Telah dilakukan uji aktivitas antioksidan terhadap isolat kapang endofit dari Garcinia forbesii King dan Garcinia porrecta Wall. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan isolat hasil fermentasinya serta mendapatkan pola kromatogram KLT isolat yang memiliki aktivitas antioksidan. Uji aktivitas antioksidan dilakukan menggunakan metode DPPH (1,1-difenil-2-pikrilhidrazil). Dari uji pendahuluan, didapat 3 isolat yang menunjukkan aktivitas antioksidan. Hasil pengujian dengan spektrofotometri UV-Vis menunjukkan isolat DP 1 memiliki aktivitas antioksidan tertinggi dengan nilai IC50 sebesar 123,97 μg/mL, sedangkan IC50 vitamin C adalah 4,8 μg/mL. Penentuan pola kromatogram KLT dilakukan terhadap dua isolat yang memiliki nilai IC50 terkuat yaitu isolat DP 1 dihasilkan dengan fase gerak terpilih n-heksana-aseton (3:2) dan isolat DF 1 dihasilkan dengan fase gerak terpilih aseton-etil asetat (3:2). Setelah elusi, terlihat delapan bercak pada isolat DP 1 dan satu bercak pada isolat DF 1. Setelah disemprot DPPH 0,2% dalam metanol, pada isolat DF 1 terlihat bercak kuning pucat berlatar belakang ungu, sedangkan pada isolat DP 1 tidak terlihat jelas. Berdasarkan hasil penelitian yang telah dilakukan menunjukkan bahwa kapang endofit dari daun, akar Garcinia forbesii King dan daun Garcinia porrecta Wall memiliki aktivitas antioksidan. A study has been done to test an antioxidant activity of endophytic fungi isolates from Garcinia forbesii King and Garcinia porrecta Wall. This study was aimed to investigate the antioxidant activity of extracts produced by fermentation of isolates and to obtain those TLC chromatogram isolates which possessed antioxidant activity. The assay for antioxidant activity was based on the DPPH (1,1-diphenyl-2-picrylhydrazil) method. Initial analysis showed that 3 isolates had significant antioxidant activities. An observation used spectrophotometry UV-Vis showed that DP 1 isolate had the highest antioxidant activity with an IC50 value of 123.97 μg/mL, while IC50 vitamin C was 4.8 μg/mL. The chromatogram profile determination was done towards two isolates that had the strongest IC50 value, DP 1 isolate which was developed using n-hexane-acetone (3:2) as the selected mobile phase and DF 1 isolate which was developed using the selected mobile phase of acetone-ethyl acetate (3:2). After elusion, there were seen eight spots on DP 1 isolate and one spot on DF 1 isolate. DF 1 isolate displayed the presence of antioxidant activity after spraying with methanolic solution of 0.2% DPPH which was a pale yellow spot with purple background, while it wasn?t seen clearly on DP 1 isolate. This study has shown that endophytic fungi from the root, the leaf of Garcinia forbesii King and the leaf Garcinia porrecta Wall have antioxidant activities. |
