"Kanker paru merupakan salah satu kanker yang memiliki angka mortalitas yang tinggi di Indonesia. Sargassum polycystum merupakan salah satu spesies rumput laut yang banyak ditemui di perairan laut Indonesia dan memiliki potensi sebagai agen antikanker namun belum banyak diteliti. Penelitian ini bertujuan untuk mengetahui kandungan metabolit sekunder dan aktivitas sitotoksik dari ekstrak Sargassum polycystum terhadap sel kanker paru A-549. Sargassum polycystum diekstraksi menggunakan empat jenis pelarut, sehingga menghasilkan empat jenis ekstrak Sargassum polycystum, yaitu ekstrak n-heksana, ekstrak etil asetat, ekstrak kloroform, dan ekstrak etanol. Uji fitokimia pada keempat ekstrak Sargassum polycystum menunjukkan metabolit sekunder berupa flavonoid, tannin, glikosida, alkaloid, triterpenoid, dan steroid. Pada uji KLT menunjukkan bahwa ekstrak Sargassum polycystum memiliki tiga hingga empat senyawa kimia. Uji in vitro aktivitas sitotoksik keempat ekstrak Sargassum polycystum terhadap sel kanker paru A549 menggunakan metode MTT assay pada panjang gelombang 492 nm dengan 5 variasi konsentrasi, yaitu 3,125 g/mL; 6,25 g/mL; 12,5 g/mL; 25 g/mL; dan 50 g/mL. Data yang diperoleh dianalisis dengan menggunakan microsoft excel sehingga diperoleh nilai IC50. Berdasarkan nilai IC50 yang diperoleh, semua ekstrak memiliki nilai IC500,05. Hal ini menunjukkan ekstrak n-heksana memiliki aktivitas sitotoksik paling kuat terhadap sel kanker paru A-549 dibandingkan ekstrak lainnya, sehingga ekstrak n-heksana Sargassum polycystum berpotensi dikembangkan sebagai agen antikanker paru.

Lung cancer is one of cancer that has high mortality rate in Indonesia. Sargassum polycystum is one of macroalgae species that commonly found in Indonesian marine and can be developed as anticancer agent but has not been widely studied. Objective of this research is to know second metabolites compounds and cytotoxic activity of extract Sargassum polycyctum on A 549 lung cancer cell. Sargassum polycystum was extracted using four solvent to produce four extract, there are n hexane extract, ethyl acetate extract, chloroform extract, and ethanol extract. Phytochemical test of each extract showed positive result of flavonoid, tannin, glycoside, alkaloid, triterpenoid, and steroid. Besides, TLC test showed there were three to four chemical compound in each extract of Sargassum polycystum. In vitro cytotoxic activity of four extract Sargassum polycystum on A 549 lung cancer were tested using MTT assay on 492 nm wavelength and devided into five different concentration, there are 3,125 g ml 6,25 g ml 12,5 g ml 25 g ml and 50 g ml. The obtained data were analyzed using microsoft excel to get IC50 value. The result showed that each extract of Sargassum polycycstum had IC50 values less than 100 g ml. This means each extract of Sargassum polcystum has cytotoxic activity against A 549 lung cancer cell. Among four extract of Sargassum polycytum, n hexane extract has the lowest IC50 value 24,1 g ml and shows the most potent cytotoxic activity against A 549 lung cancer cell. Statistical analytic showed p value 0,05. So, n hexane extract of Sargassum polycystum potential to be developed as antilung cancer agent."

