampsia and normotensive pregnant women. Placenta were collected after delivery from normotensive pregnancies (N=10), severe preeclampsia (N=10) and mild preeclampsia (N=10). Placenta was cultured in RPMI and 20% FBS, and supernatant were collected in day 3. MDA was measured using spectrophotometer and absorbance read in 530nm. HSP70 was measured using enzyme-linked immunosorbent assay. The mean MDA concentration did not differ significantly between patients with severe preeclampsia (7.13+5.36 nmol/ml) and mild preeclampsia (4.82+2.47 nmol/ml) when compared with normotensive pregnancies (4.57+2.4 nmol/ml). The mean HSP70 concentration in mild preeclampsia is highest (10.15+12.39 nmo/ml) when compared with severe preeclampsia (3.78 +3.07 nmol/ml) and normotensive pregnant women (3.76+4.65nmol/ml), but the difference was not significant. Although the difference was not significant, is indicates homeostasis response in mild preclampsia women is relative good. This response was abated in severe preeclampstic women. Although MDA and HSP70 concentration did not differ significantly between groups, however the high HSP70 concentration is indicates homeostasis response relatively good in mild preeclamptic women.

Anak usia remaja sangat rentan mengalami gangguan, diantaranya pola hidup yang tidak teratur, behavior, pertumbuhan fisik dan psikis. Peranan aktifitas fisik atau olahraga menjadi sarana untuk mencegah anak usia remaja dari pengaruh negatif. Akan tetapi aktivitas olahraga ini tidak dilakukan secara teratur dan durasinya sangat singkat baik dilingkungan sekolah maupun dilingkungan sekitarnya. Olahraga disekolah cenderung lebih singkat. Saat ini, banyak anak usia remaja melakukan kegiatan olahraga permainan Futsal. Olahraga futsal bermanfaat untuk meningkatkan kebugaran jasmani, neuroplastisitas. Akan tetapi olahraga futsal juga dapat memengaruhi terhadap stres fisik dan stres oksidatif. Desain pre-post test pada anak usia remaja 13-15 tahun. Subjek penelitian 27 orang (tidak biasa bermain futsal) dibagi secara acak kedalam 3 kelompok perlakuan, frekuensi olahraga (FO) 1x, 3x dan 5x perminggu. Tahap awal penelitian persiapan olahraga futsal dan pengambilan data parameter, tahap perlakuan olahraga futsal selama 8 minggu dan tahap terakhir pengambilan data paramater dilakukan 1 hari setelah perlakuan. Penelitian ini sudah lolos kaji etik dari Komisi Etik FKUI. Kebugaran jasmani ditemukan meningkat secara bermakna pada FO3 dan FO5. Atensi dan memori ditemukan peningkatan pada FO1, FO3 dan FO5. Peningkatan BDNF hanya ditemukan pada kelompok FO3. IGF-1 ditemukan meningkat secara bermakna pada kelompok FO1, FO3 dan FO5. Peningkatan kadar Kortisol dan MDA hanya ditemukan pada kelompok FO5. Hasil penelitian ditemukan olahraga yang optimal dan aman pada anak usia remaja yaitu frekuensi olahraga futsal 3 kali perminggu dibandingkan olahraga futsal 1 kali dan 5 kali terhadap kebugaran jasmani, neuroplastisitas, stres fisik dan stres oksidatif pada anak usia remaja.


Adolescents are susceptible to disorders, including unconventional lifestyle, behavior, physical and psychological growth. The role of physical activity or sports becomes a means to prevent adolescent children from negative influences. However, this sporting activity is not taken out regularly, and the duration is very short both in the school and the enclosing circumstances. Sports at school tend to be shorter. At present, many teenagers do Futsal sports. Futsal exercise is useful for improving physical fitness, neuroplasticity. However, futsal can also affect physical stress and oxidative stress-pre-post test design in adolescents aged 13-15 years. The research subjects were 27 people (unusual for playing futsal) divided randomly into three treatment groups, exercise frequency (FO) 1x, 3x, and 5x per week. The initial stage of the research is preparation for futsal and parameter data collection, the stage of treatment for futsal for eight weeks, and the last stage of data taking post parameters, one day after treatment. This research has passed the ethical review of the FKUI Ethics Commission. Physical fitness normally found to increase significantly at FO3 and FO5. Attention and memory are found to increase in FO1, FO3, and FO5. Surprisingly, BDNF was only detected in only the FO3 group. IGF-1 was found to increase significantly in the FO1, FO3, and FO5 groups. Increased levels of Cortisol and MDA was simply observed in the FO5 group. The results of the study found optimal and safe exercise in adolescents, namely the frequency of futsal exercise 3 times a week compared to futsal one times and five times for physical fitness, neuroplasticity, physical stress and oxidative stress in adolescents.

