Pengaruh teh hitam dibandingkan dengan teh hijau terhadap kualitas spermatozoa manusia yang berasal dari semen astenozoospermia secara invitro

ABSTRAK Huang Lingkup dan Cara Penelitian : Telah dilakukan penelitian mengenai pengaruh pemberian teh hijau dan teh hitam terhadap kualitas spermatozoa yang berasal dari semen astenozoospermia. Penelitian ini dilakukan di bagian Biologi Fakultas Kedokteran Indonesia. Seluruh sampel semen diperoleh dari 30 pasangan infertil. Tiap sampel semen dibagi dalam 4 kelompok, yakni kontrol tanpa perlakuan, kontrol dengan perlakuan (penambahan 50 μL aquabides), dan 2 kelompok perlakuan masing-masing yaitu 500 μL semen ditambahkan 50 μL larutan teh hitam dan larutan teh hijau. Motilitas, kecepatan gerak, penetrasi ke dalam getah serviks sapi, dan integritas membran spermatozoa diteliti satu jam setelah inkubasi. Hasil dan Kesimpulan : Hasil penelitian yang diperoleh menunjukkan, bahwa motilitas spermatozoa diberi perlakuan dengan larutan teh hitam memperlihatkan peningkatan yang bermakna (p<0,01) dibandingkan dengan kelompok teh hijau dan kelompok - kelompok kontrol. Sebaliknya antara kelompok teh hijau dan semua kelompok kontrol tidak terdapat perbedaan motilitas yang bermakna. Kecepatan gerak kelompok kontrol tidak dipengaruhi oleh perbedaan perlakuan. Dengan analisis variansi jelas terlihat bahwa tidak ada perbedaan yang bermakna (P<0,05) pada kecepatan gerak antara kelompok perlakuan dengan kelompok kontrol. Persentase uji HOS positif antara kedua kelompok perlakuan dan kelompok kontrol tidak bermakna. Jarak yang terjauh (5 cm) dari penetrasi sperma ke dalam getah serviks sapi terdapat pada kelompok kontrol tanpa perlakuan dan kelompok perlakuan teh hitam, dibandingkan dengan teh hijau (4 cm). Materials and methods : The effects of black tea and green tea invitro on the quality of spermatozoal from asthenozoospermia semen was investigated. This study was carried out in the Department of Biology, Faculty of Medicine University of Indonesia. All semen were obtained from 30 infertile couples. Each sample of semen was devided into 4 groups, that were untreated control, treated control (added 50 μL aquabidest), and 2 treated groups namely 50μL of semen added to 50 µL black tea solution and green tea solution, respectively. The motility, velocity, penetration into the bovine cervical mucus, and the integrity of spermatozoal membrane were studied one hour after incubation. Result and conclusion : The results of this investigation showed that motility of spermatozoa which treated with black tea solution were significantly increased (P<0.01) as compared to green tea group and to the control groups. However, no significant differences were found between the green tea compared to all control groups. The velocity in the treated groups were unaffected by the different treatments. With an analysis of variance, it was clear that there was no significant difference (P>0.05) in the velocity between experimental groups and the control groups. The percentage of positive HOS test between both treated groups and the control groups were not significant as well. Highly distance (5 cm) of sperm penetration into the bovine. cervical mucus were noted from that found in the untreated control group and from black tea group of treatment, compared to green tea (4 cm). |