"ABSTRAK
Leukemia adalah salah satu kanker dengan angka prevalensi dan mortalitas yang tinggi di dunia dan di Indonesia, salah satunya karena terapi yang digunakan saat ini masih banyak menimbulkan efek samping dan tidak bersifat spesifik. Salah satu jenis rumput laut, yaitu Sargassum vulgare, memiliki potensi sebagai agen anti kanker leukemia. Tujuan dari penelitian ini adalah untuk mengetahui kandungan metabolit skunder dan efek antiproliferatif makroalga Sargassum vulgare terhadap sel kanker leukemia P388 yang diekspresikan dengan nilai IC50. Metode MTT assay digunakan untuk menguji efek antiproliperatif Sargassum vulgare terhadap sel leukimia P388. Pada penelitian ini digunakan empat jenis ekstrak Sargassum vulgare, yaitu ekstrak n-heksana, etanol, kloforom, dan etil asetat dengan lima variasi konsentrasi. Uji fitokimia dilakukan unltuk menganalisis kandungan metabolit sekunder pada setiap ekstrak, sedangkan metode Kromatografi Lapis Tipis KLT digunakan untuk mengetahui jumlah komponen senyawa kimia yang terdapat dalam tiap ekstrak. Uji fitokimia memperlihatkan hasil positif untuk saponin, tannin, alkaloid, dan triterpenoid. Analisis kualitatif metode kromatografi lapis tipis menunjukkan bahwa ekstrak Sargassum vulgare mengandung dua hingga enam senyawa yang berbeda. Hasil uji efek antiproliperatif keempat jenis ekstrak Sargassum vulgare terhadap sel leukimia P388 memberikan nilai IC50 kurang dari 100 ?g/ml, yang diinterpretasikan sebagai agen sitotoksik yang baik terhadap penghambatan proliferasi sel Leukemia P388. Efek antiproliperatif terbaik ditunjukkan oleh esktrak etil asetat Sargassum vulgare, dengan nilai IC50 sebesar 13,7 ?g/ml.

ABSTRACT
Leukemia is the blood cancer with high prevalence and mortality in the world and in Indonesia, due to technology and chemotherapy for treating patients with leukemia has many side effects and poor to treat cancer cell specifically. One of the brown seaweed which has potential to be used as cytotoxic and anticancer agent, especially for leukemia, is Sargassum vulgare. The objective of this research is to know secondary metabolit composition and antiproliferative effect of extract Sargassum vulgare on leukemia P388 cell which can be expressed by IC50 value. The method that was used in this research to analyze antiproliferative effect of Sargassum vulgare extract on leukemia P388 cell. Researcher used four varians of Sargassum vulgare extract which were extracted using n hexane, ethanol, chloforom, and ethyl acetate with five varians of concentration. Phytochemical methode was used to analyzed secondary metabolite composition of fours extract Sargassum vulgae, while qualitative analysis to find out the quantity of chemical compound component of the fours extracts used Thin Layer Chromatography TLC methode. The result of phytochemical methode are positive for saponin, tannin, alkaloid, and triterpenoid. While the result of qualitative analysis using Thin Layer Chromatography showed that the extract contains two to six chemical compounds. Then, measuring of absorbance and continued by analyzing data to obtain IC50 value. The results showed that all of Sargassum vulgare extract had IC50 values less than 100 g ml which could be interpreted as good cytotoxic agen for inhibiting proliferation of P388 leukemia cell. The lowest IC50 value among four extracts of Sargassum vulgare is showed byethyl acetate extract 13,7 g ml , thus has the best potency to be good cytotoxic agent against leukemic cell."

"ABSTRAK
Jumlah kasus kanker paru terus mengalami peningkatan secara global. Disamping kasusnya yang terus bertambah, terapi kanker paru hingga saat ini masih banyak menimbulkan efek samping yang tidak jarang menggangu kualitas hidup pasien. Maka dari itu, skripsi ini mengevaluasi aktivitas antioksidan dan antikanker dari rumput laut Eucheuma cottonii dan Eucheuma spinosum yang berasal dari pantai Labuan Aji, Nusa Tenggara Barat terhadap sel paru A-549.  Pada penelitian ini digunakan empat jenis ekstrak, yakni heksana, kloroform, etil asetat, dan etanol. Uji aktivitas antioksidan dilakukan dengan metode DPPH sedangkan untuk aktivitas antikanker dievaluasi dengan metode MTT cell proliferation assay. Masing-masing ekstrak akan dinilai laju inhibisinya dan nilai IC50. Berdasarkan hasil percobaan, ekstrak etanol Eucheuma cottonii dengan nilai IC50 35,51 ppm memiliki aktivitas antioksidan yang paling poten dibandingkan dengan ekstrak lain yang diuji. Ekstrak etanol E. cottonii juga memiliki aktivitas antikanker terbaik, dengan nilai IC50 54,83 mg/mL. Untuk Eucheuma spinosum, ekstrak etil asetat memiliki aktivitas antioksidan dan antikanker yang paling baik, dengan nilai IC50 10,16 mg/mL. Dengan demikian, dapat disimpulkan bahwa baik Eucheuma cottonii maupun Eucheuma spinosum berpotensi sebagai antioksidan alami dan memiliki efek antikanker terhadap sel paru A-549. 

