Penurunan fungsi memori merupakan salah satu karakteristik dari proses penuaan yang dapat menurunkan kualitas hidup seseorang. Hipokampus merupakan bagian otak yang paling rentan mengalami perubahan seiring dengan proses penuaan yakni dengan adanya penurunan fungsi memori ditandai dengan adanya perubahan plastisitas sinaps. Plastisitas sinaps merupakan mekanisme selular yang mendasari proses pembentukan memori. Terdapat dua protein yang penting dalam plastisitas sinaps dan sering dijadikan marker plastisitas sinaps yakni Synaptophysin SYP dan Postsynaptic density-95 PSD-95 . Berbagai upaya telah dilakukan untuk mengatasi masalah penurunan fungsi memori, salah satunya melalui terapi herbal. Tanaman Centella asiatica memiliki kandungan triterpenoid dan flavonoid telah lama dikenal berperan dalam meningkatkan fungsi memori. Tujuan penelitian ini adalah untuk mengetahui pengaruh ekstrak etanol Centella asiatica CeA terhadap ekspresi protein SYP dan PSD-95 di hipokampus tikus. Penelitian ini merupakan studi eksperimental in-vivo menggunakan 18 ekor tikus Wistar jantan usia 6 bulan yang dibagi secara acak menjadi 3 kelompok: 1 kelompok kontrol K 2 kelompok CA300 dan 3 kelompok CA600. Kelompok kontrol diberikan akuades, kelompok CA300 diberikan CeA dosis 300 mg/kg.BB dan kelompok CA600 diberikan CeA dosis 600 mg/kg.BB yang dilakukan selama 28 hari berturut-turut secara oral. Setelah 28 hari, tikus didekapitasi dan hipokampus diisolasi dari jaringan otak. Ekspresi protein SYP dan PSD-95 di jaringan hipokampus dianalisis menggunakan teknik imunohistokimia pada regio CA1 hipokampus. Hasil penelitian menunjukkan, pemberian ekstrak etanol Centella asiatica dosis 600 mg/kg.BB dapat meningkatkan ekspresi protein SYP dan PSD-95 secara signifikan. Decreased memory function is one of the characteristics of the aging process that can reduce the quality of life. Hippocampus is the most vulnerable part of the brain undergoing changes along with the aging process that is with the decline in memory function characterized by the change in synaptic plasticity. Synaptic plasticity is the cellular mechanism that underlies the process of memory formation. There are two important proteins in synaptic plasticity and are often used as synaptic plasticity markers Synaptophysin SYP and Postsynaptic density 95 PSD 95 . Various efforts have been made to overcome the problem of memory function decline, one of them through herbal therapy. Centella asiatica CeA plants contain triterpenoids and flavonoids have long been known to play a role in improving memory function. The purpose of this study was to investigate the effect of Centella asiatica ethanol extract CeA on expression of SYP and PSD 95 protein in rats hippocampus. The study was an in vivo experimental study using 18 male Wistar rats aged 6 months randomly divided into 3 groups 1 control group K 2 CA300 group and 3 CA600 group. The control group was given aquadest, a group of CA300 given a 300 mg kg.BW CeA and a CA600 group administered a 600 mg kg.BW CeA administered for 28 consecutive days orally. After 28 days, rat were decapitated and the hippocampus were isolated from brain. The expression of the SYP and PSD 95 proteins in the hippocampal tissue was analyzed using immunohistochemical techniques in the hippocampal CA1 region. The results showed, giving Centella asiatica ethanol extract with dose 600mg kg.BW can increase expression of SYP protein and PSD 95 significantly. |
