Penggunaan hidrogel berbasis biopolimer sudah banyak diteliti untuk pembalut luka karena efektif dalam membunuh bakteri. Pada penelitian ini, telah dilakukan sintesis terhadap biopolimer natrium alginat (NaAlg) diperkuat dengan biopolimer polivinil alkohol (PVA) dengan modifikasi nanopartikel perak yang didoping seng oksida (Ag doping ZnO NP) sebagai agen antibakteri. Keberhasilan sintesis hidrogel nanokomposit NaAlg-PVA/Ag doping ZnO dikarakterisasi dengan FTIR, XRD, SEM, AAS dan TGA. Morfologi hidrogel nanokomposit NaAlg-PVA/Ag doping ZnO menggunakan SEM diperoleh struktur berpori tidak beraturan dengan permukaan ditutupi nanopartikel Ag doping ZnO. Hidrogel nanokomposit NaAlg- PVA/Ag doping ZnO dengan variasi biopolimer 1:1 dan konsentrasi Ag doping ZnO 2,33% merupakan komposisi terbaik, memberikan hasil swelling maksimum dalam medium aquadest yakni sebesar 490% selama 90 menit. Hasil uji kapasitas release maksimum diperoleh untuk ion Ag+ sebesar 72% dan ion Zn2+ sebesar 91%. Kinetika swelling air hidrogel nanokomposit 2,33% mengikuti orde pseudo satu dengan nilai regresi R2 = 0,9499. Persamaan laju swelling dari hidrogel nanokomposit NaAlg-PVA/Ag doping ZnO (2,33%) adalah V = k [absorbat]1. Hidrogel nanokomposit NaAlg-PVA/Ag doping ZnO memberikan aktivitas antibakteri terhadap gram positif Staphylococcus aureus dan bakteri gram negatif Escherichia coli dengan zona hambat 12mm dan 23mm. Since biopolymer-based hydrogels are good at killing bacteria, they have been extensively researched for use as wound dressings. As an antibacterial agent, modified zinc oxide-doped silver nanoparticles (Ag-doped ZnO NP) were added to polyvinyl alcohol (PVA) biopolymer to enhance sodium alginate (NaAlg) biopolymer. The successful synthesis of hydrogel nanocomposite SA-PVA/Ag doped ZnO was characterized by FTIR, XRD, SEM, AAS and TGA. The morphology of the hydrogel nanocomposite SA-PVA/Ag doped ZnO using SEM obtained an irregular porous structure with the surface covered with Ag doped ZnO nanoparticles. Hydrogel nanocomposite SA-PVA/Ag doped ZnO with biopolymer concentration variation of 1:1 and Ag-doped ZnO nanoparticles concentration of 2.33% was the best composition, giving maximum swelling results in aquadest medium which is 490% for 90 minutes. The maximum release capacity test results were obtained for 72% Ag+ ions and 91% Zn2+ ions. With a regression value of R2 = 0.9499, the swelling kinetics of the water hydrogel nanocomposite at 2.33% followed pseudo first order. The hydrogel nanocomposite SA-PVA/Ag doped ZnO (2.33%) has a swelling rate equation of V = k [absorbate]1. Hydrogel nanocomposite SA-PVA/Ag doped ZnO provided antibacterial activity against gram-positive Staphylococcus aureus and gram-negative bacteria Escherichia coli with inhibition zones of 12mm and 23mm. |