ASTRACT
Number of people suffering from lung cancer continue to increase globally. Yet, the treatment methods for lung cancer still have many effects that often interfere patients quality of life. Therefore, objective from this paper is to evaluate the antioxidant and anticancer activity from macroalgae Eucheuma cottonii and Eucheuma spinosum, which was taken from Labuan Aji beach, Nusa Tenggara Barat, against A-549 lung cell. This research used four types of extracts, including hexane, chloroform, ethyl acetate, and ethanol. Antioxidant activity was evaluated using DPPH method. As for anticancer activity was evaluated by MTT cell proliferation assay. Each extracts have its own inhibitory rate and IC50 value. Based on this experiment, ethanol extract of Eucheuma cottonii with IC50 value 35,51 ppm had the most potent antioxidant activity than other extracts that were tested. Ethanol extract also showed the best anticancer activity with IC50 value 54,83mg/mL. As for Eucheuma spinosum, ethyl acetate extract has the best antioxidant and anticancer activity, with IC50 value 10,16 mg/mL. Thus, it can be concluded that both Eucheuma cottonii and Eucheuma spinosum are potent natural antioxidant and have anticancer effect against A-549 lung cacner cell. "

"ABSTRAK
Kanker serviks merupakan salah satu jenis penyakit kanker dengan prevalensi dan mortalitas yang tinggi di Indonesia. Eucheuma cottonii merupakan salah satu jenis rumput laut yang banyak dijumpai di perairan laut Indonesia dan dapat dikembangkan sebagai agen anti kanker. Penelitian ini bertujuan untuk mengetahui komposisi fitokimia dan efek sitotoksik ekstrak makroalga Eucheuma cottonii terhadap sel kanker serviks HeLa yang dinyatakan dengan nilai IC50. Pada penelitian ini, Eucheuma cottonii diekstraksi masing-masing menggunakan pelarut etanol, etil asetat, n-heksana, dan kloroform. Ekstrak yang diperoleh kemudian diuji kandungan fitokimianya, dan menunjukkan hasil positif mengandung flavonoid. Sedangkan analisis kualitatif dengan kromatografi lapis tipis, menunjukkan bahwa ekstrak mengandung tiga hingga lima senyawa kimia. Selanjutnya, masing-masing ekstrak sebanyak 20 L dengan 5 variasi konsentrasi, yaitu 3,125 g/ml; 6,25 g/ml; 12,5 g/ml; 25 g/ml; dan 50 g/m, dimasukkan ke lini sel HeLa yang sudah ditambahkan 100 L DMEM dan diinkubasi selama 24 jam. Setelah diinkubasi selama 24 jam, dilakukan uji MTT assay dengan panjang gelombang 492 nm. Data yang diperoleh kemudian dianalisis sehingga diperoleh nilai IC50 untuk keempat ekstrak Eucheuma cottonii yang diujikan. Hasil menunjukkan bahwa keempat ekstrak Eucheuma cottonii memiliki nilai IC50

