Abstrak :
Gamma oryzanol dalam Rice Bran Oil (RBO) telah diteliti melindungi mukosa dengan menghambat sekresi asam dan bertindak sebagai prekusor pada sintesis prostaglandin sehingga diduga berpotensi sebagai gastroprotektif. Penelitian ini bertujuan untuk mendapatkan formulasi mikrokapsul RBO dengan kadar γ-oryzanol tertinggi dan untuk mengetahui efek gastroprotektif mikrokapsul RBO pada tikus yang diinduksi indometasin (80 mg/kg bb). Response Surface Metodology (RSM) Box-Behnken dengan 3 faktor digunakan untuk mengoptimasi kondisi pembuatan mikrokapsul. Dilakukan karakterisasi terhadap kadar tertinggi γ-oryzanol, Scanning Electron Microscope (SEM), particle size distribution, swelling ratio, efisiensi penjerapan, dan disolusi in vitro. Uji aktivitas gastroprotektif dibagi menjadi 7 kelompok yang diberikan aquades, indometasin (80 mg/kg bb), omeprazole 40 mg/kg bb, mikrokapsul RBO (160 mg/200 g bb, 320 mg/200 g bb) dan RBO (0.1 ml/200 g bb; 0.2 ml/200 g bb) secara oral selama 7 hari. Pengamatan dilakukan pada % inhibsi ulkus, derajat keasaman lambung, determinasi mukus serta gambaran histopatologi lambung tikus. Hasil optimasi RSM menunjukkan formula R 11 memberikan kadar γ-oryzanol paling tinggi dengan perbandingan 1,5% natrium alginat:RBO:emulgator (60%:30%:10%) sebesar 0,75%. Hasil SEM menunjukkan permukaan yang berpori dengan distribusi ukuran partikel rata-rata 647,013±41,68 µm. Hasil % swelling ratio tertinggi pada medium HCl sebesar 44,43%±1,01 pada menit ke 60 dan pada medium aquades sebesar 191,74%±1,85 pada menit ke 240. Hasil efisiensi penjerapan tertinggi sebesar 2,48% dan hasil uji disolusi menunjukkan jumlah γ-oryzanol yang terdisolusi sebesar 9,56 % pada medium HCl pH 1,2 dan 50,65% pada medium dapar fosfat. Hasil uji aktivitas gastroprotektif menunjukkan pemberian mikrokapsul RBO dan RBO dapat menaikkan % inhbisi ulkus secara signifikan dibandingkan dengan kontrol negatif (p <0.05). Hasil pemeriksaan derajat keasaman lambung menunjukkan mikrokapsul RBO dapat menurunkan derajat keasaman lambung (1,7919 µek/200 g bb). Hasil determinasi mukus menunjukkan lambung tikus yang diberikan mikrokapsul RBO memiliki ketebalan mukus lebih tinggi (0,89586/g bb) jika dibandingkan dengan kelompok negatif. Analisis lebih lanjut pada evaluasi histopatologi menunjukkan peningkatan proteksi dilihat dari rendahnya edema, erosi hemoragik, kongesti kapiler dan infiltrasi leukosit pada submukosa. Hasil penelitian menunjukkan bahwa Mikrokapsul RBO berpotensi sebagai gastroprotektif pada tikus yang diinduksi indometasin. ...... Gamma oryzanol content in Rice Bran Oil (RBO) has been studied to protect gastric mucosa by inhibition of acid secretion that stimulated by histamine receptor and also act as a precursor to prostaglandin synthesis. The objective of this research was to obtained the formulation of RBO microcapsules with highest γ-oryzanol content and to evaluated the gastroprotective effect of RBO microcapsules on indomethacin-induced gastric ulcers in rats. The Response Surface Metodology (RSM) Box-Behnken design on three factors was used to optimize the microencapsulation conditions. Gamma oryzanol content, Scanning Electron Microsope (SEM), particle size distribution, swelling ratio, entrapment efficiency and in vitro dissolution was also characterized. The gastroprotective study was performed on 7 groups who are administered orally the treatment of aquadest, indomethacin (80 mg/kg BW), omeprazole (40 mg/kg BW), RBO microcapsules (160 mg/200 g BW and 320 mg/200 g BW) and RBO (0,1 ml/200 g BW and 0,2 ml/200 g BW) for seven days. The effect on % inhibition, gastric content, determination of mucous stomach and histopathological analysis were also evaluated. The optimization result was on formula R 11 by comparison of 1,5% sodium alginate:RBO:emulgator (60%:30%:10%) was 0,75%. The SEM shown porous surface and the average of particle distribution was 647±68 µm. The highest swelling ratio was 44,43%±1,01 in HCl medium and 191,74%±1,85 in aquadest. The highest entrapment efficiency was 2,48% and the in vitro dissolution was 9,56% in simulated gastric fluid and 50,65% in simulated intestine fluid. The gastroprotective result showed that RBO microcapsules treatment could significantly reduced ulcer index in indomethacininduced gastric ulcers compared with negative control (p < 0.05). The RBO microcapsules can reduce gastric content (1,7919 µek/200 g BW) and also highest determination of mucous (0,89586/g BW) compared with negative control. Further evaluation on HE revealed higher protection characterized by lower edema,capiler congestion, mild hemorrhagic erosion and lower leucocyte infiltration in submucosa. This research shown that RBO microcapsules have potential to be gastroprotective agent by indomethacin-induced gastric ulcers in rats.

