Abstract
Streptococcus macedonicus MBF10-2 adalah suatu galur bakteri asam laktat yang
bersifat GRAS (generally recognized as safe). S. macedonicus MBF10-2 telah
dikembangkan selama ini untuk pemanfaatan dalam bidang farmasi dan kesehatan, salah
satunya sebagai bahan aktif perawatan kesehatan kulit dalam bentuk lisat antibakteri.
Penggunaan lisat dalam produk perawatan kesehatan kulit beberapa dekade terakhir telah
terbukti efektif, misalnya untuk mengatasi masalah penuaan kulit. Parameter uji aktivitas
antioksidan dalam penuaan kulit akibat radikal bebas diuji cobakan dalam penelitian
terhadap S. macedonicus MBF10-2 yang berupa lisat hasil pelisisian sel maupun cell free
supernatant (CFS) sebagai pembanding. Tujuan dari penelitian ini adalah menguji
aktivitas antioksidan dan sensitivitas lisat serta CFS S. macedonicus MBF10-2 terhadap
kulit manusia. Lisat dan CFS didapatkan dari penelitian terdahulu yang selanjutnya
difraksinasi dan disimpan dalam bentuk kering beku. Uji antioksidan dilakukan dengan
metode DPPH berdasarkan nilai IC50 dan uji sensitivitas dilakukan dengan metode patch
test. Hasil percobaan dengan 3 kali pengulangan menunjukkan bahwa metode fraksinasi
dan pengeringan beku bersifat reproducible. Nilai IC50 lisat sebesar 840 ?g/ml
menunjukkan aktivitas antioksidan sangat lemah, sementara nilai IC50 fraksi-fraksi CFS
sebesar 16.050 ?g/ml; 11.945 ?g/ml; dan 13.488 ?g/ml berarti tidak ada aktivitas
antioksidan. Uji sensitivitas kulit memberikan hasil negatif dimana fraksi lisat dan CFS
tidak menyebabkan alergi dan iritasi sehingga aman terhadap kulit manusia.

group that is generally recognized as safe (GRAS). S. macedonicus MBF10-2 has been
developed for pharmaceutical and healthcare benefits, one of which is an active ingredient
for skin care in the form of antibacterial lysate. In the last decade, application of lysate as
skin care products has proven to be effective to overcome premature skin aging. The
parameter of antioxidant in skin aging due to free radicals were tested in a study of S.
macedonicus MBF10-2 lysate in comparison with cell free supernatant (CFS). The
purpose of this study is to examine the antioxidant activity and the sensitivity of S.
macedonicus MBF10-2 lysate and CFS on human skin. The lysate and CFS are obtained
from previous study which are then fractionated and stored in freeze dried form.
Antioxidant assay is carried out by DPPH method based on the IC50 value and skin
sensitivity test is demonstrated by patch test. The results show that the fractionation and
freeze dry method are reproducible which was carried out in triplicates. The IC50 value of
the lysate is 840 ?g/ml indicating the lysate is a very weak antioxidant, meanwhile the
IC50 value of the CFS fractions are 16.050 ?g/ml; 11.945 ?g/ml; and 13.488 ?g/ml
showing that the CFS fractions do not possess any antioxidant activity. Skin sensitivity
test resulted negative, which proves that each fraction of lysate and CFS is non-allergenic
and non-irritating therefore it is safe for the human skin.