ABSTRACT
Cervical cancer is one kinds of cancer with high prevalence and mortality in Indonesia. Eucheuma cottonii is one kind of seaweed which is commonly found in Indonesian marine and can be developed as anticancer agent. This research aims to know the phytochemical composition and cytotoxic effect of extract of makroalgae Eucheuma cottonii on HeLa cervical cancer cells that is expressed by IC50 value. In this research, Eucheuma cottonii were extracted each using ethanol, ethyl acetate, n hexane, and chloroform. The obtained extracts were then tested for its phytochemical content, and showed positive result containing flavonoids. While qualitative analysis with Thin Layer Chromatography TLC , showed that the extract contains three to five chemical compounds. Furthermore, 20 L of each extract in five variation of concentration, i.e. 3,125 g ml 6,25 g ml 12,5 g ml 25 g ml and 50 g ml, inserted into the HeLa cell line that has been added 100 L of DMEM and incubated for 24 hours. After 24 hour incubation, MTT assay with a wavelength of 492 nm was performed to generate data which was then analyzed to obtain IC50 value for the four extracts tested of Eucheuma cottonii. The results showed that all four Eucheuma cottonii extract had IC50 values less than 100 g ml, so it can be concluded that all extracts have cytotoxic activity against HeLa cervical cancer cell HeLa. Among four extracts of Eucheuma cottonii, ethyl acetate extract has the lowest IC50 value and shows the most potent cytotoxic activity against HeLa cervical cancer cells. "Alga coklat (S. polycystum) yang diekstraksi dengan berbagai macam pelarut diketahui memiliki aktivitas sitotoksik pada sel kanker kolon HCT-116. Nilai IC-50 paling baik ditunjukkan pada alga coklat (S. polycystum) yang diekstraksi menggunakan pelarut etil asetat. Penelitian ini bertujuan untuk membuktikan efek ekstrak alga coklat (S. polycystum) dalam menghambat kolitis terkait kanker kolon secara in vivo. Sebanyak 30 ekor mencit jantan galur Balb/C dibagi acak dalam 5 kelompok yaitu normal, negatif, dosis 1 (18 mg/kgBB), dosis 2 (90 mg/kgBB), dosis 3 (450 mg/kgBB). Hewan model dibuat dengan mengadministrasikan Dekstran Sodium Sulfat (DSS) selama 24 hari secara berturut-turut dengan konsentrasi 2% selama 7 hari, konsentrasi 1% selama 10 hari, dan konsentrasi 2% selama 7 hari. Pemberian ekstrak alga coklat (S. polycystum) diberikan pada hari ke-8 berlanjut hingga 14 hari setelah induksi. Selama penelitian berlangsung dilakukan pengukuran berat badan hewan, kemampuan bertahan hidup, dan penilaian Disease Activity Index (DAI). Setelah pengorbanan hewan dilakukan isolasi kolon untuk mengukur panjang, berat kolon, kerusakan jaringan, jumlah sel goblet dengan metode histopatologi kolon serta analisis ekspresi Caspase-3 pada jaringan kolon menggunakan metode ELISA. Hasil penelitian menunjukkan bahwa ekstrak alga coklat (S. polycystum) pada dosis 1 (18 mg/kgBB), menunjukkan perbaikan kerusakan jaringan kolon paling optimal dan memiliki konsentrasi Caspase-3 paling tinggi dibandingkan kelompok dosis lainnya.

Brown algae (S. polycystum) extracted with various solvents is known to have cytotoxic activity on HCT-116 colon cancer cells. IC-50 value is best shown in brown algae (S. polycystum) extracted using ethyl acetate solvent. This study aims to prove the effect of brown algae extract (S. polycystum) in inhibiting colitis associated with colon cancer in vivo. Thirty male mice Balb/C strain were randomly divided into 5 groups: normal, negative, dose 1 (18 mg/kgBB), dose 2 (90 mg/kgBB), dose 3 (450 mg/kgBB). Animal models were made by administering Dextran Sodium Sulphate (DSS) for 24 consecutive days with a concentration of 2% for 7 days, 1% concentration for 10 days, and 2% concentration for 7 days. Brown algae extract (S. polycystum) was given on the 8th day and continued until 14 days after induction. During the study, animal body weight, survival ability, and Disease Activity Index (DAI) were measured. After animal sacrifice, colon isolation was carried out to measure the length, weight of the colon, tissue damage, number of goblet cells using the colon histopathology method and analysis of Caspase-3 expression in colon tissue using the ELISA method. The results showed that brown algae extract (S. polycystum) at dose 1 (1.8 mg/kg BW) showed the most optimal colonic damage improvement and had the highest concentration of Caspase-3 compared to the other dose groups."

