Kitosan dan deksametason merupakan material yang digunakan dalam rekayasa jaringan tulang. Kitosan biasa digunakan sebagai scaffold, sedangkan deksametason sering digunakan sebagai sinyal. Salah satu penanda diferensiasi osteoblas adalah Alkaline phosphatase (ALP). Penelitian ini bertujuan menganalisis efek kitosan dibandingkan deksametason dalam menginduksi diferensiasi osteoblas melalui aktivitas ALP pada sel punca pulpa gigi (SPPG). Aktivitas ALP dianalisis dengan ALP assay. Terdapat 4 perlakuan yang memiliki aktivitas ALP diatas kontrol, yakni kitosan 5 ng/ml, deksametason 10 nM dan 100 nM, serta campuran kitosan 5 ng/ml dan deksametason 10 nM. Peningkatan konsentrasi deksametason meningkatkan aktivitas ALP. Peningkatan konsentrasi kitosan menurunkan aktivitas ALP. ......Chitosan and dexamethasone are materials that can be used in bone tissue engineering. Chitosan is often used as a scaffold, while dexamethasone is often used as signal. One of the markers of osteoblast differentiation is Alkaline phosphatase (ALP). This research objective is to analyse the effects of chitosan compared to dexamethasone in inducing osteoblast differentiation through ALP activity in Dental Pulp Stem Cells (DPSCs). ALP activity determined by ALP Assay. There were four treatments that have higher activity than the control, they are chitosan 5 ng/ml, 10 nM dexamethasone and 100 nM, and mixture of chitosan 5 ng/ml and 10 nM dexamethasone. The increased concentrations of dexamethasone increases ALP activity and the higher concentration of chitosan will decrease the ALP activity.

Kitosan telah diketahui sebagai material scaffold poten pada rekayasa jaringan tulang. Penelitian ini bertujuan menganalisis potensi lain kitosan sebagai induktor diferensiasi sel punca pulpa gigi (SPPG) menjadi osteoblas dibandingkan dengan deksametason yang telah umum digunakan, melalui ekspresi mRNA Col1A1. SPPG dikultur pada medium yang mengandung kitosan, deksametason dan kombinasi keduanya selama 7 hari. Ekspresi mRNA Col1A1 diuji dengan metode comparative CT real-time PCR. Medium mineralizing yang ditambahkan 5 ng/ml kitosan dapat meningkatkan ekspresi mRNA Col1A1 pada SPPG dibandingkan dengan kontrol dan kelompok perlakuan lainnya (p<0,05). Kitosan dapat menginduksi diferensiasi tahap awal SPPG menjadi osteoblas bergantung pada konsentrasi yang diberikan.
Chitosan has been proven as potential scaffold in bone tissue engineering. This research intends to analyze the other potency of chitosan as inductor in dental pulp stem cell (DPSC) differentiation into osteoblast compared to dexamethasone which is commonly used, by Col1A1 mRNA expression. DPSC were cultured in medium loaded with chitosan, dexamethasone and combination of both for 7 days. Col1A1 mRNA expression was analyzed by comparative CT method real time PCR. Mineralizing medium loaded with 5 ng/ml chitosan could enhance Col1A1 mRNA expression compared to control and another treated group on p<0,05. Chitosan could stimulate early stage of osteoblast differentiation. The effect was dose-dependent.

Ekstrak daun sukun memiliki efek antibakteri. S. sanguinis diketahui sebagai bakteri yang berperan pada pembentukkan awal plak. Penelitian ini bertujuan untuk menganalisis potensi antibakteri ekstrak daun sukun terhadap viabilitas biofilm S. sanguinis secara in vitro. Bakteri S. sanguinis ATCC 10556 dikultur pada 96-well plate dan diinkubasi 370C selama 20 jam (fase akumulasi) dan 24 jam (fase maturasi) kemudian dipaparkan ekstrak daun sukun dengan konsentrasi 5; 10; 15; 20; 40; 80; dan 100%. Viabilitas biofilm S. sanguinis diuji menggunakan MTT assay dengan panjang gelombang 490 nm. Hasil dianalisis dengan one-way ANOVA. Hasil penelitian ini menunjukkan bahwa terdapat penurunan bermakna viabilitas S. sanguinis setelah pemaparan dengan ekstrak daun sukun pelbagai konsentrasi pada fase akumulasi dan fase maturasi dibandingkan dengan kelompok kontrol (p<0.05). Viabilitas S. sanguinis setelah pemaparan ekstrak daun sukun konsentrasi 20; 80; dan 100% pada fase akumulasi lebih rendah dibandingkan fase maturasi.
Breadfruit leaf?s extract has a function as antibacterial. S. sanguinis is known as an early agent of formation of bacterial plaque.This research had purpose to analyze the antibacterial effect of breadfruit leaf?s extract against S. sanguinis growth. S. sanguinis ATCC 10556 were cultured in 96-well plate and incubated for 20 hours (accumulation phase) at 37 and 24 hours (maturation phase) then added breadfruit leaf?s extract concentrations 5; 10; 20; 40; 80; and 100%. Viability test was using MTT assay with wavelength of 490 nm. The results were analyzed by one-way ANOVA. The results showed that the viability of S. sanguinis after breadfruit leaf?s extract exposured in all concentrations on accumulation and maturation phase was lower than the control group (p<0.05). The viability of S. sanguinis after added breadfruit leaf?s extract concentration 20; 80; and 100% on accumulation phase was lower than maturation phase.

