Penelitian ini bertujuan untuk melakukan pemetaan suhu rutin terhadap Chiller Pharma (2-8°C) untuk mengetahui distribusi suhu penyimpanan dan mengidentifikasi titik terpanas Chiller Pharma (2-8°C). Pemetaan suhu dilakukan dengan menggunakan Electronic Data Logging Monitor (EDLM) selama tiga hari. Hasil penelitian menunjukkan bahwa distribusi suhu memenuhi kriteria rentang suhu 2-8°C dengan nilai Mean Kinetic Temperature (MKT) sebesar 5,86 (Upper) dan 5,41 (Lower). Titik terpanas pada chiller pharma berada pada titik 3 pada rak bagian tengah. Saran yang diajukan adalah pemasangan sensor suhu pada titik terpanas chiller pharma untuk memonitoring suhu kritis secara lebih efektif. Dengan demikian, pemantauan suhu dapat lebih akurat dan konsisten, menjaga kestabilan sediaan farmasi yang disimpan dalam chiller.

Pharmaceutical preparations, including medicines, pharmaceutical ingredients, traditional medicines, and cosmetics, must meet safety, quality, and efficacy standards. The distribution of pharmaceutical preparations requires special attention, especially in procurement, storage, and distribution. Pharmaceutical Wholesalers (PWFs) play a crucial role in ensuring that this process runs smoothly. They are responsible for distributing pharmaceutical preparations, including products that require special handling such as Cold Chain Products (CCPs), such as vaccines and insulin. CCPs must be stored in rooms or containers that can maintain temperature stability, such as using a cold room or chiller, with the required temperature being 2-8°C. This study aims to conduct routine temperature mapping of the Chiller Pharma (2-8°C) to determine the distribution of storage temperature and identify the hottest point of the Chiller Pharma (2-8°C). Temperature mapping is performed using an Electronic Data Logging Monitor (EDLM) for three days. The results show that the temperature distribution meets the criteria for the temperature range of 2-8°C with a Mean Kinetic Temperature (MKT) value of 5.86 (Upper) and 5.41 (Lower). The hottest point of the chiller pharma is at point 3 on the middle rack. The proposed suggestion is to install temperature sensors at the hottest points of the chiller pharma for more effective temperature monitoring. Thus, temperature monitoring can be more accurate and consistent, maintaining the stability of pharmaceutical preparations stored in the chiller."

Chest Freezer machines are used to store ice packs which are useful for sending cold chain products. Machine qualification consists of several qualification stages which are carried out before the machine is used for daily operations. In this research, operational qualification was carried out in accordance with the company's qualification procedures. Operational qualification (OQ) is performed to establish that the installation and all systems and subsystems operate effectively and consistently when the storage area is empty. Operational qualification results for Chest Freezer AB-1200-T-X ID. P05 empty condition has been implemented with the results meeting the acceptance criteria with justification. Therefore, qualification testing can proceed to the performance qualification stage."

Untuk menjaga kualitas produk rantai dingin yang didistribusikan sesuai dengan CDOB, maka diperlukan salah satu fasilitas berupa Chest Freezer yang digunakan untuk menyimpan ice pack yang digunakan untuk menjaga suhu produk rantai dingin saat pengiriman. Penelitian ini dibuat untuk melakukan pengujian kualifikasi operasional pada salah satu Chest Freezer yang ada di PT. Enseval Putera Megatrading Tbk untuk memastikan freezer tersebut memenuhi spesifikasi yang telah ditentukan berdasarkan protokol kualifikasi operasional dan dapat digunakan untuk menyimpan ice pack. Penelitian dilakukan dengan pengujian dan pengamatan kualifikasi operasional Chest Freezer AB-1200-T-X ID.P04 sesuai dengan protokol yang berlaku. Pengujian terdiri atas beberapa prosedur yaitu menyalakan dan mematikan daya, pengaturan suhu, distribusi dingin pada keadaan kosong dan operasi defrost. Setelah pengujian, hasil didokumentasikan dan diolah ke tabel dan grafik Microsoft Excel untuk melihat distribusi temperatur freezer dan keberterimaannya. One chapter within GDP discusses specific provisions for Cold Chain Products (CCPs), which are pharmaceutical products sensitive to temperature, necessitating continuous temperature control during storage and transportation to ensure that product characteristics, stability, efficacy, and safety remain unchanged. To maintain the quality of distributed Cold Chain Products in accordance with GDP, facilities such as a Chest Freezer are required to store ice packs used to maintain the temperature of the products during transportation. This research was conducted to perform operational qualification testing on one of the Chest Freezers at PT. Enseval Putera Megatrading Tbk to ensure that the freezer meets specified requirements based on operational qualification protocols and can be used to store ice packs. The study involved testing and observing the operational qualification of the Chest Freezer AB-1200-T-X ID.P04 according to applicable protocols. Testing procedures included powering on and off, temperature setting, cold distribution in empty condition, and defrost operation. After testing, results were documented and processed into tables and graphs using Microsoft Excel to assess freezer temperature distribution and acceptance criteria. The research findings indicate that the results meet acceptance criteria."