"Latar belakang: Kanker kolorektal adalah satu jenis kanker dengan jumlah kasus terbanyak di Indonesia. Dalam berbagai penelitian, bunga kecombrang memiliki zat antikanker dengan efek sitotoksik serta antioksidan. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dan sitotoksik ekstrak heksana bunga kecombrang terhadap sel kanker kolon HT-29.
Metode: Penelitian ini menggunakan desain eksperimental in vitro menggunakan sel kanker kolon HT-29. Sebanyak 45sampel sel HT-29 dibagi menjadi kelompok perlakuan dan kelompok kontrol positif. Analisis kandungan ekstrak heksana bunga kecombrang akan dilakukan menggunakan uji fitokimia dan uji KLT. Pengukuran efek antioksidan akan dilakukan menggunakan metode uji dengan reagen DPPH dan untuk efek sitotoksisitas menggunakan uji MTT assay. Nilai absorbansi dan persen inhibisi dari kedua metode ini lalu dibandingkan antara kelompok ekstrak dan kelompok kontrol positif menggunakan analisis regresi linear.
Hasil: Ekstrak heksana bunga kecombrang mengandung senyawa saponin, flavonoid, triterpenoid, dan alkaloid. Ekstrak memiliki efek antioksidan yang tidak aktif dengan nilai IC50 pada uji DPPH sebesar 21.337,99 ppm. Ekstrak memiliki efek sitotoksik dengan klasifikasi sedang dengan nilai IC50 pada uji MTT sebesar 111,097 ppm. Dibandingkan dengan kontrol positif, ekstrak memiliki efek antioksidan dan sitotoksik yang lebih rendah.
Kesimpulan: Ekstrak heksana bunga kecombrang tidak aktif memberikan efek antioksidan dan memberikan efek sitotoksik yang sedang, meski lebih rendah daripada kontrol positif. Meskipun begitu, tetap terdapat hubungan antara konsentrasi ekstrak heksana bunga kecombrang dengan persentase inhibisi. Oleh karena itu, diperlukan penelitian dengan desain dan jumlah sampel yang lebih baik.

ntroduction: Colorectal cancer is one type of cancer with the highest number of cases in Indonesia. Etlingera elatior have anticancer substances with cytotoxic and antioxidant effects. This study aims to determine the antioxidant and cytotoxic activity of hexane extract from Etlingera elatior against HT-29 cells.
Method: This study used an in vitro experimental design using HT-29 colon cancer cells. A total of 45 samples of HT-29 cells were divided into a treatment group and a positive control group. Analysis of the hexane extract from Etlingera elatior was carried out using phytochemical tests and TLC tests. Measurement of the antioxidant effect was carried out using the test method with DPPH reagent and for the cytotoxicity effect using the MTT assay test. The absorbance values ​​and percent inhibition of these two methods were then compared between the extract group and the positive control group using linear regression analysis.
Result: Hexane extract from Etlingera elatior contained saponins, flavonoids, triterpenoids, and alkaloids. The extract had an inactive antioxidant effect with an IC50 value of 21,337.99 ppm in the DPPH test. The extract had a moderate cytotoxic effect with an IC50 value of 111,097 ppm in the MTT test. Compared with the positive control, the extract had lower antioxidant and cytotoxic effects.
Conclusion: Hexane extract from Etlingera elatior did not have an antioxidant effect and had a moderate cytotoxic effect, although it was lower than the positive control. Even so, there was still a relationship between the concentration of hexane extract of kecombrang flowers and the percentage of inhibition. Therefore, more research is needed with a better design and number of samples.

