ABSTRAK
Metformin sekarang menjadi perhatian yang cukup besar karena telah dikaitkan dengan penurunan yang signifikan dalam trigliserida plasma, kolesterol total plasma, kolesterol LDL, dan peningkatan yang signifikan dalam kolesterol HDL plasma tanpa menyebabkan hipoglikemia. Penelitian ini bertujuan untuk mengetahui pengaruh metformin pada tikus hiperlipidemia yang diinduksi diet tinggi lemak. Dalam penelitian ini menggunakan 30 ekor tikus yang dikelompokkan menjadi 6 kelompok yaitu kelompok kontrol normal, positif, negatif, dan 3 kelompok perlakuan lainnya (n = 5). Kontrol normal tidak diberikan perlakuan, kelompok negatif diberi CMC 0,5%, kelompok positif menggunakan dosis simvastatin 1,8 mg / kg BB tikus / hari secara oral dan kelompok variasi dosis 45 mg / 200gBB tikus / hari secara oral; 90 mg / 200gBW tikus / hari secara oral; dan 180 mg / 200gBW tikus / hari secara oral. Induksi diberikan selama 28 hari dengan menggunakan diet tinggi lemak. Diet tinggi lemak diberikan melalui pakan dan usap lambung. Komposisi pakan tinggi lemak terdiri dari pakan standar: kuning telur puyuh: mentega (50:40: 10) dan minum PTU 0,01%. Komposisi induksi lambung sonde menggunakan kuning telur puyuh: sirup jagung fruktosa tinggi: mentega (80: 15: 5) dan PTU 0,1%. Kemudian dilanjutkan selama 14 hari pemberian obat oral. Pengukuran profil lipid dilakukan dengan metode enzymatic colori. Semua dosis metformin secara signifikan menurunkan kolesterol total dan LDL (p <0,05) dibandingkan dengan kontrol normal dan negatif. Metformin dosis 3 juga secara signifikan menurunkan kadar trigliserida (p <0,05) dan meningkatkan kadar HDL (p <0,05) dibandingkan dengan kontrol normal dan negatif. Berdasarkan hasil penelitian tersebut, metformin memiliki efek antihiperlipidemik dengan dosis 45 mg / 200 g BB.
ABSTRACT
Metformin is now of considerable concern because it has significant reductions in plasma triglycerides, total plasma cholesterol, LDL cholesterol, and significant increases in plasma HDL cholesterol without causing hypoglycemia. This study aims to determine the effect of metformin on hyperlipidemic rats induced by a high-fat diet. In this study, 30 rats were grouped into 6 groups, namely the normal, positive, negative control group and 3 other treatment groups (n = 5). Normal control was not given treatment, the negative group was given CMC 0.5%, the positive group used a dose of simvastatin 1.8 mg / kg of rats / day orally and the group with a variation of the dose of 45 mg / 200gBW rats / day orally; 90 mg / 200gBW rat / day orally; and 180 mg / 200gBW rat / day orally. All dose measurements significantly lowered total and LDL cholesterol (p <0.05) compared to normal and negative controls. Metformin dose 3 also significantly lowered triglyceride levels (p <0.05) and increased HDL levels (p <0.05) compared to normal and negative controls. Based on the results of these studies, metformin has an antihyperlipidemic effect at a dose of 45 mg / 200 g BW. |
