Abstrak :
Kanker kolorektal adalah penyakit neoplasma ganas yang tumbuh dan berkembang pada saluran usus besar dan atau rektum. Terapi anti EGFR menggunakan agen biologis antibodi monoklonal cetuximab dan panitumumab diketahui memberikan tingkat penyembuhan yang baik pada pasien kanker kolorektal. Pasien dengan mutasi pada gen NRAS dan KRAS cenderung resisten terhadap terapi anti EGFR, sehingga penting dilakukan pemeriksaan kedua gen tersebut sebelum pemberian terapi. Pemeriksaan gen NRAS belum tersedia di Indonesia karena minimnya data mengenai mutasi gen tersebut pada populasi Indonesia. Penelitian dilakukan untuk mengetahui profil mutasi pasien gen NRAS pada 58 sampel pasien kanker kolorektal di Jakarta. Pemeriksaan mutasi dilakukan pada exon 2(codon 12 & 13) dan exon 3 (codon 61) gen NRAS menggunakan metode sekuensing. Analisis elektroferogram sekuensing menunjukan mutasi gen NRAS ditemukan pada 6,9% (n=58) sampel uji. Hasil uji statistik fischer exact test dua arah menunjukan tidak adanya asosiasi mutasi gen dengan kelompok usia pasien dan jenis kelamin. Gen NRAS ditemukan termutasi pada codon 12 (1,7 %) dan codon 61 (5.2%). Tidak ditemukan adanya mutasi pada codon 13 gen NRAS.
Colorectal cancer is a neoplasm disease that arise in inner lining of colon or rectum. Anti EGFR therapy such as cetuximab and panitumumab were widely used to suppress metastases colorectal cancer in patient and decided as gold standard therapy. Mutation either KRAS or NRAS gene will reduced effectifity of anti EGFR therapy, hence genotyping of KRAS and NRAS gene must be executed before. NRAS genotyping test not yet available in Indonesia due to lack of information about this gene. This study was subjected to understanding profile of NRAS gene mutation in Jakartans colorectal cancer patient. Mutation screening was perform by sequencing method, notably exon 2 (codon 12&13) and exon 3 (codon 61). Electropherograms analysis shows that NRAS mutation found in 6,9% samples (n=58). NRAS mutation found in codon 12 (1,7%), codon 61 (5,2%), and there was no mutation found in codon 13. Fischer exact test statistical analysis summarized that there was no significant association of NRAS mutation with both sex and age.

ABSTRAK

Telah dilakukan penelitian yang bertujuan mengetahui potensi hepatoprotektif madu PS terhadap kadar alkali fosfatase (ALP) mencit (Mus musculus L.) jantan galur DDY. Dua puluh empat ekor mencit jantan dibagi ke dalam 4 kelompok hewan uji, yaitu kelompok kontrol normal (KK1) yang diberikan akuades dan minyak kelapa; kelompok kontrol perlakuan (KK2) yang diberikan akuades dan CCl4; serta 2 kelompok perlakuan (KP1 dan KP2) yang diberikan madu PS 10% dan 20% selama 14 hari berturut-turut, kemudian CCl4 2 jam setelah pemberian madu terakhir. Darah diambil 24 jam setelah injeksi CCl4. Kadar ALP diukur dengan metode kolorimetri. Hasil uji anova satu arah (P<0,05) menunjukkan adanya pengaruh nyata pemberian madu PS terhadap kadar ALP semua hewan uji. Dibandingkan kadar ALP KK2, kadar ALP KP1 lebih rendah 30,5% dan KP2 lebih rendah 52,9%. Namun, uji LSD (P<0,05) menunjukkan hanya kadar ALP KP2 yang tidak berbeda nyata dengan KK1. Berdasarkan hasil tersebut, disimpulkan bahwa potensi hepatoprotektif madu PS 20% lebih besar dibandingkan madu PS 10%.


ABSTRACT

The study has been conducted to know the hepatoprotective potency of PS honey administration on male-DDY mice’s alkaline phosphatase level of blood plasma. Twenty four male mice were divided into four groups, namely normal control group (KK1) which was administered with aquadest and coconut oil; treatment control group (KK2) which was administered with aquadest and CCl4; and two treatment groups which was administered with PS honey 10% (KP1) and 20% (KP2) within 14 consecutive days and three groups (KK2, KP1,and KP2) were injected with CCl4 on the 14th day. Alkaline phosphatase was measured based on colorimetry method. One-way anova test (P<0,05) showed that alkaline phosphatase levels were significantly different. Compared with KK2, the alkaline phosphatase levels of KP1 and KP2 were 30,5% and 52,9% lesser than KK2, consecutively. However, LSD test (P<0,05) showed that only alkaline phosphatase level of KP2 was not significantly different. In conclusion, dose 20% of PS honey is more potential on hepatoprotective than those of 10%.