Pendahuluan: Kanker paru merupakan salah satu jenis kanker dengan angka kejadian dan kematian yang tinggi baik di dunia maupun di Indonesia. Perawatan saat ini untuk kanker paru-paru adalah operasi, kemoterapi, terapi radiasi dan terapi bertarget. Namun, berbagai terapi ini dapat menyebabkan beberapa efek samping. Makroalga Eucheuma cottonii merupakan salah satu bahan alami yang potensial untuk pengobatan kanker paru. Penelitian ini dilakukan untuk mengetahui komposisi senyawa fitokimia, aktivitas antioksidan dan efek sitotoksik makroalga Eucheuma cottonii asal Sorong, Papua Barat pada kultur sel kanker paru A549. Metode: Penelitian ini menggunakan desain penelitian eksperimen. Sampel E. cottonii diekstraksi dalam 3 jenis pelarut, yaitu etanol, etil asetat dan n-heksana. Kemudian dilakukan uji kandungan senyawa fitokimia dan kromatografi lapis tipis (KLT). Aktivitas antioksidan ekstrak diuji pada enam konsentrasi ekstrak yang berbeda menggunakan metode DPPH. Aktivitas sitotoksik ekstrak E. cottonii terhadap sel kanker paru A549 diuji dalam delapan konsentrasi berbeda menggunakan metode MTT. Hasil: Uji fitokimia ekstrak E. cottonii menunjukkan adanya senyawa triterpenoid dan alkaloid. Analisis KLT menunjukkan bahwa ekstrak E. cottonii mengandung satu sampai tiga senyawa kimia. Uji antioksidan dengan metode DPPH ekstrak etanol dan etil asetat makroalga E. cottonii memberikan nilai IC50 masing-masing sebesar 64,2 g/ml dan 181,7 g/mL. Uji sitotoksik ekstrak etanol, ekstrak etil asetat dan ekstrak n-heksana E. cottonii terhadap sel kanker paru A549 secara in vitro memberikan nilai IC50 masing-masing sebesar 168,56 g/mL, 196,87 g/mL, dan 1485,10 g. /mL. Kesimpulan: Ekstrak E. cottonii memiliki aktivitas antioksidan sedang dan memiliki efek sitotoksik lemah terhadap sel kanker paru A549.
ABSTRACT
Introduction: Lung cancer is one type of cancer with a high incidence and mortality rate both in the world and in Indonesia. Current treatments for lung cancer are surgery, chemotherapy, radiation therapy and targeted therapy. However, these various therapies can cause some side effects. Macroalgae Eucheuma cottonii is one of the potential natural ingredients for the treatment of lung cancer. This research was conducted to determine the composition of phytochemical compounds, antioxidant activity and cytotoxic effects of macroalgae Eucheuma cottonii from Sorong, West Papua on A549 lung cancer cell culture. Methods: This study used an experimental research design. Samples of E. cottonii were extracted in 3 types of solvents, namely ethanol, ethyl acetate and n-hexane. Then, the phytochemical compound content test and thin layer chromatography (TLC) were tested. The antioxidant activity of the extracts was tested at six different extract concentrations using the DPPH method. The cytotoxic activity of E. cottonii extract against lung cancer cells A549 was tested in eight different concentrations using the MTT method. Result: Phytochemical test of E. cottonii extract showed the presence of triterpenoid and alkaloid compounds. TLC analysis showed that the extract of E. cottonii contained one to three chemical compounds. Antioxidant test using DPPH method of ethanol extract and ethyl acetate of macroalgae E. cottonii gave IC50 values ​​of 64.2 g/ml and 181.7 g/mL, respectively. Cytotoxic test of ethanol extract, ethyl acetate extract and n-hexane extract of E. cottonii against lung cancer cells A549 in vitro gave IC50 values ​​of 168.56 g/mL, 196.87 g/mL, and 1485.10 g, respectively. . /mL. Conclusion: Extract of E. cottonii has moderate antioxidant activity and has a weak cytotoxic effect against lung cancer cells A549."

"Sargassum polycystum diketahui secara in vitro memiliki aktivitas sitotoksik terhadap sel HCT-116 pada kolon yang diuji pada beberapa pelarut dan dengan nilai IC50 yang berbeda. Namun, penelitian secara in vivo Sargassum polycystum pada kanker kolon belum banyak dilakukan dan mekanisme sepenuhnya dalam penghambatan kanker belum diketahui. Penelitian ini bertujuan untuk mengetahui efek alga coklat (Sargassum polycystum) secara in vivo pada hewan model kolitis terkait kanker kolon yang diinduksi dengan Dekstran Sodium Sulfat (DSS). Penelitian ini menggunakan mencit jantan galur Balb/c (n = 30 ekor) yang secara acak dibagi dalam 5 kelompok: kelompok normal, kelompok negatif, kelompok dosis 1 (18 mg/kgBB), kelompok dosis 2 (90 mg/kgBB), dan kelompok dosis 3 (450 mg/kgBB). Induksi kolitis terkait kanker kolon menggunakan senyawa kimia Dekstran Sodium Sulfat (DSS) dengan konsentrasi 2% dan 1% selama total 24 hari. Pengukuran berat badan, analisis kelangsungan hidup, dan penilaian Disease Activity Index (DAI) dilakukan selama penelitian berlangsung. Pengaruh Sargassum polycystum sebagai antikanker diamati dengan memeriksa variabel inflamasi IL-1β dan pemeriksaan histologi jaringan kolon dengan periodic acid-schiff (PAS). Hasil penelitian menunjukkan bahwa hewan uji yang diberikan Sargassum polycystum pada dosis 18 mg/kgBB memiliki kelangsungan hidup lebih tinggi dan dapat menurunkan ekspresi variabel inflamasi IL-1β.