Latar belakang. Individu dewasa di masyarakat menunjukkan perubahan pola makan dan kurang aktivitas sehingga berisiko untuk menderita hiperkolesterolemia dan obesitas. Hiperkolesterolemia dapat diatasi dengan terapi nutrisi. Minyak bekatul mengandung zat aktif yang bekerja secara sinergis dan telah terbukti dari penelitian sebelumnya berperan dalam pengendalian lipid yaitu gamma-oryzanol, fitosterol, dan derivat vitamin E (tokotrienol dan tokoferol). Penelitian ini bertujuan untuk menilai perbaikan profil lipid pada pemberian minyak bekatul dengan jumlah yang berbeda tanpa merubah pola makan subyek. Metode. Uji klinis, desain paralel, alokasi acak selama 4 minggu pada laki-laki usia 19-55 tahun, kolesterol total 200-300 mg/dl, dan IMT 20-30 kg/m2. Subyek diambil secara konsekutif dan dibagi menjadi kelompok 45 ml/hari dan kelompok 15 ml/hari minyak bekatul. Sebelum perlakuan dilakukan wawancara data demografi, aktifitas fisik dan pemeriksaan antropometri. Asupan makan dinilai sebelum dan setelah perlakuan. Pemeriksaan laboratorium profil lipid dilakukan sebelum dan setelah perlakuan 4 minggu. Hasil. Dari total 20 subyek (10 subyek kelompok 45 ml/hari dan 10 subyek kelompok 15 ml/hari) didapatkan karakteristik yang setara antara kedua kelompok menurut usia, tingkat pendidikan, status gizi, aktivitas fisik, kebiasaan merokok, riwayat hiperkolesterolemia keluarga, antropometri dan profil lipid. Asupan makanan meliputi asupan energi, karbohidrat, protein, lemak dan serat sebelum perlakuan tidak berbeda bermakna antara kelompok. Asupan lemak setelah perlakuan berbeda bermakna antara kedua kelompok dikarenakan perbedaan pemberian jumlah minyak. Setelah perlakuan selama 4 minggu, didapatkan penurunan kolesterol total secara statistik berbeda bermakna antara kedua kelompok (p=0,049). Pada kelompok 45 ml/hari kadar kolesterol total turun sebanyak 14% dan pada kelompok 15 ml/hari terjadi penurunan kadar kolesterol total 7,8%. Penurunan LDL dan trigliserida serta peningkatan HDL secara statistik tidak berbeda bermakna antara dua kelompok (p >0,05). Pada penelitian ini tidak terjadi perubahan berat badan yang bermakna pada kedua kelompok. Kesimpulan. Konsumsi minyak bekatul 45 ml/hari menyebabkan perbaikan profil lipid yang lebih baik dibandingkan konsumsi minyak bekatul 15 ml.hari.

