Abstract
Pembatasan penggunaan daun tembakau kering untuk produksi rokok harus disertai dengan adanya pengembangan produk alternatif non-rokok yang berbahan dasar daun tembakau. Salah satu alternatif yang dapat dilakukan adalah dengan memanfaatkan kandungan senyawa nikotin yang telah diisolasi pada daun tembakau. Nikotin diisolasi dari ekstrak daun tembakau menggunakan metode kromatografi kolom dengan variasi rasio campuran petroleum eter dan etanol sebagai fasa gerak, dimulai dari 8:2, 6:4, 4:6, 2:8, hingga 0:10. Fraksi hasil dari setiap rasio yang berhasil didapatkan, kemudian diuji secara kualitatif dengan menggunakan kromatografi lapis tipis KLT serta secara kuantitatif dengan instrumen HPLC. Proses kromatografi yang dilakukan mampu mengisolasi 4,006 senyawa nikotin dari nilai awal sebesar 4,19 . Didapatkan pula bahwa senyawa etanol baik untuk digunakan dalam mengisolasi nikotin yang terdapat dalam ekstrak daun tembakau.
Restrictions on the use of dried tobacco leaf for cigarette production must be accompanied by the development of non cigarette alternative products that are made from tobacco leaves. One of the alternatives that can be done is to use the nicotine compound that is isolated from tobacco leaf extract. Nicotine is isolated using column chromatography method with the variation of mobile phase mixture petroleum ether and ethanol, started from 8 2, 6 4, 4 6, 2 8, to 0 10. All of the chromatographic fraction from each mobile phase rsquo s ratio is then tested qualitatively using thin layer chromatography TLC and also quantitatively using HPLC instrument. The column chromatography process can isolate 4.006 of nicotine compound from 4.19 tobacco leaf extract rsquo s nicotine. It is also known that ethanol is good to be used as chromatography rsquo s mobile phase for nicotine isolation.