Sargassum polycystum is known in vitro to have cytotoxic activity against HCT-116 cells in the colon, which were tested in several solvents with different IC50 values. However, in vivo studies of Sargassum polycystum on colon cancer have not been widely carried out, and the full mechanism of cancer inhibition is not yet known. This study aims to determine the effect of brown algae (Sargassum polycystum) in vivo on an animal model of colitis related to colon cancer induced by Dextran Sodium Sulfate (DSS). This study used male mice of the Balb/c strain (n = 30), which were randomly divided into 5 groups: normal group, negative group, dose 1 group (18 mg/kgBW), dose 2 group (90 mg/kgBW), and dose group 3 (450 mg/kgBW). Induction of colitis-associated colon cancer using the chemical compound Dextran Sodium Sulfate (DSS) with a concentration of 2% and 1% for a total of 24 days. Body weight measurements, survival analysis, and Disease Activity Index (DAI) assessments were carried out during the study. The effect of Sargassum polycystum as an anticancer agent was observed by examining the inflammatory variable IL-1β and histological examination of colonic tissue with periodic acid-schiff (PAS). The results showed that the test animals that were given Sargassum polycystum at a dose of 18 mg/kgBW had higher survival and could reduce the expression of the inflammatory variable IL-1β."

"Latar belakang: Kanker kolorektal merupakan salah satu kanker dengan prevalensi tertinggi di dunia. Pengobatan kanker yang berdasarkan pembedahan dan kemoterapi masih memberikan prognosis yang kurang baik bagi kebanyakan pasien. Tatalaksana dan zat antikanker yang tersedia dan teruji klinis juga masih memberikan efikasi yang kurang baik ditandai dengan angka mortalitas yang masih tinggi. Berbagai studi telah menunjukkan bahwa bunga kecombrang mengandung berbagai metabolit aktif yang berpotensi menjadi zat antikanker. Penelitian ini bertujuan untuk melihat profil fitokimia, sifat antioksidan dan sifat sitotoksik ekstrak etil asetat bunga kecombrang terhadap sel kanker kolon HT-29. 
Metode: Penelitian ini menggunakan desain eksperimental in vitro menggunakan galur sel kanker kolon HT-29. Sebanyak 45 sampel sel HT-29 yang dibagi menjadi kelompok perlakuan dan kelompok kontrol positif akan diletakkan kedalam well plate dan diberikan konsentrasi secara serial. Analisis kandungan metabolit ekstrak dilakukan menggunakan skrining uji fitokimia dan uji KLT. Pengukuran sifat antioksidan akan dilakukan menggunakan metode uji dengan reagen DPPH dan untuk sifat sitotoksisitas menggunakan metode MTT assay. Nilai absorbansi dan persen inhibisi dari kedua metode ini kemudian akan dibandingkan antara kelompok ekstrak dan kelompok kontrol positif menggunakan analisis regresi linear multipel. 
Hasil: Berdasarkan uji fitokimia dan uji KLT, ekstrak etil asetat bunga kecombrang mengandung 4 golongan metabolit sekunder aktif seperti saponin, flavonoid, alkaloid dan triterpenoid. Ekstrak etil asetat bunga kecombrang tidak menunjukkan adanya sifat antioksidan ditandai dengan nilai IC50 pada uji DPPH sebesar1282 ppm dan menunjukkan sifat sitotoksitas moderat ditandai dengan nilai IC50 pada uji MTT assay sebesar 71 ppm. Hasil uji regresi linear multipel juga menunjukkan bahwa sifat antioksidan vitamin C dan sifat sitotoksisitas doksorubisin sebagai kontrol positif masih lebih unggul dibandingkan dengan ekstrak etil asetat bunga kecombrang.  
Kesimpulan: Ekstrak etil asetat bunga kecombrang tidak memiliki sifat antioksidan dan memiliki sifat sitotoksik moderat. Kekuatan antioksidan dan sitotoksisitas ekstrak juga tidak sepadan bila dibandingkan dengan agen antioksidan dan sitotoksik konvensional seperti vitamin C dan doksorubisin. Oleh karena itu, diperlukan studi lanjutan dengan tingkat yang lebih tinggi dan jumlah sampel yang lebih besar lagi. 