Background. Adult individuals in Indonesian society showed changes in diet pattern and lack of physical activity that increasing risk for hypercholesterolemia and obesity. Hypercholesterolemia would be treated with nutritional therapy. Rice bran oil contains active substances (gamma-oryzanol, phytosterols, and derivatives of vitamin E (tocotrienols and tocopherols) that work in synergy and have been proven on previous research controlling lipid profil. This study aimed to assess the lipid profile improvement in intake of rice bran oil with different amounts without changing the eating patterns of the subjects. Methods. It was parallel and randomized clinical trial for 4 weeks in male with 19-55 years of age, total cholesterol level 200-300 mg/dl, and BMI of 20−30 kg/m2. All subjects were recruited consecutively and classified into two groups that received 45 ml/day or 15 ml/day rice bran oil for 4 weeks. The demographic data interviews, physical activity and anthropometric examination were taken before intervention. Food intake were assessed before and after intervention. Laboratory test of lipid profile performed before and after 4 weeks of treatment. Results. A total of 20 subjects consisting of 10 subjects with 45 ml/day rice bran oil and 10 subjects with 15 ml/day had obtained similar characteristics in age, education level, nutritional status, physical activity, smoking, family history of hypercholesterolemia, BMI and lipid profiles. Food intake includes intake of energy, carbohydrate, protein, fat and fiber before treatment did not significantly difference between two groups. Fat intake after treatment was significantly different between the two groups due to differences in the amount of oil. After 4 weeks treatment, there was a decrease in total cholesterol significantly different between the two groups (p = 0,049). Penggunaan bahan alam sebagai produk perawatan kulit sudah dilakukan oleh wanita Indonesia secara turun temurun, seperti penggunaan lulur bagi wanita Jawa dan bedak dingin bagi wanita Kalimantan yang berbahan dasar beras. Bekatul, lapisan terluar dari beras, memiliki potensi yang masih dapat dikembangkan salah satunya sebagai pencerah kulit. Tujuan dari penelitian ini yaitu untuk mengetahui manfaat ekstrak etanol bekatul beras hitam (Oryza sativa L. indica) sebagai pencerah kulit dalam bentuk sediaan losion. Pada penelitian ini, kadar gamma oryzanol ditentukan menggunakan KCKT (Kromatografi Cepat Kinerja Tinggi). Penetapan total phenolic compound (TPC) atau kadar fenol total dilakukan menggunakan reagen Folin-Ciocalteu dan dibaca serapannya oleh microplate reader pada panjang gelombang 720 nm. Sedangkan uji penghambatan in vitro enzim tirosinase dibaca serapannya oleh microplate reader pada panjang gelombang 490 nm. Ekstrak etanol bekatul beras hitam kemudian diformulasikan dalam losion minyak dalam air untuk dilakukan uji manfaat secara in vivo. Kadar gamma oryzanol dan fenol total pada ekstrak bekatul beras hitam secara berturut-turut yakni sebesar sebesar 118,57 mg/g dan 175,48 mg/g. Ekstrak etanol bekatul beras hitam menghambat enzim tirosinase secara in vitro dengan nilai IC50 sebesar 74,8 µg/ml. Losion berisi ekstrak diaplikasikan pada lengan relawan (n=34), dan pada lengan lain diaplikasikan losion kontrol. Analisis data dilakukan menggunakan aplikasi GraphPad Prism 8.3.0, data diuji menggunakan t-test berpasangan dan didapatkan hasil penurunan secara signifikan pada indeks melanin (p<0,0001) dan indeks eritema (p<0,0001) pada lengan yang dioleskan losion berisi ekstrak bekatul beras hitam. Kesimpulannya, losion mengandung ekstrak bekatul beras hitam lebih efektif dalam mencerahkan kulit dibanding dengan losion kontrol. ......Nature based skincare has been used empirically by Indonesian women. Javanese women apply rice based traditional bodycare called lulur, while Kalimantan women apply rice based face care called bedak dingin. Rice bran, the outer layer of rice grain, has potential to be utilized as skin lightening. The aim of this study is to determine the efficacy of lotion containing black rice bran (Oryza sativa L. indica) extract as skin lightening. In this study, high performance liquid chromatography was performed to measure gamma oryzanol content. Folin-Ciocalteu reagent was added to determine total phenolic content of black rice bran (Oryza sativa L. indica) and measured under microplate reader wavelength 720 nm. In vitro tyrosinase inhibitor was measured under microplate reader wavelength 490 nm and the results was simbolized with IC50. Gamma oryzanol content and total phenolic content in black rice bran (Oryza sativa L. indica) ethanolic extract were 118,57 mg/g and 175,48 mg/g respectively. Ethanolic extract had potential to inhibit tirosinase enzyme in vitro with IC50 of 74,8 µg/ml. Black rice bran (Oryza sativa L. indica) ethanolic extract was formulated into oil in water (o/w) lotion to be tested in vivo. 34 women were applying lotion containing black rice bran extract at one side forearm and base placebo lotion as control at the other side forearm. The results were tested with paired t-test by GrapdPad Prism 8.3.0 application. There was significant decreasing of melanine index and erythema index with both p value <0,0001 in forearm with lotion containing black rice bran ethanolic extract. As the resume, lotion containing black rice bran extract is more effective to be applied as skin brightening than placebo lotion.