Renal dysfunction which frequently occurs in type 2 diabetes mellitus patients caused by oxidative stress. The effectiveness of the type 2 diabetes mellitus treatment to renal dysfunction is unknown. This study compare and analyze the correlation between urinary hydrogen peroxide which is a product of oxidative stress and estimated glomerular filtration rate (eGFR) in the treatment groups of sulfonylurea and combination biguanide-sulfonylurea. This study used a retrospective cohort study design with 50 sampels that was taken in Dr. Sitanala Tangerang hospital with total sampling technique. Estimated GFR value obtained based on serum creatinine values were measured using a kinetic Jaffe method, while the urinary hydrogen peroxide using FOX 1 (Ferrous ion Oxidation Xylenol Orange1). Value of urinary hydrogen peroxide in the two treatment groups did not have significant difference (p = 0.69), while the eGFR value of two groups did not have significant differences with the Cockroft Gault is p = 0.884; MDRD p = 0.886; and CKDEPI p = 0.490. Correlation analysis of urinary hydrogen peroxide and eGFR based on the MDRD equation and CKDEPI generate significant positive correlation (r = 0.326; p = 0.021) and (r = 0.282; p = 0.047). There is no antioxidant activity in the treatment groups. Urinary hydrogen peroxide may play a role in the pathophysiologic significance of diabetic nephropathy.

Penelitian ini membahas tingkat karbonil sebagai penanda dari stress oksidatif di ginjal akibat terpapar oleh hipoksia hipobarik akut interminten. Hipoksia hipobarik rentan terjadi kepada penerbang (pilot) yang sering terpapar oleh kondisi ini. Sebagai salah satu organ penting, ginjal rentan terpapar oleh stress oksidatif akibat hipoksia hipobarik. Penelitian ini menggunakan desain eksperimental. Sampel jaringan yang dipakai adalah jaringan ginjal tikus jantan galur wistar. Sampel ini lalu dikelompokkan ke dalam empat perlakuan dengan perbedaan frekuensi paparan hipoksia hipobarik dari hypoxia chamber dan satu kelompok kontrol. Metode Cayman?s Protein Carbonyl Assay yang telah dimodifikasi oleh departemen biokimia Fakultas Kedokteran Universitas Indonesia digunakan dalam percobaan ini. Hasil penelitian menunjukkan adanya tingkat perbedaan karbonil yang bermakna antara kelompok perlakuan dan kelompok kontrol (p<0.05). Bedasarkan hasil pada penilitian ini, dapat disimpulkan terdapat peningkatan stres oksidatif secara signifikan pada keadaan hipoksia hipobarik akut intermiten di jaringan ginjal tikus.
This study discussed about carbonyl concentration level as marker of stress oxidative in kidney due to acute intermittent hypobaric hypoxia exposure. Hypobaric hypoxia is prone to occur in aviators (pilots) who usually expose to this condition. As one of important organs, kidney is prone to be exposed by stress oxidative due to hypobaric hypoxia. This study uses experimental design. The sample used in this study was kidney tissue from male rats wistar. This sample then grouped into four different exposed groups which is differed in frequency of hypobaric hypoxia given in hypoxic chamber and a control group. The method used to measure carbonyl concentration was the method from Cayman's Protein Carbonyl Assay Procedure which then modified by biochemistry department Universitas Indonesia. The result from this experiment revealed that there was a significant difference of carbonyl concentration between exposed and control group (p<0.05). This study concluded that there was a significant increase of stress oxidative in acute intermittent hypobaric hypoxia condition in kidney tissue.

[ABSTRAK Gangguan fungsi ginjal yang sering terjadi pada pasien diabetes melitus tipe 2 diperankan oleh stres oksidatif. Belum diketahui efektivitas pengobatan diabetes melitus tipe 2 terhadap gangguan fungsi ginjal. Penelitian ini membandingkan dan menganalisis hubungan hidrogen peroksida urin yang merupakan produk stress oksidatif dan estimasi Laju Filtrasi Glomerulus (eLFG) pada kelompok pengobatan sulfonilurea dan kombinasi biguanid-sulfonilurea. Penelitian ini menggunakan desain penelitian kohort retrospektif dengan jumlah sampel 50 orang yang diambil di RSK Dr. Sitanala Tangerang dengan teknik total sampling. Nilai eLFG diperoleh berdasarkan nilai kreatinin serum yang diukur menggunakan metode kinetik Jaffe, sedangkan hidrogen peroksida urin menggunakan metode FOX (Ferrous ion Oxidation Xylenol Orange) 1. Nilai hidrogen peroksida urin pada dua kelompok pengobatan tidak memiliki perbedaan yang bermakna (p = 0,69). Sedangkan nilai eLFG pada dua kelompok juga tidak memiliki memiliki perbedaan yang bermakna dengan Cockroft Gault adalah p = 0,884; MDRD p = 0,886; dan CKDEP p= 0,490. Analisis hubungan hidrogen peroksida urin dengan eLFG berdasarkan persamaan MDRD dan CKDEPI menghasilkan hubungan positif bermakna (r = 0,326; p = 0,021) dan (r = 0,282; p = 0,047).