Telah dilakukan penelitian yang bertujuan untuk mengetahui potensi hepatoprotektif madu PS (Pollen Substitute) terhadap kadar SGPT dan SGOT pada mencit (Mus musculus L.) jantan galur DDY yang diinduksi karbon tetraklorida (CCl4). Dua puluh empat ekor mencit jantan dibagi ke dalam 4 kelompok hewan uji, yaitu kelompok kontrol normal (KK1) yang diberi pakan standar dan akuades, kelompok kontrol perlakuan (KK2) yang diberi akuades dan larutan karbon tetraklorida, dan dua kelompok perlakuan (KP1 dan KP2) yang diberi madu PS 10% dan 20% b/v dan larutan karbon tetraklorida. Hasil uji ANOVA (P< 0,05) menunjukkan adanya pengaruh nyata pemberian madu PS terhadap kadar SGPT dan SGOT mencit pada semua kelompok perlakuan. Hasil uji LSD (P<0,05) menunjukkan adanya perbedaan nyata kadar SGPT dan SGOT mencit antar kelompok perlakuan (KK1, KK2, KP1 dan KP2). Persentase penurunan kadar SGPT dan SGOT terbaik dicapai oleh KP1 yaitu sebesar 82.93% dan 85.25% mendekati kadar normal (mengacu pada KK1), jika dibandingkan dengan dosis 20% pada KP2. Dengan demikian, dosis optimum dicapai oleh KP1 yaitu sebesar 10%.
The objective of this study was to screen for the potential hepatoprotective role of PS (Pollen Substitute) honey administration on male DDY mice (Mus musculus L.)?s SGPT and SGOT level in blood serum. Twenty four mice were divided into four groups consisting of normal control group (KK1) which administrated with aqudes and coconut oil; treatment control group (KK2) which administrated with aquades and carbon tetrachloride; and two treatment groups (KP1 and KP2) which administrated with PS honey 10% and 20% also carbon tetrachloride, respectively, for 14 consecutive days. SGPT and SGOT level was measured in day 15 using SGPT and SGOT kit assay in laboratory. ANOVA one way test (P<0.05) shows that SGPT and SGOT level were significantly different between groups of animals. LSD test (P<0.05) shows that there is significantly different of SGPT and SGOT levels between KK1, KK2, KP1 and KP2. According to normal control group (KK1), the decrease percentage of SGPT and SGOT level of KP1 is better than KP2, that is 82.93% dan 85.25%. Its concluded that administration of PS honey reached its optimum dosage at 10%.

Telah dilakukan penelitian yang bertujuan untuk mengetahui pengaruh pemberian fortifikan NaFeEDTA dalam tepung tahu terhadap kadar zat besi plasma darah tikus (Rattus norvegicus L.) jantan galur Sprague- Dawley. Metode penelitian menggunakan Rancangan Acak Lengkap (RAL), terdiri atas 25 ekor tikus putih jantan yang dibagi ke dalam 5 kelompok perlakuan, yaitu KK1 yang diberi larutan CMC 0,5%; KK2 yang diberi CMC 0,5% dan suspensi tepung tahu tanpa fortifikan; dan KP1, KP 2, dan KP3 yang diberi CMC 0,5% dan tepung tahu dengan fortifikan NaFeEDTA dosis 1,35 mg Fe/ kgBB; 2,7 mg Fe/ kg BB; dan 5,4 mg Fe/ kgBB selama 21 hari. Pengambilan darah dilakukan pada hari ke-7, 14, dan 21. Darah ditentukan kadar Fe nya dengan AAS (Atomic Absorption Spectrophotometer). Hasil uji ANAVA satu arah (P < 0,05) menunjukkan pengaruh nyata pemberian fortifikan NaFeEDTA dalam tepung tahu terhadap kadar zat besi antar kelompok perlakuan. Hasil uji LSD (P < 0,05) menunjukkan bahwa terdapat perbedaan kadar zat besi (Fe) dalam darah yang nyata antara KK1 dengan semua kelompok perlakuan (KP1, KP2, dan KP3) dan KK2 dengan semua kelompok perlakuan pada t14. Hasil uji paired-samples T test (P < 0,05) menunjukkan terdapat perbedaan yang nyata kadar zat besi semua kelompok perlakuan pada t14 dan t21. Peningkatan kadar zat besi tertinggi terjadi pada KP3, yaitu sebesar 53,18 % di t14.
The effect of fortificant NaFeEDTA inserted in tofu flour intake on plasma iron concentration in male Sprague-Dawley rats (Rattus norvegicus L.) has been studied. Twenty five rats were divided into five groups, consist of normal contol group (KK1) which was administered with CMC 0,5%, treatment control group (KK2) which was administered with CMC 0,5% and tofu flour non fortificant, and three treatment groups wich was adminestered with tofu flour added with fortificant NaFeEDTA 1,35 mg Fe/kgbw (KP1); 2,7 mg Fe/kgbw (KP2); and 5,4 mg Fe/kgbw (KP3). All of the five groups were treated for 21 consecutive days. The plasma iron concentration was measured by Atomic Absorption Spectrophotometer (AAS). One way ANAVA test ( P < 0,05) showed significant effect of fortificant NaFeEDTA inserted in tofu flour intake on plasma iron concentration in all treatment groups. LSD test (P<0,05) showed that the plasma iron concentration significantly different between KK1 towards all treatment groups and KK2 towards all treatment groups. Paired-samples T Test (P < 0,05) showed that the plasma iron concentration significantly different in all treatment groups on t14 and t­21. The higest increase of plasma iron concentration was detected on KP3 at t14, whic is 53,18 %.

