"Pedagang Besar Farmasi atau PBF merupakan salah satu unit terpenting dalam kegiatan penyaluran sediaan farmasi ke fasilitas pelayanan kesehatan agar dapat sampai ke tangan masyarakat. PBF sebagai sarana distribusi kefarmasian memiliki tanggung jawab penuh dalam menjaga kualitas dan mutu produk yang didistribusikan khususnya untuk produk rantai dingin atau Cold Chain Product (CCP). Oleh karena itu, untuk memastikan peralatan penyimpanan yang digunakan bekerja sesuai dengan kriteria yang telah ditentukan maka dilakukan pengujian kualifikasi di PT Enseval Putera Megatrading DC1 Jakarta. Tujuan penelitian dilakukan untuk memastikan Upright Medical Freezer yang akan digunakan dapat memenuhi spesifikasi yang telah ditentukan dan dapat digunakan untuk menyimpan frozen product dengan melakukan Kualifikasi Instalasi, Kualifikasi Operasional dan penentuan titik pemantauan suhu. Penelitian dilaksanakan pada tanggal 11 September sampai dengan 22 September 2023. Hasil dari Kualifikasi Instalasi dan Kualifikasi Opeasional yang dilakukan pada Upright Medical Freezer DC 1 Jakarta telah memenuhi kriteria penerimaan tanpa ada penyimpangan dengan nilai rata-rata suhu yaitu -18,91 pada setting suhu -22°C. Titik pemantauan suhu akan diletakkan pada posisi 5 karena memiliki suhu maksimal dan rata-rata paling rendah yaitu -13.20°C dan -17.23°C.

Pharmaceutical Wholesalers or PBF are one of the most important units in the distribution of pharmaceutical supplies to health service facilities so that they can reach the hands of the public. PBF as a pharmaceutical distribution facility has full responsibility for maintaining the quality and quality of the products distributed, especially for cold chain products (CCP). Therefore, to ensure that the storage equipment used works according to predetermined criteria, qualification testing is carried out at PT Enseval Putera Megatrading DC 1 Jakarta. The aim of the research was to ensure that the Upright Medical Freezer that will be used can meet the specified specifications and can be used to store frozen products by carrying out Installation Qualification, Operational Qualification and determining temperature monitoring points. The research was carried out from 11 September to 22 September 2023. The results of the Installation Qualification and Operational Qualification carried out on the Upright Medical Freezer DC 1 Jakarta met the acceptance criteria without any deviation with an average temperature value of -18.91 at the temperature setting - 22°C. The temperature monitoring point will be placed in position 5 because it has the lowest maximum and average temperatures, namely -13.20°C and -17.23°C."

Oleh sebab itu, dibutuhkan fasilitas penyimpanan yang memadai. Container chiller adalah salah satu fasilitas penyimpanan untuk produk rantai dingin yang digunakan di PT. Enseval Putera Megatrading. Menurut CDOB (2020), fasilitas penyimpanan chiller / cold room / freezer harus dikualifikasi terlebih dahulu saat awal penggunaan. Selain itu, untuk memastikan bahwa container chiller dapat beroperasi pada suhu yang sesuai dengan persyaratan maka perlu dilakukan kegiatan pemetaan suhu. Pada makalah ini dibahas mengenai kualifikasi operasional serta pemetaan suhu terhadap container chiller di PT. Enseval Putera Megatrading. Pemetaan suhu dilakukan saat container chiller belum terisi oleh produk rantai dingin yang akan disimpan. Hasil pemetaan suhu menunjukkan bahwa kondisi suhu pada container chiller yang diuji memenuhi kriteria yang dibutuhkan dalam penyimpanan produk rantai dingin karena masih berada pada rentang yang dipersyaratkan yaitu 2-8oC, dengan nilai MKT (Mean Kinetic Temperature) tertinggi dan terendah secara berturut-turut adalah 5,14 (Upper) dan 4,37 (Lower). Suhu tertinggi terdapat pada titik 8, sedangkan suhu terendah berada pada titik 11. Hasil kualifikasi operasional menunjukkan bahwa sistem refrigerasi container chiller dapat berfungsi dengan baik. Alarm pada container chiller juga dapat beroperasi dengan baik.

This paper discusses operational qualifications and temperature mapping of container chillers at PT. Enseval Putera Megatrading. Temperature mapping is carried out when the chiller container is not yet filled with the cold chain products to be stored. The temperature mapping results show that the temperature conditions in the chiller container tested meet the criteria required for storing cold chain products because they are still within the required range, namely 2-8oC, with the highest and lowest MKT (Mean Kinetic Temperature) values respectively being 5,14 (Upper) and 4,37 (Lower). The operational qualification results show that the container chiller refrigeration system can function well. The alarm on the container chiller can also operate properly."