"Produk rantai dingin merupakan salah satu bentuk dari rantai pasok produk yang sangat menjaga suhu produk agar tetap stabil selama proses penyimpanan dan distribusi obat. Penyimpanan produk rantai dingin harus memenuhi persyaratan suhu dengan rentang 2-8°C. Hasil pemetaan suhu menunjukkan bahwa kondisi suhu pada container chiller yang diuji memenuhi kriteria yang dibutuhkan dalam penyimpanan produk rantai dingin karena masih berada pada rentang yang dipersyaratkan yaitu 2-8oC, dengan nilai MKT (Mean Kinetic Temperature) tertinggi dan terendah secara berturut-turut adalah 5,14 (Upper) dan 4,37 (Lower). Suhu tertinggi terdapat pada titik 8, sedangkan suhu terendah berada pada titik 11. Hasil kualifikasi operasional menunjukkan bahwa sistem refrigerasi container chiller dapat berfungsi dengan baik. Alarm pada container chiller juga dapat beroperasi dengan baik.

The temperature mapping results show that the temperature conditions in the chiller container tested meet the criteria required for storing cold chain products because they are still within the required range, namely 2-8oC, with the highest and lowest MKT (Mean Kinetic Temperature) values respectively being 5,14 (Upper) and 4,37 (Lower). The operational qualification results show that the container chiller refrigeration system can function well. The alarm on the container chiller can also operate properly."

The performance qualification of Container Chiller 2 was conducted using door-opening studies, with the door opened ten times every 10 minutes, a holding time study to measure the time required for the temperature to exceed the specified range, and a recovery time study to determine the time needed for the temperature to return to the specified range after deviation. Based on the results of the door-opening study, the minimum temperature in Container Chiller 2 at the coldest point was 4.4°C, and the maximum temperature at the warmest point reached 7.6°C. The results indicate that the components of the operational and performance qualification protocols include objectives, scope, responsibilities, machine descriptions, and testing procedures for the stand freezer. Operational qualification testing includes powering on/off, temperature adjustment, cold distribution in empty condition, and defrost operation. Furthermore, performance qualification testing involves cold distribution testing with load on the freezer."

Hasil pengujian kualifikasi kinerja freezer yang telah dilakukan, ditemukan beberapa data distribusi suhu dari termometer yang berada diluar rentang suhu yang dipersyaratkan yaitu -25°C s/d -15°C. Suhu yang didapatkan cenderung lebih dingin dari rentang suhu yang dipersyaratkan. Walaupun begitu pendistribusian suhu pada kinerja freezer terbilang sudah merata sehingga kualifikasi kinerja ini dinyatakan lulus. Selain itu, tidak ada ice gel yang mencair pada pengaturan suhu -18°C sehingga freezer tersebut dapat digunakan lebih lanjut untuk membuat material pendingin (ice gel) dengan pengaturan suhu -18°C. Hasil laporan ini dapat dijadikan sebagai sumber informasi dalam melakukan proses Kualifikasi Kinerja (KK) untuk fasilitas pendingin seperti freezer oleh pedagang besar farmasi atau distributor farmasi.

The research was carried out by following freezer performance qualification testing procedures following the applicable protocols at PT Enseval Putera Mega Trading DC 1 Jakarta. As a result of the freezer performance qualification tests that had been carried out, it was found that several temperature distribution data from the thermometer were outside the required temperature range, namely -25°C to -15°C. The temperature obtained tends to be colder than the required temperature range. However, the temperature distribution in the freezer performance is fairly even so this performance qualification is declared passed. Apart from that, no ice gel melts at a temperature setting of -18°C so that the freezer can be further used to make cooling material (ice gel) with a temperature setting of -18°C. The results of this report can be used as a source of information in carrying out the Performance Qualification (PQ) process for cooling facilities such as freezers by pharmaceutical wholesalers or pharmaceutical distributors."

Kesenjangan dapat terjadi pada proses ini ketika pelaksanaan teknis yang terjadi di lapangan memiliki sedikit banyak perbedaan dengan yang tercantum dalam pedoman. Oleh karena itu, perlu dilakukan Gap assessment sebagai alat penilaian untuk mengevaluasi proses distribusi tersebut. Tugas khusus ini disusun dengan melakukan proses pengamatan atau observasi pada proses pelaksanaan distribusi di PT. Enseval Putera Megatrading (EPM) Cabang Jakarta 1 terhadap penerapan Cara Distribusi Obat yang Baik (CDOB). Selanjutnya, dilakukan penilaian terhadap kesesuaian penerapan atau implementasi CDOB berdasarkan daftar aspek penerapan CDOB. Bedasarkan daftar periksa yang digunakan, terdapat 121 aspek detail terkait CDOB pada proses distribusi oleh PT EPM Cabang Jakarta 1 yang perlu dinilai kesesuaian dengan yang terlaksana. Namun, terdapat 6 aspek yang tidak terpenuhi. Semua aspek yang tidak sesuai dengan ketentuan diharapkan dapat dibuat aksi perbaikan dan pencegahannya.

Gaps can occur in this process when the technical implementation that occurs in the field is more or less different from what is stated in the guidelines. Therefore, it is necessary to carry out a Gap assessment as an assessment tool to evaluate the distribution process. This special task is prepared by carrying out an observation process or observations on the distribution implementation process at PT. Enseval Putera Megatrading (EPM) Jakarta Branch 1 regarding the implementation of Good Distribution Practice (GDP). Next, an assessment is carried out on the suitability of the application or implementation of GDP based on the list of GDP implementation aspects. Based on the checklist used, there are 121 detailed aspects related to GDP in the distribution process by PT EPM Jakarta Branch 1 which need to be assessed for suitability with what is being implemented. However, 6 aspects are not fulfilled. It is hoped that all aspects that are not by the provisions can be taken to corrective and preventive actions"