[Penicillium marneffei merupakan fungi patogen yang ditemukan di Asia Tenggara, khususnya Thailand. Penisiliosis dapat menyebabkan mikosis sistemik sehingga membahayakan nyawa penderita immunocompromised, khususnya penderita HIV/AIDS. Antifungi seperti Fluconazole dan Ketoconazole, digunakan untuk mengatasi infeksi P. marneffei. Akan tetapi, penggunaan antifungi secara jangka panjang dapat memicu kemungkinan munculnya mutan resisten P. marneffei. Resistensi pada fungi dapat dipengaruhi beberapa faktor, salah satunya, overekspresi transporter pengeluaran obat (drug efflux transporter). Mekanisme pompa pengeluaran obat diatur oleh berbagai transporter. Transporter yang paling umum diketahui ialah transporter ABC (ATP-binding-cassette) dan MFS (Major Facilitator Superfamily). Transporter ABC multidrug (MDR) pada P. marneffei telah dipelajari dengan baik, sedangkan transporter MFS MDR pada fungi tersebut, belum mendapatkan perhatian yang sama. Penelitian ini fokus pada satu transporter MFS MDR P. marneffei, yakni PMAA 067100, yang diekspresikan pada Saccharomyces cerevisiae ADΔ; sistem ekspresi yang sangat rentan terhadap berbagai macam antifungi. Pengamatan melalui mikroskop konfokal dan uji Disk Diffusion menunjukkan bahwa transporter PMAA 067100 terlokalisasi pada membran sel S. cerevisiae ADΔ dan resisten terhadap Fluconazole dan Terbinafine.;Penicillium marneffei has been known as a pathogenic fungi which is found in Southeast Asia, especially Thailand. The infection by this fungi recognized as Penicilliosis, that caused systemic mycosis, might be lethal in immunocompromised patient, specifically HIV/AIDS patient. Antifungal such as Fluconazole and Ketoconazole, had been used against P. marneffei infection. However, the long-term-use of antifungal might cause an emerging resistant strain of P. marneffei. The resistance phenomenon in fungi is caused by several factors, one of it is the overexpression of drug efflux transporter. Mechanism of this efflux pump is regulated by some of transporters such as ABC (ATP-binding-cassette) and MFS (Major Facilitator Superfamily) transporter. The ABC multidrug (MDR) transporter of P. marneffei has been studied well, yet the underrated MFS MDR transporter of the same fungi has not received the same attention. This study focus on one of P. marneffei MFS MDR transporter, known as PMAA 067100, which was expressed in Saccharomyces cerevisiae ADΔ; an expression system which is very susceptible to many kind of antifungal. Telah dilakukan penelitian mengenai kriopreservasi ikan koi Cyprinus carpio, Linnaeus 1758 menggunakan berbagai konsentrasi susu skim yang dikombinasikan dengan metanol 10. Tujuan penelitian adalah mengetahui pengaruh konsentrasi susu skim 0, 5, 10, 15, 20, 25 yang dikombinasikan dengan metanol 10 terhadap kualitas spermatozoa ikan koi 24 jam pascakriopreservasi. Hasil uji ANAVA satu faktor menunjukkan pemberian berbagai konsentrasi susu skim memiliki nilai rata-rata persentase motilitas, viabilitas, dan abnormalitas spermatozoa ikan koi 24 jam pascakriopreservasi yang berbeda nyata.
Cryopreservation of koi Cyprinus carpio, Linnaerus 1758 spermatozoa using various concentrations of skim milk combined with 10 methanol has been studied. The objective of the study was to know the effect of skim milk in various concentration on spermatozoa quality of koi Cyprinus carpio, Linnaeus 1758 postcryopreservation. The one factor ANOVA showed that various concentrations of skim milk had a significant difference.

