Pengobatan kanker payudara saat ini sedang dipertimbangkan karena toksisitas kemoterapi tradisional. Oleh karena itu, penting untuk mencari alternatif berbasis alam dengan toksisitas lebih rendah. Rimpang temulawak (Curcuma xanthorrhiza Roxb.) mengandung senyawa kurkuminoid dan xantorizol yang berpotensi sebagai agen antikanker. NADES sebagai salah satu pelarut hijau telah digunakan dalam banyak ekstraksi karena kelebihannya. Namun, belum ada penelitian tentang aktivitas antikanker dari ekstrak NADES temulawak. Penelitian ini bertujuan untuk mengevaluasi dan membandingkan aktivitas antikanker dari ekstrak NADES kolin klorida-gliserol (ChCl-Gly) dan ekstrak etanol 96% temulawak pada sel kanker payudara MCF-7 menggunakan uji MTT. Metode antipelarut digunakan untuk memulihkan kurkuminoid dan xantorizol dari ekstrak NADES ChCl-Gly karena toksisitasnya terhadap sel MCF-7. Hasil penelitian menunjukkan persentase pemulihan kembali kurkuminoid dan xantorizol berturut-turut sebesar 17,1% dan 16,47%. Kadar kurkuminoid dan xantorizol dalam ekstrak NADES setelah pemulihan adalah 187,97 ± 0,002 mg/g dan 132,30 ± 0,005 mg/g ekstrak, sedangkan ekstrak etanol mengandung 161,96 ± 0,038 mg/g dan 566,37 ± 0,074 mg/g ekstrak. Nilai IC50 untuk ekstrak NADES dan etanol temulawak berturut-turut adalah 1771,29 µg/mL dan 793,19 µg/mL. Dapat disimpulkan bahwa ekstrak NADES temulawak tidak menunjukkan aktivitas antikanker, sementara ekstrak etanol memiliki aktivitas antikanker yang lemah. Breast cancer treatment was being reconsidered due to the toxicity of traditional chemotherapy. Therefore, it was important to search for alternative natural options with lower toxicity. The rhizome of temulawak (Curcuma xanthorrhiza Roxb.) contained curcuminoid and xanthorrhizol compounds, which had the potential as anticancer agents. NADES as one of the green solvents had been widely used in various extractions due to its advantages. However, there had been no research on the anticancer activity of temulawak NADES extract. This study aimed to evaluate and compare the anticancer activity of temulawak NADES extract (ChCl-Gly) and 96% ethanol extract on MCF-7 breast cancer cells using the MTT assay. The anti-solvent method was used to recover curcuminoid and xanthorrhizol from temulawak NADES extract due to its toxicity to MCF-7 cells. The results showed a recovery percentage of 17.1% for curcuminoid and 16.47% for xanthorrhizol. The content of curcuminoid and xanthorrhizol in the recovered NADES extract was 187.97 ± 0.002 mg/g and 132.30 ± 0.005 mg/g, respectively, while the ethanol extract contained 161.96 ± 0.038 mg/g of curcuminoid and 566.37 ± 0.074 mg/g of xanthorrhizol. The IC50 values obtained for temulawak NADES and ethanol extracts were 1771.29 µg/mL and 793.19 µg/mL, respectively. It was concluded that temulawak NADES extract did not exhibit anticancer activity, while the ethanol extract showed weak anticancer activity. |