Daun Sukun memiliki efek antibakteri. S.mutans berperan dalam pembentukan biofilm. Penelitian ini bertujuan menganalisis potensi ekstrak daun Sukun terhadap viabilitas S.mutans secara in vitro. S.mutans ATCC 25175 dikultur, diinkubasi 20 jam (fase akumulasi) dan 24 jam (fase maturasi), kemudian dipaparkan ekstrak Daun Sukun 5;10;20;40;80;100%. Uji viabilitas menggunakan MTT assay, dianalisis dengan one-way ANOVA. Terdapat penurunan bermakna viabilitas S.mutans fase 20 jam setelah pemaparan ekstrak daun Sukun pada semua konsentrasi, dan fase 24 jam hanya konsentrasi 5% terjadi peningkatan bermakna dibandingkan kelompok kontrol (p<0.05). Viabilitas biofilm S.mutans pada kelompok perlakuan fase 20 jam lebih rendah dibandingkan fase 24 jam. ......Breadfruit leaf's has an antibacterial property. S.mutans has a function to form biofilm. The purpose of this study was to analyze the potential of breadfruit leaf’s extract toward the viability of S.mutans in vitro. S.mutans ATCC 25175 were cultured and incubated for 20 hours (accumulation phase) and 24 hours (maturation phase). Then added with the following breadlfruit leaf’s concentration :5;10;20;40;80;100%. The viability test was using MTT assay and would be analyzed by one-way ANOVA. There was significant decreased of the viability biofilm S.mutans on 20 hours phase after had been added the various concentration of bread fruit leaf’s extract, and on 24 hours phase only concentration 5% viability had increased significantly compared to the control group (p <0.05). The Viability biofilm S.mutans on 20 hours phase for all treatment group was lower than 24 hours phase.

Porphyromonas gingivalis dalam plak gigi merupakan penyebab utama terjadinya penyakit periodontal. Aloe vera memiliki sifat antibakteri karena kandungan senyawa fenol. Tujuan penelitian ini untuk menguji efek antibakteri kulit lidah buaya terhadap Porphyromonas gingivalis dengan metode ekstraksi terpilih. Dilakukan ekstraksi dengan metode maserasi dan infundasi untuk menarik senyawa aktif antibakteri. Uji antibakteri terhadap Porphyromonas gingivalis dilakukan dengan metode dilusi (KHM dan KBM) dan metode difusi (zona hambatan). Hasil metode dilusi menunjukkan nilai KHM dan KBM pada konsentrasi 70%. Sedangkan, metode difusi menunjukkan zona hambat tertinggi sebesar 2.25 mm pada konsentrasi 80%. Dapat disimpulkan bahwa infusum kulit Aloe vera mengandung senyawa fenol, tanin, dan antrakuinon serta memiliki efek bakteriostatik dan bakterisidal terhadap Porphyromonas gingivalis secara in vitro. Porphyromonas gingivalis in dental plaque has been the primary cause of periodontal disease. Aloe vera has antibacterial properties because of its active compounds such as phenol. The aim of this study was to examine antibacterial effects of Aloe vera rind on Porphyromonas gingivalis using the chosen method. It was performed by doing xtraction with maceration and infusion methods to attract antibacterial active compounds. Antibacterial tests on Porphyromonas gingivalis were carried out using dilution (MIC and MBC)) and diffusion (inhibition zone) methods. The result of the dilution method showed MIC and MBC values at 70% concentration while the diffusion method showed the highest inhibition zone of 2.25 mm at 80% concentration. Hence, Aloe vera rind infuse revealed the presence of phenol, tannin, and anthraquinon and along with bacteriostatic and bacterisidal effects on Porphyromonas gingivalis, in vitro.