Sindrom ovarium polikistik (SOPK) merupakan kelainan reproduksi yang ditandai dengan anovulasi, menstruasi tidak teratur, dan hirsutisme yang seringkali menyebabkan infertilitas. Meski etiologinya belum sepenuhnya dipahami, namun telah diketahui bahwa sebagian besar wanita dengan SOPK mengalami obesitas dengan prevalensi mencapai 40—80%. Obesitas merupakan kelebihan akumulasi lemak tubuh yang dicirikan dengan hipertrofi. Salah satu gen yang diduga terkait dengan obesitas adalah gen fat mass and obesity associated (FTO). Gen FTO menyebabkan peningkatkan nilai BMI. Penelitian ini bertujuan untuk mengetahui perbedaan ekspresi mRNA gen FTO dan korelasinya dengan BMI pada wanita SOPK dan normal dengan obesitas dan non-obesitas. Jaringan darah digunakan sebagai sumber mRNA yang diambil pada 30 wanita non obesitas, 30 wanita normal obesitas, 30 wanita SOPK non-obesitas, dan 30 wanita SOPK obesitas. Kuantitas ekspresi mRNA gen FTO ditentukan dengan menggunakan quantitative real-time PCR. Hasil penelitian menunjukkan terdapat perbedaan ekspresi mRNA gen FTO pada kelompok wanita SOPK dan normal dengan obesitas, serta tidak terdapat korelasi antara ekspresi mRNA gen FTO dengan BMI. Gen FTO merupakan gen yang bertanggung jawab terhadap obesitas akan tetapi tidak memiliki keterkaitan dengan sindrom ovarium polikistik, serta tidak memiliki korelasi dengan BMI ......Polycystic ovary syndrome (PCOS) is a reproductive disorder characterized by anovulation, irregular menstruation, and hirsutism which often causes infertility. Although the etiology is not fully understood, it is well known that most women with PCOS are obese with a prevalence of 40-80%. Obesity is an excess of body fat accumulation which is characterized by hypertrophy. Fat mass and obesity associated gene (FTO) is known to correlate with obesity. The study found that FTO gene causes an increase in the BMI. This reserach aim to determine the differences in FTO mRNA gene expression and its correlation with BMI in normal and PCOS woman with obesity and lean. This study used blood tissue as a source of mRNA taken in 30 normal lean woman, 30 normal obese women, 30 PCOS lean women, and 30 PCOS obese women. The quantity of FTO mRNA gene expression was determined using quantitative real-time PCR. The result shows that there is differences in FTO mRNA gene expression in PCOS and normal woman with obesity dan non-obesity. FTO mRNA expression in PCOS and normal obesity woman is higher than those in the PCOS and normal lean women, and there is no correlation between FTO gene mRNA expression and BMI. Thus, the FTO gene is a gene responsible for obesity but has no association with polycystic ovary syndrome, and does not have a correlation with BMI.

