ABSTRAK Fosfor (P) adalah salah satu nutrien utama penyebab eutrofikasi di perairan yangdapat menimbulkan terjadinya blooming alga. Eutrofikasi dapat terjadi karena prosespelepasan senyawa fosfat dari sedimen yang dapat meningkatkan konsentrasi fosfatdalam air. Tujuan dari penelitian ini adalah untuk mengkaji interaksi antara sedimen danair pada proses pelepasan fosfat dan penyerapan oleh alga Oscillatoria sp dengan teknikdiffusive gradient in thin film (DGT). Teknik DGT menggunakan ferrihidrit sebagaibinding gel untuk penentuan fosfat dengan efisiensi penyerapan sebesar 98,17% padainkubasi selama 24 jam. Respon unit DGT terhadap akumulasi fosfat pada rentangkonsentrasi 0-1,0 mg/L memberikan korelasi linier (R2=0,9792). Teknik DGTmemberikan respon pada rentang pH 2 -10 untuk spesi fosfat H2PO4-, HPO42- dan PO43-.Penelitian ini menggunakan sedimen laut Teluk Jakarta pada 6 titik stasiun. Fraksinasifosfat dalam sedimen diperoleh fraksi H2O-P (0,27-0,76 μg/g), NH4Cl-P (0,54-2,27μg/g), NaHCO3-P (3,53-9,69 μg/g), NaOH-P (1,63-11,23 μg/g) dan HCl-P (1,20-2,21μg/g). Studi pelepasan fosfat dilakukan dengan variasi suhu, pH, agitasi, waktu kontak,salinitas dan kondisi oksigen. Suhu maksimum terjadinya pelepasan fosfat yaitu 35oCsebesar 99, 88 μg/L dan massa fosfat terakumulasi unit DGT sebesar 0,9876 μg. RentangpH 5-10 menghasilkan konsentrasi fosfat yang lepas dari sedimen sebesar 59,33-100,16μg/L dan MDGT sebesar 1,8331-2,9734 μg. Secara umum pengadukan tidakmempengaruhi pelepasan fosfat dari sedimen. Pelepasan fosfat maksimum terjadi padasalinitas dengan konsentrasi NaCl 30 g/L dengan inkubasi selama 15 hari diperoleh Clarutansebesar 113,99 μg/L dan MDGT sebesar 4,7723 μg. Pengaruh kondisi aerasi menunjukkanpelepasan fosfat pada kondisi anoxic lebih besar dari kondisi oxic. Kondisi anoxicdengan inkubasi selama 21 hari menunjukkan pelepasan fosfat dari sedimen ke air sebesar208,62 μg/L dan MDGT sebesar 6,1081 μg. Bioavailabilitas fosfat terhadap mikroalgaOscillatoria sp dipengaruhi oleh waktu inkubasi. Semakin lama waktu inkubasi makasemakin banyak jumlah fosfat yang diserap. P-tersedia dalam medium dengan aerasi padakonsentrasi fosfat 10, 20, 30, 40 dan 50 ppm sebesar 5,74, 11,52, 18,14, 23,12 dan 26,48ppm. P-tersedia yang berasal dari sedimen diserap oleh alga Oscillatoria sp. Fraksi fosfatyang diserap tersebut menunjukkan fraksi NaOH-P ˃ NaHCO3-P ˃ H2O-P ˃ HCl-P ˃NH4Cl-P. Teknik DGT dapat digunakan untuk memprediksi biovailabilitas fosfatterhadap alga Oscillatoria sp. Hubungan linier antara Palga dengan PDGT pada salinitas 0g/L, 15 g/L dan 30 g/L diperoleh berturut-turut 0,9820 ; 0,9449 dan 0,9677. Pelacakanfosfat dengan isotop 32P menunjukkan terjadi penyerapan yang sangat cepat 32P oleh algaOscillatoria sp setelah inkubasi selama 1 jam dengan % incorporation sebesar 99 %. ABSTRACT Phosphorus (P) is one of the most nutrient contributors in aquatic eutrophicationand causes the algae blooms. Sediment plays an important role in the overall phosphatereleased. Phosphate can be released from sediment and can give effect on phosphateconcentration in overlying water. The objective of this research is to study the phosphatereleased from sediment and phosphate uptake in Oscillatoria sp algae using diffusivegradient in thin film (DGT) technique. DGT technique with ferryhidrite as binding gelshowed the adsorption efficiency as 98,17% with incubation for 24 hours. The DGTdevice gave the range of phosphate concentration was 0.2-1.0 mg/L with linearcorrelation (R2=0,9649). The DGT technique can be used in the range of pH 2 -10 for thespeciation of phosphate as H2PO4-, HPO42-, and PO43-. This study use the marine sedimentof Jakarta Bay. The research indicated the fractions of six stations were H2O-P (0.27-0.76μg/g), NH4Cl-P (0.54-2.27 μg/g), NaHCO3-P (3.53-9.69 μg/g), NaOH-P (1.63-11.23μg/g), and HCl-P (1.20-2.21 μg/g). The study of phosphate released from sedimentincluding the effect of temperature, pH, agitation, salinity and oxygen concentration onphosphate concentration in the overlying water. The temperature maximum of phosphatereleased at 35oC with the concentration of phosphate was 99.88 μg/L and theaccumulation of phosphate in DGT device was 0.9876 μg. The range of pH 5-10 resultedthe phosphate released was 59.33-100.16 μg/L and MDGT range 1.8331-2.9734 μg. Theagitation did not influence the phosphate released from sediment. Salinity with theconcentration of 0-35 g/L showed the maximum of phosphate released at concentrationof NaCl 30 g/L, for incubation of 15 days, Csolution was 113.99 μg/L andMDGT was 4.7723μg/L. The effect of aeration condition showed the phosphate released in anoxic conditionwas higher than the oxic condition. The anoxic condition with incubation of 21 daysshowed of phosphate released at 208.62 μg/L, and MDGT 6.1081 μg. Bioavailability ofphosphate in medium at phosphate concentrations of 10, 20, 30, 40, and 50 ppm were5.74, 11.52, 18.14, 23.12, and 26.48 ppm, respectively. The available P from sedimentwas uptake by Oscillatoria sp algae.The phosphate fraction in sediment which uptake itshown the fraction of NaOH-P ˃ NaHCO3-P ˃ H2O-P ˃ HCl-P ˃ NH4Cl-P. The DGTtechnique is applied to predict the bioavailability of phosphate for algae uptake(Oscillatoria sp). The linear correlation between Palga and PDGT at salinity of 0 g/L, 15g/L, and 30 g/L were 0.982, 0.9449, and 0.9677, respectively. The tracer of 32Pradioisotope showed the uptake of 32P in Oscillatoria sp microalgae for 1 hour and itrelated to the fast uptake of Oscillatoria sp for phosphate in the solution with %incorporation was 99 % |