Kesimpulan, infusum lidah buaya mengandung senyawa aktif fenol, tanin dan antrakuinon serta memiliki sifat bakteriostatik dan tidak bersifat bakterisidal terhadap Porphyromonas gingivalis secara in vitro.
Porphyromonas gingivalis is the main etiologic agent of periodontal disease. Aloe vera has antibacterial effect because of its phenolic compound. The aim of this study is to investigate the antibacterial effectivity of Aloe vera on Porphyromonas gingivalis. The study was performed by extracting Aloe vera using maceration and infusion extraction methods in order to attract the antibacterial active compounds. The test method of the antibacterial effect was carried out by dilution method (MIC and MBC) and diffusion method (inhibition zone). The results of dilution method showed that MIC value was at 70% concentration while MBC value could not be determined. The largest inhibition zone of the diffusion method was 1,75 mm at 90% concentration. In summary, Aloe vera infuse contained antibacterial active compounds such as phenol, tannin and anthraquinone and showed bacteriostatic effect on Porphyromonas gingivalis, in vitro.

Industri farmasi sebagai produsen produk obat memiliki peran besar dalam peningkatan upaya kesehatan yaitu melalui pengadaan obat-obatan. Industri farmasi memproduksi obat dengan berpedoman pada cara pembuatan obat yang baik (CPOB). Industri farmasi harus memiliki minimal tiga orang apoteker sebagai penanggungjawab masing-masing pada bidang pemastian mutu, produksi, dan pengawasan mutu. Tujuan praktik kerja profesi (PKP) ini supaya mahasiswa apoteker memiliki wawasan, pengetahuan, keterampilan, dan pengalaman praktis untuk melakukan pekerjaan kefarmasian di industri farmasi dan paham penerapan CPOB. Mahasiswa apoteker di tempatkan di bagian quality assurance (QA) dan melakukan validasi metode pembersihan. Dari PKP yang dilakukan, mahasiswa PKP mendapat pengetahuan pekerjaan kefarmasian di industri farmasi, memperoleh keterampilan dalam melakukan validasi pembersihan alat, dan lebih memahami penerapan aspek-aspek CPOB di industri farmasi. Industri farmasi merupakan salah satu sarana tempat dilakukan pekerjaan kefarmasian. Praktik kerja profesi yang dilaksanakan di PT Medifarma Laboratories ini, bertujuan agar mahasiswa mendapatkan pengalaman nyata dalam melakukan pekerjaan kefarmasian di industri farmasi. Industri farmasi adalah badan usaha yang memiliki izin untuk melakukan kegiatan pembuatan obat atau bahan obat. Dalam pembuatan obat, industri farmasi harus memenuhi ketentuan Cara Pembuatan Obat yang Baik CPOB agar obat yang dihasilkan aman, berkhasiat dan bermutu. Seluruh aspek CPOB telah diterapkan oleh PT Medifarma Laboratories. Selain itu, PT Medifarma Laboratories juga menerapkan ketentuan GMP dari PIC/s sebagai jaminan tambahan untk keperluan ekspor produk. Seluruh proses produksi dan aspek terkait telah dikendalikan, dan setiap tahapnya telah terdokumentasikan. Salah satu upaya pengendalian dalam proses produksi adalah validasi proses yang menjadi tugas khusus yang diberikan kepada mahasiswa. Validasi proses merupakan usaha untuk membangun bukti terdokumentasi bahwa suatu proses produksi akan senantiasa menghasilkan produk yang memenuhi kriteria yang ditentukan. Validasi ini termasuk dalam validasi konkuren, dilakukan terhadap pada tiga bets tablet suplemen XYZ berturut-turut, dimana tiap bets berukuran 100.000 tablet. Validasi proses telah dilakukan mulai dari tahap wet compounding, dry mixing, compression dan coating. Validasi terhadap proses blistering tablet suplemen XYZ juga telah dilakukan. Dari hasil validasi, disimpulkan bahwa proses produksi tiga bets tablet suplemen XYZ telah tervalidasi.
ABSTRACT
The pharmaceutical industry is one of the places to do the pharmaceutical works. The internship that held at PT Medifarma Laboratories, intended to make students get the real experience in performing the pharmaceutical works in the pharmaceutical industry. The pharmaceutical industry is an entity that has a license to conduct the manufacture of drugs or drug ingredients. In the manufacture of drugs, the pharmaceutical industry must comply Good Manufacturing Practice GMP so that the resulting drug meet the criteria of safe, efficacy and quality. All aspects of GMP has been applied by PT Medifarma Laboratories. In addition, PT Medifarma Laboratories also applying the GMP provisions of PIC s as an additional guarantee for export products. The whole process of production and its related aspects are controlled, and every stage is documented. One of the actions in order to control the production process is validation process that became task given to the student. The validation process is establishing documented evidence to determine that process consistently produce a product meet its predetermined specifications and quality characteristics. Validation that had been done is classified as concurrent validation, and were carried out on three batches of Tablet Supplements XYZ, where the size of each batch were 100.000 tablets. Validation process were started from wet compounding, dry mixing, compression, and coating. Validation of the blistering process Tablets Supplement XYZ had also been performed. The results showed that the production process of three batches of Tablet Supplements XYZ had been validated.