Telah dilakukan penelitian tentang pengaruh pemberian madu PS (pollen substitute) terhadap gambaran histologi pulau Langerhans pankreas pada mencit (Mus musculus L.) jantan galur DDY yang diinduksi aloksan. Sebanyak 24 ekor mencit dibagi dalam 4 kelompok 6 ulangan, yaitu: kelompok kontrol normal (KK1), kelompok kontrol diabetes (KK2), kelompok perlakuan pemberian madu PS 10% (KP1), dan kelompok perlakuan pemberian madu PS 20% (KP2). Pencekokan madu PS dilakukan setiap hari selama 14 hari berturut-turut. Pada hari ke-15, mencit dikorbankan, organ pankreas diisolasi dan dibuat sediaan histologi menggunakan metode parafin dengan pewarnaan Hematoksilin Eosin (HE). Sediaan diamati secara mikroskopik menggunakan mikroskop cahaya dan mikroproyektor. Data rerata diameter pulau Langerhans mencit dan jumlah sel β pankreas adalah sebagai berikut: KKI (115,03 ± 4,94) µm, (59,47 ± 2,08); KK2 (51,09 ± 8,39) µm, (15,24 ± 2,54); KP1 (106,70 ± 4,75) µm, (40,89 ± 2,33); KP2 (114,24 ± 10,85) µm, (46,78 ± 3,2). Hasil uji LSD (P < 0,05) menunjukkan terdapat perbedaan nyata antara kelompok perlakuan dengan kelompok kontrol KK2. Hal ini berarti bahwa pemberian madu PS (pollen substitute) memengaruhi rerata diameter pulau Langerhans dan jumlah sel β pankreas pada dosis 10% dan 20%.
ABSTRACT
The study was undertaken to assess the effect of PS (pollen substitute) honey intake on recovery diameter islet of Langerhans pancreas of alloxan-induced male-DDY mice (Mus musculus L.). Tweenty-four male mice were randomly devided into four groups, consisting of normal control group (KK1), treatment control group (KK2), two treatment groups (KP1 and KP2) which was administered with alloxan and PS honey in concentration of 10% and 20%, respectively. Treatments were carried out orally within 14 consecutive days. The mice were sacrified at day 15 (T15). Histology slides was made with paraffin method and stained with Haematoxyline Eosin (HE) and observed with microscope and microprojector. Mean of diameter of islet of Langerhans and pancreatic cell β number : KKI (115,03 ± 4,94) µm, (59,47 ± 2,08); KK2 (51,09 ±8,39) µm, (15,24 ± 2,54); KP1 (106,70 ± 4,75) µm, (40,89 ± 2,33); KP2 (114,24 ±10,85) µm, (46,78 ± 3,2). Least significant difference (LSD) (P < 0,05%) test showed a significant effect of treatment. The result demonstrated the potential beneficiary effect of PS (pollen substitute) Honey for recovery diameter of islet of Langerhans and pancreatic cell β number in concentration of 10% and 20%.

Telah dilakukan penelitian untuk mengetahui pengaruh pemberian infusa daun kluwih (Artocarpus camansi Blanco) terhadap penurunan kuantitas dan kualitas spermatozoa mencit (Mus musculus) jantan galur DDY. Sebanyak 24 ekor mencit dibagi kedalam 4 kelompok, yaitu: kelompok kontrol (KK), kelompok perlakuan yang diberikan infusa daun kluwih dengan dosis berturut-turut, yaitu 2,5; 5; dan 10 g/kg BB (KP1, KP2, dan KP3). Infusa daun kluwih diberikan selama 36 hari. Kemudian dilakukan analisis kuantitas dan kualitas spermatozoa. Data rerata jumlah spermatozoa per militer (x106) pada KK, KP1, KP2, dan KP3 berturutturut ialah (42,92± 3,28), (39,57± 2,08), (36,49± 2,73), dan (33,37± 1,26) spermatozoa per mililiter. Data rerata persentase motilitas spermatozoa pada KK, KP1, KP2, dan KP3 berturut-turut ialah (82,92 % ± 1,74) , (68,54 % ± 6,32), (61,23 % ± 7,13), dan (46,12 % ± 3,90). Data rerata persentase abnormalitas spermatozoa pada KK, KP1, KP2, dan KP3 berturut-turut ialah (37,63% ± 1,32), (52,24 % ± 0,95), (61,93 % ± 1,26), dan (68,83% ± 0,66). Hasil uji LSD (P < 0,05) menunjukkan terdapat perbedaan nyata antara kelompok perlakuan dengan kelompok kontrol KK. Hal tersebut menunjukkan bahwa pemberian infusa daun kluwih (Artocarpus camansi Blanco) berpengaruh terhadap kuantitas dan kualitas spermatozoa mencit jantan pada dosis 2,5; 5; dan 10 g/kg BB. ......The present study was done to determine the effect of Kluwih’s leaf’s infusion on the quantity and quality of spermatozoa of male mice DDY strain. 24 male mice have divided into 4 experimental group; control group and treament group which were given infusion with doses 2,5;5;10 g/kg bw. Test material administated for 36 consecutive days. Mean of sperm total per militer: KK (42,92± 3,28), KP1 (39,57± 2,08), KP2 (36,49± 2,73), and KP3 (33,37± 1,26) sperm/ml. Mean of percentage of sperm motility: KK (82,92 % ± 1,74) , KP1(68,54 % ± 6,32), KP2(61,23 % ± 7,13), and KP3(46,12 % ± 3,90). Mean of percentage of sperm abnormality: KK (37,63% ± 1,32), KP1(52,24 % ± 0,95), KP2(61,93 % ± 1,26), and KP3 (68,83% ± 0,66). Based on LSD test (P<0.05) the result showed that the data has differences between treatment and control group. The result indicated that the treatment group have impact on quantity and quality of spermatozoa of male mice with doses 2,5;5;10 g/kg bw.