PT Enseval Putera Megatrading Tbk. merupakan salah satu PBF di Indonesia yang mendistribusikan sediaan farmasi, alat kesehatan, dan barang konsumsi. Suhu dalam proses pendistribusian dapat mempengaruhi mutu sediaan farmasi akibat terjadinya perubahan secara fisik atau kimia pada sediaan yang menyebabkan penurunan potensi zat aktif dan aktivitas farmakologinya. Salah satu cara dalam memenuhi standar CDOB adalah dengan rutin dilakukan kualifikasi kinerja. Dengan meletakan thermometer data logger sesuai dengan titik-titik uji yang telah ditentukan, proses perekaman data (setiap 10 menit) dan akhiri setelah 24 jam. Suhu pendistribusian produk pada mobil berpendingin dengan suhu 2-8°C belum memenuhi syarat keberterimaan dan untuk mobil berpendingin dengan suhu 15-25°C memenuhi syarat keberterimaan.

Temperature in the distribution process can affect the quality of pharmaceutical preparations due to physical or chemical changes in the preparations that cause a decrease in the potential of active substances and their pharmacological activity. One way to meet CDOB standards is to routinely carry out performance qualifications. Berdasarkan hasil studi buka tutup pintu, suhu minimum Container Chiller 2 pada titik terdingin yaitu 4,4°C, suhu maksimum pada titik terpanas mencapai 7,6°C. Container Chiller 2 memiliki holding time selama 47 menit dan nilai recovery time selama 13 menit. Hasil kualifikasi kinerja menunjukkan bahwa kinerja Container Chiller 2 di PT Enseval Putera Megatrading Tbk cabang Bekasi telah sesuai dengan spesifikasi yang ditentukan, yaitu masih berada pada rentang 2°−8°C.

Dari penelitian ini dihasilkan Container chiller 1 dan 2 PT Enseval Putera Megatrading cabang Bekasi memenuhi persyaratan suhu yakni diantara 2-8o C dan titik kritisnya berada pada posisi terpanas dan terdingin di container chiller 1 terletak pada titik 3 dan 19, sedangkan pada container chiller 2 titik terpanas dan terdingin terletak pada titik 10 dan 5. Berdasarkan hasil penelitian tersebut sebaiknya limit suhu diatur pada suhu atas 7o C dan bawah 3oC untuk penanggulangan lebih cepat jika suhu berada diluar standar dan sebaiknya dalam proses penyimpanan dan pengiriman sediaan-sediaan cold chain product perlu dipantau dengan baik, terutama pada vaksin-vaksin yang memiliki indikator VVM yang mempermudah pengecekkan kestabilan obat.

This research aims to determine the location of the highest and lowest temperatures as well as critical temperature points in chiller containers which are checked by placing temperature data loggers at various points and seeing whether the available chiller containers meet the requirements and standards for storing cold chain products or not. This research was carried out observationally for 3 days, recording temperature data with a temperature data logger every 10 minutes, and then pulling data from the thermometer data logger using the Testo Comfort Software Basic 5.0 application. From this research it was produced that Container chiller 1 and 2 of PT Enseval Putera Megatrading Bekasi branch met the temperature requirements, namely between 2-8o C and the critical points were at the hottest and coldest positions. Container chiller 1 was at points 3 and 19, while container chiller 2 was at point The hottest and coldest are located at points 10 and 5. Based on the results of this research, the temperature limit should be set at an upper temperature of 7o C and a lower temperature of 3o C for quicker response if the temperature is outside the standard and it is best to monitor the process of storing and sending cold chain products. Well, especially for vaccines that have a VVM indicator which makes it easier to check drug stability."

Penelitian ini bertujuan untuk memastikan suhu pada ruang penyimpanan produk di Gudang A PT. Enseval Putera Megatrading sesuai dengan standar yang ditetapkan dan menganalisis hasil kalibrasi Data Logger Testo Saveris sebagai pemantau suhu otomatis. Penelitian dilakukan dengan metode kalibrasi menggunakan perangkat Testo 735-2 pada berbagai titik penyimpanan di gudang, termasuk Chiller FA, Chiller FB, Chiller FD, dan Cool Room A. Data suhu yang diperoleh dibandingkan dengan standar acuan yang telah dikalibrasi. Calibration of automatic temperature monitoring devices is crucial in the pharmaceutical industry to ensure optimal drug storage conditions. This study aims to ensure that the storage temperature of products in Warehouse A of PT. Enseval Putera Megatrading meets established standards and to analyze the calibration results of the Testo Saveris Data Logger as an automatic temperature monitor. The study was conducted using the Testo 735-2 device for calibration at various storage points in the warehouse, including Chiller FA, Chiller FB, Chiller FD, and Cool Room A. The obtained temperature data were compared with calibrated reference standards. The calibration results show that the Testo Saveris Data Logger provides accurate and consistent temperature readings at all tested storage points. This device is effective in monitoring and maintaining temperatures according to the set parameters. Implementing the Testo Saveris Data Logger in Warehouse A of PT. Enseval Putera Megatrading has proven to enhance the quality of pharmaceutical distribution management, reduce the risk of product damage due to incorrect temperatures, and ensure product safety until it reaches consumers. Periodic calibration is recommended to maintain the accuracy of the device."

Laporan ini akan berfokus terhadap pelaksanaan Kualifikasi Instalasi dan Kualifikasi Operasional terhadap salah satu mesin Freezer yang akan digunakan untuk membekukan ice gel.