Introduction: Colorectal cancer is one of the most prevalent cancers in the world. Current anticancer treatments, which are based on surgery and chemotherapy still provide poor prognosis for most of the colorectal patients. Many studies have been conducted to learn about the extract from torch ginger flower as a potential candidate for anticancer agent. The goal of this study is to gather knowledge about the phytochemistry profile, antioxidant activity, and cytotoxic activity from ethyl acetate extract against HT-29 colon cancer cells.
Method: This was an in-vitro experimental study using HT-29 colorectal cancer cell line. 45 samples of HT-29 cells were divided into 2 groups which were the positive control group and the extract group. We used phytochemical screening test and thin plate chromatography to analyse the metabolite content from the extract sample. The antioxidant, the extract’s cytotoxic activity, and the positive control group were analysed using the DPPH method and the MTT assay respectively. We then compared the absorbance value and the inhibition percentage from the extract group to the positive control group using multiple linear regression. 
Result: Based on phytochemical screening and thin plate chromatography, ethyl acetate extract from torch ginger flower contained 4 groups of active metabolites such as  saponin, flavonoid, triterpenoid, and alkaloid. The extract group did not have antioxidant activity with IC50 1282 ppm and showed moderate cytotoxic activity with IC50 71 ppm. From multiple linear regressions, there was a significance difference between the extract and the positive control group for both antioxidant and cytotoxic activity. This study showed that the positive control group which was vitamin C for antioxidant and doxorubicin for cytotoxic, had better activity than the extract group. 

Conclusion: Ethyl acetate extract did not have any antioxidant activity and had moderate cytotoxic activity. However, the cytotoxic activity was also disproportionate when compared to the conventional positive control group. Therefore, further studies with a higher rate and a larger number of samples are needed.

Penelitian mengenai potensi makroalga alami Sargassum polycystum dan makroalga budidaya Eucheuma cottonii dalam menyerap dan menyimpan karbon serta nutrien di Pulau Panjang, Teluk Banten telah dilakukan pada bulan Oktober hingga November 2014. Penelitian bertujuan untuk mengetahui laju penyerapan karbon, kandungan nutrien dan produktivitas primer dari makroalga S. polycystum dan E. cottonii. Lokasi penelitian terletak di bagian hamparan gosong karang dan berlumpur dengan kedalaman 0,5-5 m. Pengamatan pertumbuhan dan laju penyerapan karbon menggunakan metode penandaan thallus pada 30 sampel makroalga setiap hari selama 7 hari. Sampel makroalga selanjutnya dianalisis kandungan nutriennya. Hasil penelitian didapatkan estimasi laju penyerapan karbon S. polycystum dan E. cottonii adalah 0,0081 gC/hari dan 0,0083 gC/hari. Kandungan karbon, nitrogen dan fosfat di S. polycystum adalah sebanyak 6,84%, 1,72% and 0,009% sedangkan kadar karbon, nitrogen dan fosfat di E. cottonii adalah 5,99%, 0,67% dan 0,006%. Berdasarkan analisis statistik dengan uji t, terdapat perbedaan yang signifikan pada laju pertumbuhan, kandungan nitrogen dan fosfat S. polycystum dengan E. cottonii. Sedangkan untuk kandungan karbon tidak terdapat perbedaan yang signifikan antara S. polycystum dengan E. cottonii. Selanjutnya, uji korelasi didapatkan bahwa adanya korelasi antara laju pertumbuhan S. polycystum dan E. cottonii dengan kandungan nitrogen masing- masing sedangkan antara laju pertumbuhan dengan kandungan karbon dan fosfat tidak terdapat korelasi. Produktivitas primer diukur dengan metode botol terang dan botol gelap yang dimodifikasi. Pengukuran produktivitas primer dilakukan pada kedalaman 0,5 m untuk S. polycystum dan 0,1 m untuk E. cottonii. Kandungan oksigen terlarut diukur dengan DO meter. Hasil penelitian didapatkan rata-rata produktivitas primer S. polycystum sebesar 512,99 ± 169,26 mgC/m3/hari dan E. cottonii sebesar 387,88 ± 219,93 mgC/m3/hari.

The research about the potency of wild macroalgae Sargassum polycystum dan cultivation macroalgae Eucheuma cottonii to absorb and store carbon also nutrient in Panjang Island, Banten Bay was held on October until November 2014. The research was aimed to estimate carbon sequestration, nutrient content and primary productivity from macroalgae S. polycystum and E. cottonii. Location of the research in the stretch of reef and muddy with a depth of 0,5-5 m. The observation of growth rate and carbon sequestration used thallus marking method in 30 macroalgae shoots everyday for 7 days. Macroalgae was analyzed its nutrient content. The results showed that estimation carbon sequestration by S. polycystum and E. cottonii were 0,0081 gC/day dan 0,0083 gC/day.