Dedak Padi Oryza sativa L. adalah produk sampingan hasil penggilingan beras yang mengandung tokoferol, tokotrienol, ?-oryzanol, dan senyawa bioaktif lainnya. ?-Oryzanol merupakan salah satu komponen dari minyak dedak padi yang berfungsi sebagai antioksidan, antiinflamasi, antineoplastik, hipolipidemia, dan berpotensi dikembangkan dalam bidang obat-obatan, kosmetik dan produk nutrisi. Penelitian ini bertujuan mengoptimisasi kondisi ?-oryzanol dengan IL- MAE (Ionic Liquid-Microwave Assisted Extraction) dan aktivitas penghambatan radikal bebas dengan metode DPPH 1,1 difenil-2-pIkrilhidrazil . Dedak padi diekstraksi menggunakan MAE Microwave Assisted Extraction dengan pelarut cairan ionik [Bmim]BF4. Kondisi ektraksi dibuat dalam empat parameter yaitu konsentrasi pelarut, rasio pelarut banding sampel, waktu ekstraksi, dan daya MAE. Proses optimasi dilakukan dengan bantuan RSM Response Surface Methodology . Hasil yang didapat yaitu kondisi optimal untuk memperoleh kadar ?-oryzanol adalah pada run 3 dengan kondisi konsentrasi pelarut 0,7 M dengan rasio sampel banding pelarut 1 : 15 gr/mL , waktu ekstraksi 10 menit dan daya MAE 30 Dan untuk aktivitas penghambatan diperoleh kondisi optimal pada run 1 dengan konsentrasi pelarut 0,7 M, rasio sampel banding pelarut 1 : 15 gr/mL , waktu ekstraksi 15 menit dan daya MAE 10 . Berdasarkan penelitian dapat disimpulkan kadar ?-oryzanol yang didapat pada kondisi optimum adalah 0,41 mg/g indeks desirability 0,876 . serta persen penghambatan yang didapat pada kondisi optimum adalah 31,14 indeks desirability 0,780 . Hasil menunjukkan IL-MAE sebagai metode ekstraksi yang cocok untuk mengekstraksi bahan alam. ......Rice Oryza sativa L. bran is a by product of rice milling process that contain tocopherol, tocotrienol, oryzanol and others bioactive compound. Oryzanol is one of the components of rice bran oil which have the function as antioxidant, antiinflammatory, antineoplastic, hypolipidemia and potential to be used in pharmaceuticals, cosmoceutical and nutraceutical. This study aimed to optimization condition of Oryzanol using IL MAE Ionic Liquid Microwave Assisted Extraction and the Inhibition activity with DPPH 1,1 diphenyl 2 pycrilhidrazil . Rice bran was extracted by MAE Microwave Assisted Extraction using Ionic Liquid Bmim BF4 as the solvent. This extraction used four parameters to determine optimum condition, including concentration, solid ratio, times, and power. The result of the optimization was processed using Response Surface Methodology RSM in Design Expert 10.03. The optimum condition of oryzanol content was found in run 3. The conditions was the IL concentration 0.7 M, solid liquid ratio 1 15 gr mL , extraction time of 10 min, and extraction power of 30 beside that, the result of free radical scavenging activity showed that the optimum condition was found in run 1. The optimum condition was in the IL concentration 0.7 M, solid liquid ratio 1 15 gr mL , extraction time of 15 min, extraction power of 10 . Based on this research, it can be concluded that the levels of oryzanol in optimum condition was 0.41 mg g desirability index of 0.876 and the optimum inhibition percentage of free radical scavenging activity was 31.14 desirability index 0.780 . The result showed that IL MAE was suitable extraction s method of natural product.

ABSTRACT
The study investigated the soxhlet extraction of Leum Pua rice bran oil (Oryza sativa L.) using 12 hours of extraction time, with comparison of product with hexane and ethanol as solvent. The chemical properties and gamma-oryzanol content in the rice bran oil were investigated. The results showed that the highest yield of rice bran oil was obtained from extraction using 70:30 %v/v ethanol:water solvent with percent yield of 21.89±0.35 g (mean+SD)/100g of dry rice bran) and maximum amount of gamma-oryzanol of 0.4647 mg/L of rice bran oil. The chemical properties and gamma-oryzanol content of Leum Pua rice bran oil indicates acceptable quality of oil when stored for a long time causing foul odour due to lipid peroxidation.

ABSTRAK
Rice bran oil soapstock (RBOS) is a by-product from the chemical refining process of rice bran oil production. It contains a large amount of -oryzanol. The main objective of this study was to investigate the amount of -oryzanol obtained by solvent extraction using soxhlet apparatus. RBOS was saponified and was then dehydrated and extracted with ethyl acetate. The optimum conditions were determined using response surface methodology (RSM) with a Box-Behnken experimental design (BBD). BBD was used to investigate the effects of three independent variables, namely solid to solvent ratio (w/v), extraction temperature (C), and extraction time (h). The results showed that the most suitable conditions for the extraction of RBOS that can provide the highest yield of -oryzanol (9.04% dry basis) were 1:12 solid to solvent ratio, 70.1C extraction temperature and 7.26 h extraction time.
