[Lab-on-chip merupakan teknologi baru dengan konsep miniaturisasi pekerjaanlaboratorium ke dalam sebuah chip. Dengan merekayasa material yang ramah sel,lab-on-chip ini diharapkan mampu menjadi media studi sel. Lab-on-chip terbuatdari polydimethyl siloxane (PDMS) berpermukaan kasar (groovy) hasil daribiomachining. Lab-on-chip terdiri dari inlet, empat saluran utama dengan duasaluran berpermukaan kasar (groovy) dan dua sisanya tanpa modifikasi, danoutlet. Sebelum mendesain lab-on-chip, bahan lab-on-chip diuji kekasaranpermukaan, wettability, dan viabilitas sel pada permukaannya. Langkahselanjutnya adalah membuat desain cetakan lab-on-chip komponen atas dankomponen bawah dengan Autodesk Inventor 2012. Selanjutnya, cetakan dibuatdengan CNC milling. Lalu, cetakan di-biomachining selama 24 jam untukmembentuk kontur kasar (groovy). Kemudian, PDMS & agent dituang padacetakan tersebut. Komponen atas dan komponen bawah dirakit dengan bantuanakrilik. Candida albicans kemudian dikultur pada permukaan saluran utamadengan bantuan Fetal Bovine Serum (FBS). Setelah diinkubasi 24 jam, viabilitassel diukur menggunakan ELISA Reader. Hasil menunjukkan bahwa kekasarandan coating reagen berpengaruh pada viabilitas sel. Jadi, untuk mendapatkan LoCyang berpotensi memberikan viabilitas sel paling baik, modifikasi kekasaran dancoating reagen perlu dilakukan., Lab-on-chip is a new technology with concept of miniaturization of laboratorywork into a chip. Lab-on-chip is made of polydimethyl siloxane(PDMS) with rough (groovy) surface as the result of biomachining. Lab-on-chipconsists of inlet, the four main channels with two rough surface (groovy) channelsand the remaining two without modification, and the outlet. Before designing thelab-on-chip, lab-on-chip material is tested in terms of surface roughness,wettability, and on-surface cell viability. The next step is designing mold of upperand lower lab-on-chip components with Autodesk Inventor 2012. Subsequently,the mold is made by CNC milling. Then, the molds are biomachined for 24 hoursto form a rough (groovy) surface. Then, PDMS and agent are poured on the mold.Upper and lower component are assembled with acrylic support. Candidaalbicans is then cultured on the surface of the main channel with the help of FetalBovine Serum (FBS). After 24 hours incubation, cell viability was measuredusing ELISA Reader. The result shows that surface roughness and reagent coatinginfluence cell viability. So, to get potential lab-on-chip with good cell viability,roughness modification and reagent coating are needed to be proceeded.] |
